Development and optimization of sectioning technique for the study of migration and differentiation potential of testicular stem cells in X. tropicalis tadpoles

The rapid development of regenerative medicine and the urgent need for cells which are able to modulate the immune system, or even differentiate into variable cell types, have led to the research of mesenchymal stem cells. The Sertoli cells, which are essential for the proper development of sperm in the testis, have a strikingly similar character. In the previous research, a cell culture expressing markers of mesenchymal stem cells and Sertoli cells from juvenile male testes of X. tropicalis was established. At the same time, the cells were modified by the introduction of gene for the red fluorescent protein (RFP) in their genome. The aim of this diploma thesis was to clarify their characteristics after microinjection to X. tropicalis tadpoles (allogeneic transplantation). For these experiments, it was necessary to develop a reliable technique for the preparation of sections, which won't be harmfull for the samples. Using the vibratome sectioning method along with immunohistochemical labeling, the cell culture has been found to contain precursors of Sertoli and peritubulare myoid cells.
Strengthening intergenerational relationships in the family through reminiscence

This bachelor thesis deals with the use of the method of reminiscence specifically to boost intergenerational relationships in a family. It discusses especially the possibilities of strengthening relationships between grandparents and grandchildren by means of common thinking back and of time spent together. It works on the conviction of importance and uniqueness of the relationship between grandparents and grandchildren. The thesis introduces a reminiscence pilot project "Treasury of memories" for pupils of fourth year of primary school, discusses its signification, evaluates its contributions and concerns areas which proved to be insufficiently functional.
Analysis of gene expresion via next generation sequencing techniques

1 Abstract The main task of this work is create review today's available methods for gene expression analysis, introduce advantages and disadvantages of this metods and compare them. Nowadays sequencing is one of the most usable molecular methods. Sequencing methods are divided to three groups, a first generation sequencing, a second generation sequencing and a third generation sequencing. The most useful are the second generation sequencing. However, the third generation sequencing have a big potencial too. It is not necessary amplificate samples using PCR thanks them. Amount of data raises rapidly, thanks decreasing costs and increasing efficiency. Demands on data starage, computers output are growing. And softwares for data analysis are much clever than ever before.
Aspects of smoking among students of high medical school.

The thesis topic bears the name: "Aspect of smoking among students of high medical schools". Smoking is one the most often appearing drug addictions nowadays in Europe. With reference to the World Health Organization, there are 1.3 billion of smoking people in the world today whereas consequences of smoking kill approximately 4.8 million of people every year, which are 560 people each hour. If the current trend continues, then in the current pace 10 million of people will die of the consequences of cigarette smoking during the year of 2020. Smoking invokes an enormous addiction to nicotine which bears itself considerable economic, social and health problems. There were some students from 1st and 4th grade of the present form of studies of all high medical schools selected. Those were students particularly from the schools: SZŠ and VOŠZ České Budějovice, SOŠZ and SOU Český Krumlov, SZŠ Jindřichův Hradec, SZŠ Tábor and SZŠ Písek. In total, 591 of students were responded whereas most of them were girls (87%). The average age of the responded ones was 17 years. The thesis consists of theoretical and empirical parts. The theoretical part deals with history of cigarette smoking, a definition of drug addiction, chemical composition of cigarette smoke and malignity of individual toxic elements. Further, there is a chapter concerning problem issues of electronic cigarette smoking in the theoretical part. In the conclusion of this part of the thesis, there are some statistical data referring to cigarette smoking stated. There are some findings of quite a few of authors dealing with problem issues of cigarette smoking and addiction to nicotine summarized here. The empirical part consists of findings from my own research analysis which was done with the intention to map the prevalence of smoking among future health workers and their opinion to cigarette smoking among healthcare staff and people preparing for their healthcare career. There are also the respondents' opinions and attitudes included. The aim of the thesis was: 1.: To find the prevalence of smoking of 1st and 4th grade of high medical schools students. 2.: To find what influence the respondents' surroundings have on the fact of their smoking. 3.: To find what attitude high medical school students have to healthcare staff smoking cigarettes. To reach the above mentioned aims, there were three hypotheses determined: H1: There is a statistically significant difference existing in the prevalence of smoking at 1st and 4th grade of high medical school students. H2: There is a statistically significant difference existing in the prevalence of cigarette smoking among students coming from families where at least one member smokes. H3: There are statistically significant differences in the attitude of 1st and 4th grade students to cigarette smoking among healthcare workers. To prove or disprove the mentioned hypotheses there is a method of quantitative research selected. A questionnaire was used to collect the data. The questionnaires including 23 questions were distributed among 591 students of high medical schools of the South Bohemia Region. Their rate of return was 85%. The collected data were processed in the form of transparent graphs and the hypotheses were statistically tested. Pursuant to the evaluation of respondents' answers including the statistical data testing were proved by the hypothesis which presumed that there were statistically significant differences in the prevalence of cigarette smoking 1st and 4th grade students. The difference really exists that the 4th grade students smoke more often. It is interesting that the most considerable difference is between the schools of SZŠ and VOŠZ České Budějovice. On the contrary, the percentage of smoking students of SOŠZ Český Krumlov in the 1st grade is considerably high. Hypothesis no. 2 was statistically proved as well. Experiences of kindergarten teachers with shared custody and their attitude to this custody.

