The diploma thesis is focused on the comparison of the quality characteritics of fallow deer meat originating from farm breeding, in cluding the assessment of the influence of different methods of slaughter, gender, and suplementary feeding of mothers during the lactation period on the occurence of qualitative changes in the texture of the meat literature rewiev is devoted to the definition and composition of meat, and then selected characteristics of meat are described individually. At the end of literary part, information about farm breeding is sumarized. In the practical part, the farm breeding of fallow deer of the Friedberger family is presented. Then the results from the analysis of the chemical composition of deer meat are presented, followed by an assessment of the occurrence of qualitative changes in the texture of meat depending on the monitored factors.
The Progress of Nursing in the 21st Century from a Patient's Perspective

Intoduction: Nursing is a human-oriented scientific discipline. Due to various world events, pivotal changes have occurred in this field. These changes have led to improvement in nursing care that is being provided in health facilities today. The Aim of the Thesis: The aim of the thesis was to determine changes to the field of nursing through the eyes of care recipients (i.e. patients) that happened during the 21st century. Furthermore, the goal was also to identify factors that influenced these changes and evaluate nursing care through the eyes of patients. Methodology: The thesis was carried out by means of a qualitative research survey in the form of semi-structured interviews with patients. The selection criteria for choosing an informant were residence in the South Bohemian Region and 18 years of age (or older). These informants were chosen by snowball sampling. The research survey included 20 informants. The interviews were recorded on an answering machine, literally transcribed and consequently analysed. Results: The thesis identified a total of 8 categories and 23 subcategories. The category of Scientific and Technological Progress was recognized as the central category. It is the driving force for most changes, including changes in education. The patients thought that the biggest change was modern equipment, new aids and devices in health facilities. They also mentioned innovations in the education of nurses and they agreed with the changes. The patients rated nursing care as a highly professional, expert and demanding field. They understood that more and more demands were being placed on the nurses. Moreover, the patients described the profession as prestigious, but also said that the public should be more informed about it. Conclusion: It is certain that nursing care will be influenced by further advances and innovations because science and technology cannot be stopped. In this context, the demands on nursing personnel will increase and the patients will be more demanding as well. Therefore, it is necessary to continue working on nursing care. However, it has to be taken into account how these changes and developments are perceived by the patients.
Porovnání vybraných reprodukčních ukazatelů mezi zakrslými a zakrslými beraními králíky

The bachelor thesis is focused on the evaluation and comparison of selected reprodu ctive indicators between two breeds of dwarf rabbits. The beginning of the literary part is devoted to the origin, domestication and history of rabbit breeding not only in the Czech Republic. In the next section, the standards of the dwarf white sanded rabbit and the wildly colored dwarf rabbit, including their origin , are presented and describe. The second half of the literature is focused on the reproduction of rabbits. The conc lusion of the literature section summarizes the most common pathological conditions and behavioral disorders in reproduction. In the practical part is the comparison of selected reproductive indicators between two dwarf breeds of rabbits.
The effect of morphine withdrawal on G protein-coupled receptors and redox balance in rat brain

Although morphine is one of the most effective analgesics available, its long-term use is severely limited by the development of tolerance and dependence. Molecular mechanisms responsible for these effects have been the focus of intensive research for decades aimed at developing safer and more effective pain treatments. The aim of this thesis was to contribute to a better understanding of the molecular actions of morphine, as an understanding of these processes is essential to develop better therapeutic approaches. Using western blot analysis, we investigated the effect of 10-day treatment with increasing doses of morphine and its subsequent withdrawal on key components of opioid signalling in the rat cerebellum and cerebral cortex. Amounts of opioid receptors, various heterotrimeric G protein subunits, GRK3 kinase and β-arrestin 2 were examined. Furthermore, the levels of adenosine A3 receptor and cannabinoid receptor type 1 were detected. Finally, we also investigated the effect of morphine and its withdrawal on oxidative stress markers. Morphine administration resulted in a significant decrease in GRK3 kinase levels in both brain regions, and these changes remained even after withdrawal. Quality of amateur and professional translation of film subtitles

