Studium prevalence a subtypové diverzity střevního prvoka rodu Blastocystis sp. ve zdravé lidské populaci v České republice

Blastocystis sp. is a common intestinal protist colonizing the human intestine. Its role in the human gut ecosystem remains unclear. Here, we aimed to expand the knowledge of the epidemiology of Blastocystis sp. in the gut-healthy humans in the Czech Republic, including the distribution of its subtypes, the correlation between its occurrence and several factors such as lifestyle, contact with animals, age, and sex. A total of 288 stool samples were obtained from asymptomatic individuals over the entire age-range and 136 samples from animals with which the volunteers were in frequent contact. All samples were examined in parallel by PCR and xenic in vitro culture.

The main aim of this study was to optimize the qPCR diagnostic protocol for determination of the prevalence of intestinal protist, Giardia intestinalis, in a volunteer group across the Czech Republic. A total of 428 samples were collected from gut-healthy humans (296) and from animals (132) with which these people were in close contact. The overall prevalence of G. intestinalis was 7 % in humans and 19 % in animals. In addition, we compared the sensitivity of two molecular methods for detecting of the presence of giardia, specifically qPCR and conventional PCR. Based on our results, we found out that qPCR is more sensitive method than PCR.
F. Kupka and his letters to V. Hynais

This bachelor thesis, named F. Kupka and his letters to V. Hynais deals with mutual correspondence between František Kupka and Vojtěch Hynais. Their letters are currently placed in the National Gallery Archive in Prague. Dealing with this correspondence required translation from French to Czech and critical analysis of the text in comparison to already published studies. Many of the currently known information concerning Kupka has been confirmed while some new facts have also been uncovered. Although it may have seemed formal at first sight, the relationship between the two artists was truly warm. They shared not only their artistic points of view but also the joys and sorrows of their family lives. They often included magazine articles dealing with art and the current situation in the Czech and French art world into their letters. Kupka often mention his work, mainly book illustration, and expressed his longing for painting. His correspondence reassures us of his punctual preparation for each art project and also sometimes helps withs its dating. The reader is also confronted with Kupka's successes and failures with his search for a new artistic expression, although this can only be found in slight indications, not in connection to concrete paintings. Studium prevalence \kur{Giardia intestinalis}

The present project is focused on the study of a prevalence of the gut protist, Giardia intestinalis, in human and animal samples using two diagnostic approaches: (i) coproscopical diagnostics, specifically Sheather flotation technique, and (ii) PCR. First, three PCR protocols for different genes (triosephosphate isomerase, beta-giardin and small ribosomal subunit) were optimized by gradient PCR. Based on these result, the gene TPI was chosen as the best option for molecular diagnostics of G. intestinalis in the collected stool and fecal samples. Giardia was detected only in the animal samples (2/41, dog and rabbit samples), all the analysed human samples remained negative. Further, the TPI protocol showed low sensitivity for diagnostics of G. intestinalis.

English Abstract The goal of my master thesis is to present a comprehensive study of the life and work of Sobeslav Pinkas (1927 - 1901), one of the most important Czech realistic painters. Painter's personal and artistic development is introduced in the wider historical context to allow us to understand why he had decided to become a painter. In 1948 Pinkas, as a young student of law, joined the armed revolution and personally attended the riots. Later he became a caricaturist for the Sotek magazine. His craving for democracy in the political life was mirrored in his attempts to find a new way of artistic expression. Neither the Academy in Prague nor the one in Mnichov could stand up to his expectations, and he consequently chooses Johann Baptist Berdelle as his personal teacher. Later, following a short Prague intermezzo, he, as one of the first Czech artist, leaves for Paris, where he studies under the guidance of Thomas Couture. Later on he discovers the work of Jean-François Millet and is heavily influenced by it. His conversion to the modern style of French painting wasn't received in Prague with much enthusiasm, which may be one of the reasons why he gradually, after his return to his homeland, stops painting. Attitude discourse markers: a contrastive analysis of selected texts

The aim of this Master´s thesis is the contrastive analysis of discourse markers on a selected type of texts in Spanish and Czech language. The thesis is divided in two parts theoretical and practical part. Firstly, in the theoretical part of the thesis, is introduced the textual linguistic the field which studies discourse markers. For wholeness, are briefly mentioned all of the seven standards of textuality with profound focus on textual coherence. Other part of the thesis aims on terminology of discourse markers (especially on conectores), which is problematic, and various classifications according to Czech and Spanish authors. The last part of the theoretical part is dedicated to attitude markers the main theme of this thesis, their characteristics, functions and examples. The practical part of the thesis is introduced by contrastive analysis of attitude markers carried out on selected sample of texts. Firstly the attitude markers are identified and secondly described and classified. Also is mentioned the absolute and relative frequency and comparison of the contrasts and coincidences in both languages. The thesis is finally summarized in Spanish résumé.
Taxation of the Dependent Agent´s Revenues

The thesis deals with finding definition characters of agents as permanent establishments (dependent agents) available from publicly accessible sources, including its negative definition in comparison with the concept of an independent agent. The second part of the thesis analyses all possible and suggested ways of tax base creation, i.e. application of substitute methods of profit assessment and tax base enumeration from accounting profit or difference between revenues and expenses. Further this part of the thesis focuses on the issue of appropriateness of the Proposal for a Council Directive COM(2011) 0121 of 16 March 2011 on a Common Consolidated Corporate Tax Base (CCCTB) for dependent agent's purpose. Used methods include analogy, analysis, synthesis, deduction and comparison.
The Multimedia Educational Programme for teaching regional geography of Australia and New Zealand for 2nd stage of PS

The diploma thesis deals with teaching regional geography of Australia and New Zealand on the second stage of Primary school. The main aim is to create the multimedia educational programme in the SMART notebook 11, the special programme for interactive board. The thesis also contains a methodological guide for teachers which was created with educational programme. This programme corresponds to the principles of the current educational documents (Framework Educational Programme for Elementary Education). The theoretical part deals with the didactic analysis of current educational textbooks and workbooks of geography which are designed for the second stage of Primary school and included the subject matter. There was made a research by a controlled discussion with geography teachers to get a notion of time allocation and volume in teaching regional geography of Australia and New Zealand.
Česká centrála cestovního ruchu - CzechTourism and its activities in Spain and Portugal

The aim of this bachelor thesis is to deal with the organization Czech central office of tourism - CzechTourism and its activities, which this organization produces at its foreign representation in Spain and Portugal. At first the central office is represented in general, explaining, how it was established in these countries and which statute belongs to this organization. Alongside, this work mentions the structure of the organization, its mission, main goals and management. Also specifies the subjects, which corporate with the central office on the national and international level. In the following part is marginally mentioned, how Czech central office of tourism works in whole and what uses for the support of incoming tourism of Czech Republic. The next part is dedicated to foreign representation in Spain and Portugal and specific activities, which the foreign representation uses for promoting Czech Republic, as an attractive destination for travelling in these countries. Finally, I will evaluate the importance of the activities and also possible contribution for mutual relations in the field of tourism.
