DNA damage response in mammalian oocytes

During early embryonic development oocytes are arrested in prophase I of the first meiotic division, in which they can persist for years. After reaching sexual maturity and the luteinizing hormon surge resumption of meiosis and meiotic maturation occur. Oocytes are arrested again at metaphase of the second meiotic division. At this stage they are ovulated and waiting for a fertilisation. Oocytes are during their development exposed to factors that cause DNA damage, of which DNA double-strand breaks (DSBs) are the most serious threat. The maintaining of genome integrity is crucial for quality of oocytes, fertility and proper embryonic development. The mechanism of the oocyte response to DSBs presence is not fully understood and it seems to differ from somatic cells. We assume that DSBs are repaired during meiotic maturation probably by a mechanism of homologous recombination (HR). In this thesis we focuse on essencial recombinase RAD51, which participates in the repair by HR. We found that RAD51 inhibition leads to an increase of segregation errors in anaphase I. Using high resolution live cell imaging we observed chromosomal fragments and anaphase bridges. Immunofluorescence detection of DSBs-marker γH2AX showed increased amount of DSBs in prophase I and MII stage after RAD51 inhibition. Our data...
Technical Condition Monitoring of Radar Systems Components

The goal of this thesis is to design a diagnostic subsystem for secondary surveillance radar and to construct a functional prototype of communication and testing generator unit. The first part of the thesis aims at description and division of radar systems, with emphasis on air traffic control radars.  Main attention is being focused on ATC secondary surveillance radars and description of their function. Following part deals with requirements specification for design of diagnostic system and the system design itself. Part of the thesis forms design, production and testing of prototype of block enabling communication of diagnostic subsystem and function of testing generator for secondary surveillance radar.

Universal Sensoric Platform with Programmable Features

The aim of this thesis is to design universal sensoric platform for data collection in manufacturing environment. Device is capable of collecting data from variety of sensors, but it is also able to control external systems. Platform is controled by Lua scripts. The scripts are generated in user friendly visual programming environment Blockly. Platform is based on WiFi module ESP-WROOM-02.
Effect of sperm ubiquitination on the early embryonic development in pig

The ubiquitin-proteasomal system which is the major pathway for intracellular protein degradation is also involved in sperm quality control in the mammalian epididymis. Defective sperm become surface- ubiquitinated during epididymal passage. The level of sperm surface ubiquitination negatively correlates with their quality. Hypothetically it is possible that after fertilization, highly ubiquitinated sperm, naturally present in mammalian ejaculates, would be actively recognized by oocyte (probably via 26S proteasomal complex). Subsequent partial or total sperm degradation should negatively affect the development of the potentially defective embryo. In this study, we examined the effect of sperm ubiquitination on the early embryonic development in pig (Sus scrofa f. domestica) using the method of intracytoplasmic sperm injection (ICSI). In vitro embryonic development to the blastocyst stage after ICSI was comparable with other laboratories. In this study, no significant difference was observed in the formation of pronuclei between oocytes fertilized by lower and highly ubiquitinated sperm cells. System for Household Monitoring and Automation of Central Heating

In this work, which belongs to the field of embedded systems, several hardware modules and central control unit for home automation purposes were designed and subsequently implemented using 1-Wire bus and tiny computer Raspberry Pi. Furthermore, an extensive application for management of automation activities in a given system was created in C++ programming language. This piece of software is running continuously as a service from computer boot time until its shut down. There were implemented several functionalities that enable controlling the general purpose relay to switch home appliances and to operate central heating automation features, which include operating of electronic actuators on the radiator and boiler. In addition, web-based application allows direct monitoring of automation activities and provides necessary means to plan the desired actions of the available units within the automation system. Resulting system could be installed on a target site and monitor temperature, automate central heating and switch basic home appliances using relay. Main advantage of the created system can be recognized in scalability where new automation tasks or devices could be added.
Audio Player Device for Automotive Environment

The purpose of this thesis is to create an audio player device for automotive environment. Thesis describes development of modern car radio with touchscreen, built on oneboard computer Raspberry Pi 2. It describes development of hardware parts of the device from concept through selection of components to realization. It also describes development of firmware for this device, including graphical user interface. The device was built and it fulfilled set goals.
Design and Realization of Device-Prototype for Multi-Channel Monitoring of Measured Quantities via USB

The aim of this work is design and construction of a device based on microcontroller Freescale MC9S08JM60 that will extend laboratory kit used in the lab of Computer Hardware course at FIT BUT. The device allows monitoring of several measured quantities and sending the measured values to a computer via USB. The first part presents the current laboratory kit and used microcontroller. The second part contains device’s design including schematic, PCB and a list of used electronic components, a description of implementation of firmware for the microcontroller in C and control application written in C#. This application receives the measured data samples from the device and visualizes them to demonstrate the functionality of the device. In the last part of the bachelor’s thesis there are proposed steps to upgrade the laboratory kit based on this device, contained its evaluation and possible extensions.
Control Panel with Touchscreen

The aim of this work is to design and construct a printed circuit board with touchscreen connection that will serve as a universal platform for controlling a variety of devices including industrial devices. The functionality of the board is demonstrated on control of gas boiler application. The first part attends to currently used and available graphic displays, touchscreens, communication interfaces and microcontrollers that allow control of a graphic display. The main focus is put on LCD TFT displays, resistive touch panels and microcontrollers with LCD TFT controller. The second part describes the selection of system components, design of printed circuit board and implementation of thermostat. The end of the work evaluates the produced device and discusses possible future extensions.
