Photocatalytic inactivation of yeast in flow reactor

This diploma thesis focuses on the photocatalytic effect of titanium dioxide and ultraviolet light. Theoretical part shows the effect of titanium dioxide and aplication in various branches. Antibacterial effect of titanium dioxide iluminated by ultraviolet light was observed in the flow tube reactor on the yeast Hansenula anomala. After adjusting the optimal flow rate experimented with different inicial concentration of yeast cells. To increase desired effect also silver was used in the photocatalytic system. As observed in many previous studies, silver decreases the time needed for killing the microorganisms. The optained date were analysed.

Synthesis of spherical gold nanoparticles for biomedical applications

Tato práce se zabývá syntézou sférických zlatých nanočástic pro biomedicínské aplikace. Zlaté nanočástice byly syntetizovány ekologicky nezávadnými metodami, kterým je věnována i značná část rešerše. Cílem bylo nasyntetizovat stabilní koloidní zlaté nanočástice vhodné pro různé biomedicínké aplikace zejména, pro in vivo a in vitro zobrazovací metody, kterých přehled je take obsažen v teoretické části práce. Také byl proveden test cytotoxicity, jelikož částice mají být použitelné pro in vivo aplikace. Částice byly dale charakterizovány metodami SEM, DLS a UV-VIS.

Ion-selective membrane with antimicrobial effect

Tato práce se zabývá zlepšením antimikrobiálních vlastností komerčních polymerních membrán používaných v mlékárenském průmyslu pro filtraci mléčných výrobků jako je syrovátka. Tyto membrány trpí kontaminací způsobenou mikroorganismy přítomnými v mléce během procesu filtrace. Toto biologické znečištění ovlivňuje životnost membrány i samotný filtrační proces. Pro ochranu membrány před vytvořením biofilmu z mikroorganismů byly tyto membrány modifikovány pomocí plazmových metod tak, aby došlo k funkcionalizaci aminovými skupinami pro následnou imobilizaci nanočástic stříbra a selenu. Antimikrobiální aktivita obou typů nanočástic a membrán s imobilizovanými nanočásticemi byla stanovena proti buněčným kmenům Staphylococcus aureus a Escherichia coli.

Fabrication of titania mesoporous layers by material printing

This diploma thesis was focused on the preparation of mesoporous titanium dioxide layers. Mesoporous layers were prepared from titanium tetraisopropoxide as a precursor with addition diethanolamine and polyethylene glycol. These layers were deposited by material printing. This work was also focused on the characterisation of prepared layers. Hydrophilicity of titanium dioxide layers were studied. The photocatalytic activity of the printed titanium layers was tested by the photocatalytic degradation of stearic acid. Bang-gap studies were performed. Quality of prepared layers was inspected by optical microscopy, scanning electron microscopy and atomic force microscopy.

Electrophotocatalytic wather disinfection

This work focuses on the photoelectrocatalytic water disinfection by applying electrical voltage onto an electrode system. Working electrode consist of electrical conductor covered by thin film TiO2, because of its very important photoactivity. Disinfection effect was monitored on Gram-negative bacteria E. coli under different conditions. Inactivation of bacterial cells in contamined aqueous solution, first experiment was observed only with presence of oxygen, later reactor was irradiated with UV-A compact blacklight with intensity of 4 mW• m-2 or electrical voltage of 1V was applied to electrode. By adding both effects at the same time I was verifying photoelectrocatalysic water disinfection on chosen microorganism.

Functionalization of gold nanoparticles for imaging

The diploma thesis deals with synthesis of gold nanoparticles and their surface functionalization suitable for in vitro imaging. In this view there are requirements for optical properties such as stability, monodispersity and no presence of cytotocxicity. In order to fulfill these demands the synthesis by Turkevich method with surface modification by glutathione and polytethylenglycol was performed. Analytical methods such as DLS, SEM and Zeta potential measurement were utilized to characterize the physical and chemical properties of synthesized gold nanoparticles. Finally, MTT assay was performed to evaluate toxicity of gold nanoparticles using HEK 293 cell line.

Preparation of nanostructures using nanoporous template

The aim of the work described in this paper is to prepare electrochemically an optical sensor consisting of TiO2 quantum dots (QDs) using nanoporous Al2O3 template in the first step and to modify the surface of QDs with gold layer by evaporation process, again using nanoporous template, in the second step. This easy, cheap and rapid approach provides homogenous and highly ordered distribution of both nanostructures on substrate. The final objective is to characterize the physical parameters and fluorescent properties of TiO2/Au sensor designed for optical detection of biomolecules such as DNA and proteins.