(in English): The aim of this thesis is to discuss the issue of quality of audio-visual translation in amateur and professional environments and to examine the impact of the quality of subtitle translation on the viewer's understanding and overall impressions of the film. As a part of our empirical research, we conducted a translatological analysis of Czech subtitles for the French comedy Le sens de la fête (C'est la vie!, 2017) and the social drama Les misérables (2019). Subsequently, using the focus group method (group interviews), we organized an experiment with 6-7 participants, during which we showed a part of the film with professional subtitles to one group of viewers and with amateur subtitles to another group, and then we tried to verify their impressions and overall understanding of the film as well as their perception of the quality of the subtitles. The results obtained from each group were compared with each other and then with the findings from our translatological analysis. Among other findings, the study shows that quality had an impact on the overall impressions and understanding of the film, especially for the first film, which was based on long dialogues, jokes and wordplays, and where the difference in quality between the professional and amateur versions was more marked. Title of Diploma Thesis: Thiopurine S-methyltransferase - clinical importace of genotyping and phenotyping. Thiopurine S-methyltransferase catalyzes S-methylation thiopurine's drugs such as 6-mercaptopurine and thioguanine. TPMT genetic polymorphisms represent an important role in clinical pharmacogenetics. The differences in TPMT activity result from mutations in gene for TPMT. The polymorphisms are important factor in efficacy of treatment by thiopurine drugs. Patients inheriting low activity of enzyme TPMT have mutated allels, patients inheriting high activity of TPMT are usualy wild types. TPMT gen was genotypized by method real-time PCR in volunteers (n=55) with autoimmune diseases. The average of patient's age was 16,7 years. From blood collected into EDTA DNA was isolated by using QIAmp Mini Kit (Quiagen, Germany) and it was used for genotyping of TPMT. Laws of nature and humankind in rural stories of Vítězslav Hálek

The thesis deals with literary production of Vítězslav Hálek - namely his rural short stories. I have tried to observe evolution of both the writer and his work in two creative periods of his life: 1857 - 1863 and 1871 - 1874. I am focusing on comparison of the stories written in two productive stages of Vítězslav Hálek, searching for both common features and the differences. I pay special attention to images of natural scenery, their function in the storyline and the importance of nature in human life. In Halek's approach, nature is ruled by essential laws which should be abided and respected by people. Therefore the main part of my thesis is concerned with individual images of nature laws and their comparison with the laws applicable in human society. AdditionalIy, I take account of Halek's career as well as his literary production, which has been listed in the record. Hálek read well in all his life, he was considered popular and recognised author, however, some of his stories are hard to find these days. There do not even exist contemporary belles-lettres on Vítězslav Hálek. My study is based primarily on Halek's correspondence, small biographic essays and, lastly, on analysis of narrative texts. Internal structure of the Balkan refugium using Erinaceus roumanicus as a model organism

The aim of this work was to describe the spatial distribution of genetic variability of the northern white-breasted hedgehog (E. roumanicus), mainly within the Balkan Peninsula and Central Europe, as these areas are crucial for understanding the effects of Pleistocene climate oscillations on the genetic architecture of the species. Based on this spatial distribution, hypotheses about the possible structuring of the Balkan refugia were formulated. A total of 260 individuals and 9 microsatellite loci were used for the analysis. Using the approach of landscape and population genetics several possible isolated subpopulations within the area surveyed were identified. Unique status was proved for a population originating from Crete, probably influenced by mechanisms of island evolution. Differences from the rest of the range were ascertained also in population from the Czech Republic, located in a secondary contact zone with the E. europeaus. The role of interspecific interactions and possible introgression should be considered in this case. In the area of Romania and the area south of the Balkan Peninsula the highest genetic distances between individuals were identified, probably associated with the occurrence of geographical barriers and the possible presence of glacial subrefugia.
