Determination of ELOS (organic matter solubility in cellulose) in forage by domestic cellulosis from Trichoderma reesei: applied methodology with certification

Organic matter digestibility (OMD) is considered to be on of the main indicators of forage quality. In fodder research, breeding, and farming practice in the CR methods in vitro using clean cellulolytic and proteolytic enzymes in ANKOM Daisy Incubator are usually used for its determination and are expressed as ELOS. Methodology is focused on (1) the optimum procedure of determination with respect to its accuracy (i.e. the highest possible agreement of predicted values with the values of control method) and repeatability (i.e. agreement of predicted values of repeated determinations) and (2) the use of this exact and expedient method for serial evaluation with a warning about its limits.

Methodology for analysis of dairy products and their soy analogues for the presence of RoundUp Ready soybean

Dairy products may be subject to adulteration, where there is a relatively expensive raw material (e.g. cream) replaced by the addition of vegetable fats. Milk can be replaced in dairy products for example by soybean extract. Because soy belongs among strong allergens and the incidence of allergy is rather high. Therefore it is necessary to have a method for its detection. This work introduces the method for detection of soy, including analysis of genetic modified RoundUp Ready soybean. Detection of soybean in dairy products is based on analysis of amplification products melting profiles by High Resolution Melting (HRM) method. Analysis of geneticaly modified RoundUp Ready soybean is based on DNA detection via Polymerase Chain Reaction(PCR).

Alternative nitrogen nutrition of plants

The methodology provides the basic information about the usability of nitrogenfixating bacteria. The methodology describes procedures of the isolation and screening of beneficial soil bacteria Azotobacter spp. and Rhizobium spp. It also provides recommendations to promote biological nitrogen fixation. The methodology also represents the use of inoculants as a necessary part of modern farming practices.

Methodology for the phenotyping resistance to Plum pox virus in peach

Sharka disease, which is caused by the Plum pox virus (PPV), is the most economically important virus disease in fruit trees worldwide. Since it is not possible to completely eradicate PPV in the Czech Republic, it is necessary to find another way to control this pathogen. The most promising seems to be the breeding of new cultivars for resistance. This methodology provides a comprehensive protocol for testing PPV resistance in peach. It is based on visual assessment of disease symptoms on leaves and fruits, and determining the presence of the virus in plants using ELISA and RT-PCR techniques.

The Efficient Application of Near Infrared Spectroscopy with Fourier Transformation for Prediction of Wheat Technological Parameters

This method brings to the users basic information about principles and features of NIR spectroscopy as well as more detailed view of the state-of-the-art capabilities of NIR spectroscopy in wheat quality prediction. The agriculture professionals will use supportive information about possible utilization of the method including practical tips for proper NIR spectrometer selection. For both, the current and prospective users, the method provides practical guidance to device handling, management of calibration sets and calibration models development. The objective evaluation of the 21 predictive models based on FT-NIR measurement, covering the results of 14 reference analytical methods aimed at wheat quality. Furthermore, it offers information useful directly in breeding programs, mill and bakery production and also in agriculture sector, government control and at universities.

Cultivation of fodder sorrel for biogas production

The aim of the methodology is to provide farmers, agricultural consultants, operators of biogas plants and all other interested persons basic information about growing and processing for biogas of a promising non-traditional energy crop - fodder sorrel (Rumex patientia L. x Rumex tianschanicus A.Los.). Until now a comprehensive methodology for cultivation of this crop for biogas production hasn’t been processed. The methodology provides basic botanical characteristics of the crop, specifies soil and ecological requirements in habitat, describes the agricultural technologies of seeding, cultivation, fertilization, plant protection, harvesting and ensiling this crop for biogas production and provides their economic evaluation.

The publication provides basic botanical characteristics of the crop, specifies its soil and ecological habitat requirements, describes the agricultural technologies for seeding, cultivation, fertilization, plant protection, harvesting and post-harvest treatment and gives an overview of potential productivity and main utilization possibilities, especially for biogas. The importance of this legume for soil fertility is also emphasized.

Sorghum (Sorghum bicolor (L.) MOENCH) and its utilization possibilities in the condition of the Czech Republic

Sorghum is one of the oldest used crops. Presently, it is one of the five most cultivated cereals in the world for human nutrition. This methodology includes a comprehensive view of sorghum cultivation and presents the newest knowledge in range from botanical taxonomy to the possibilities of utilization. In this publication, there are included not only our own results but there are also results from the world scientific publications. There are characterized selected species of sorghum suitable for different climatic conditions of the Czech Republic. There are described all aspects and procedures, which can influence the final production of selected sorghum varieties including their quality from the point of view of biogas production.
