In the theoretical part, this diploma thesis briefly describes the origin and processing of honey. Then, some of its physical and chemical properties are characterized, focusing on the methods of their determination. The work also focuses on the effects of honey on human health and its possible applications in the treatment of various diseases, and finally with a brief presentation of other bee products. In the practical part, an analysis of selected physical and chemical properties (water content, titratable acidity, active acidity, colour, glucose content, proline content) of honey samples obtained from the market was performed. The results of the determination were compared with the values set by the legislation. A sensory evaluation was also carried out for the overall assessment. Finally, a statistical comparison of individual results was also performed.

Expression and localization of alpha V and alpha 3 integrins in male germ cells of selected mammalian species

Integrins are transmembrane glycoproteins mediating cell-cell adhesion and ensuring communication between extracellular matrix and cell cytoskeleton. The presence of integrin subunits has already been described on somatic cells, as well as on oocytes and sperm cells. In oocyte plasma membrane, integrins together with tetraspanins are involved in the formation of vast molecular webs. Analogical structures have been recently discovered in sperm plasma membrane as well. While the role of integrins on oocyte has been researched quite extensively, there is much less known about its presence and function on sperm. This Master's thesis focuses on expression and localization of integrins alpha V and alpha 3 on acrosome-intact sperm and its comparison between mouse, pig and human sperm cells. Furthermore, this thesis also describes relocation of the alpha V integrin after the acrosome reaction, which is an essential step of sperm maturation. Using confocal microscopy, the alpha V integrin localization was detected in acrosomal cap area of acrosome-intact sperm of all selected species. After acrosome reaction, the alpha V integrin relocated and was depicted in postacrosomal region and equatorial segment of acrosome-reacted sperm. Optimization of biologic therapy in children with inflammatory bowel disease (IBD) using modern biomarkers Abstract to thesis Study programme: Biochemistry and Pathobiochemistry Introduction: In adults, infliximab (IFX) levels correlate with disease activity and antibodies to IFX (ATIs) predict treatment failure. We aimed to determine the association of IFX levels and ATIs with disease activity in paediatric population. Methods: This study was performed as a prospective observational study. We prospectively collected blood, stool, and clinical data from 65 patients (age 10.5-15.1 years) with Crohn's disease (CD) before IFX administration, and measured IFX trough levels, ATIs, and faecal calprotectin levels (CPT). We used multivariate analysis to identify the predictors of IFX levels. IFX and ATIs levels were meassured using ELISA. Results: Lower levels of IFX were associated with ATIs positivity (OR [odds ratio] 0.027, CI [confidence interval] 0.009-0.077). Higher C-reactive protein (CRP) level, erythrocyte sedimentation rate (ESR) and CPT levels were found in patients with lower IFX levels. The optimal combination of specificity (50%) and sensitivity (74%) for disease activity was calculated for IFX levels ≥ 1.1 µg/ml using CRP level < 5 mg/l as a marker of laboratory remission. Immunological causes of female infertility

Currently, up to 15 % of reproductive age couples struggle with infertility. Female infertility is the cause of conception issues in nearly 40 % of these cases. The origin of female infertility consists of wide range of causes and other factors. Immunological causes are certainly one of the most significant ones. In pathological conditions, defense mechanisms of female immune system can target alloantigens presented on sperm surface. Moreover, autoimmune response and antibody production can occur as well. Antibodies can target various parts of ovarian follicles and oocytes, phospholipids, nuclear antigens, enzymes etc. These antibodies can inhibit fertilization, maturation of oocytes and embryo, embryo implantation, and also recurrent miscarriages. The aim of this thesis is to summarize various causes of female infertility focusing on immunological pathologies. The thesis also describes functioning of immune mechanisms in the female reproductive tract and briefly touches on oogenesis and female hormonal cycle.

Lifestyle and social capital "active young generation".

Diploma thesis "Lifestyle and Social Capital 'active young generation'" discusses the background of the relevant theoretical anchor, the two dimensions of individual life in society - the lifestyle that is in this work conceived as a space for identity formation (through the dimension of consumerism) and it operationalize the concept of leisure sports and related sportive activities, the second dimension is the theme of social capital, in accordance with its structural components (egocentric individual networks) and cultural aspects (defined by degree of tolerance and social trust ). The aim of this work is through the empirical sociological analysis of representative survey ISSP 2007 Volný cas a sport and Distinkce a hodnoty 2008, map to the active lifestyle of the individual in its socio-demographic and socio-economic determinants, monitors the process of sport socialization within the family and close friends and value preferences and orientation, the which may be active in leisure sports potential impact. In conclusion, this work deals with effects of sports on the components of social capital.

Corporate Social Responsibility: cooperation between businesses and non-profit organizations

The objective of the thesis is to define key points of successful cooperation between businesses and non-profit organizations. The thesis is focused on Corporate Social Responsibility in terms of cooperation with a nonprofit sector. The theoretical part of the thesis defines the introduction into CSR topic, the introduction into non-profit organizations, types of cooperation, partnership of both sectors and criticism of CSR.
The practical part of the thesis consists of a research, which is made by three research surveys. The first research survey uses the method of the secondary document analysis. Another two surveys are realized through semi-structured interviews. The interviews are taken with both sectors to deliver complex research results. Firstly with specialists from businesses and secondly with non-profit organizations specialists. Four key results come from the research and these significantly increase the success of mutual cooperation. These are accuracy prepared strategy, clear expectations on both sides, equal partnership and mutual respect of the expected standard.

Forms of Regional Co-operation in the Light of the Promotion of Production

I chose Forms of the regional cooperation of the view support of production as a theme for my bachelor thesis. I Works with Local Action Group Světovina o.p.s., which is situated in Pilsen-North and Rokycany district. My major target for my bachelor thesis is analysis of chosen forms regional coorperation and define its potentiality in the sphere support of the local production. Secondary target is determaned, if entrepreneurs in sphere of LAG Světovina o.p.s, if they might know about co-operation with this Local Action Group, alternatively if they want be able to interest in member of this group.