This Bachelor's thesis inquires shared child custody after divorce. In a theoretical part it is mentioned, when the shared custody, as a form of a child custody after divorce, was implemented. It also refers to its low frequency. The problem of shared custody is clarified here and a presentation of a research all over the world comes after. A difficult situation after divorce is mentioned afterward, an explanation haw difficult this period is, not only for parents, but also for children, who are overlooked usually, themselves. Further forms of child custody are mentioned in the end. Methods, used in the research, are presented in a methodological part. Information got from nursery school teachers are analyzed in a practical part. It is about their point of view of shared custody and of their experience with this form of custody.
Impact of the night work on the quality of life

The contents of bachelor thesis are named ?How Night Shifts Influence Quality of Life?. Shift work provides people with new jobs and wages. Work, during night shifts in particular, is usually evaluated with higher financial extras. Shift work, and particularly, work performed during night can have a negative impact on health of employees and their social and family life. Health problems are connected with irregular cycle of living. Humans naturally work during a day and relax at night. Biorhythm of a person working at night is different and some individuals hardly get used to this fact. Bad adaptable abilities of human body are the reasons for many health troubles, e.g. difficult concentrating, stomach illnesses, problems with blood circulation, and researches of some experts also recorded a higher risk of cancer occurrence. Shift work also influences social life of people, family routine and partnership. Young people often suffer from difficulty of finding a lasting partnership and parents working in shifts cannot spend much time with their families. Some employees of a company with the three-shift operation are chosen for the research. The company is involved in production of moulded pieces on injection mouldings. Approximately 126 employees work in three-shift operation, mostly women (app. 74%). 98 employees takes part in the research, from whom 77% are under 45 years old. The bachelor thesis contains theoretical and empirical parts. The theoretical part deals with the objectification of conceptions. There are ideas by many authors included, debating issues of night shift and three-shift operation and quality of living with regard to health of people, further to social relationship and partnership, and other similar issues. There are number of various impacts of night shift on the physical and mental side of human health and the influence of a night shift routine on the selected social aspects, particularly, on family and partnership. The empirical part contains my own findings of the analysis of research which is done to find the influence of night shift on the quality of living. The target of thesis: Ad 1: to find out how the subjective night shift workers? feelings of their health are influenced in three-shift routine of work. Ad 2: to find the influence of night shift of the three-shift routine of work on the social life. Hypothesis 1: The tested people feel deterioration of their health due to night shifts. Hypothesis 2: The night shift routine affects the selected aspects of social life. confirm or rebut the hypotheses mentioned above, a method of quantity research of quantitative research is chosen, and a questionnaire is used to collect the data. The questionnaires, containing 16 questions each, are distributed among 126 of respondents. The 78% of them are returned completed. The collected data are included in the survey graphs and the hypotheses of statistics are tested. The first hypothesis is confirmed on the basis of the evaluation of respondents? answers by the statistical testing. The hypothesis supposes that the surveyed workers feel their health deterioration due to their night shifts. According to the certain indicators, health deterioration is notable depending on the number of years spend in the three-shift routine during their employment. Age also plays an important role in the evaluation of results. Employees of an advanced age often have headaches than those of a younger one. The total frequency of health problems grows by ageing therefore workers of an advanced age experiencing night shifts have evidently more often health problems. Moreover, the statistics show that night shift more negatively influences health of women. On the basis of the obtained data demanding targets are achieved. The bachelor thesis can become a resource both for experts and general public who are involved in monitoring the influence of night shifts on quality of living.
