The aim of this thesis is to analyse applicability range of relations for a critical load at buckling for columns with deflection from ideal column – initially curved column or eccentric load. The solution of this thesis is analytical and it comes from solving the differential equation of the deflection curve. This equation has different general solutions for both assumptions. The result achieved in this thesis is general recommendation for maximal values of eccentricity of load according to cross section. Next result is recommendation of maximum curve according to cross section. The chosen cross sections are circular, square, rectangular and thin tubes.
Separation of pharmaceutically active compounds by mixed-mode and chiral chromatography: interaction study
Fojtíková, Sofia ; Bosáková, Zuzana (advisor) ; Kozlík, Petr (referee) ; Fischer, Jan (referee)
During the drug development process, the pharmaceutical industry often relies on chromatographic techniques. High-performance liquid chromatography (HPLC) is among the most commonly used chromatographic techniques for these purposes, with reverse phase and HILIC being the most common choices. However, despite their popularity, these techniques have several drawbacks, such as the contribution of secondary interactions that arise from the nature of the carrier material, and limitations in terms of analyzing molecules with certain properties. In this sense, multimodal (also called mixed-mode) chromatography (MMC) appears to be a promising technique capable of addressing these challenges. The first part of the dissertation is focused on the comparison of the retention of structurally different molecules in different chromatographic systems using seven different multimodal stationary phases and the effort to elucidate the interaction mechanisms applied to the individual studied stationary phases. By observing the trends, obtained under different conditions for molecules with different physicochemical properties, it was at least partially achieved. Chiral chromatography is another key tool in the analysis of pharmaceuticals. Its main purpose in the development of medicinal products is to separate...
Determination of propyl gallate on carbon paste electrode
Vysoká, Marie ; Zima, Jiří (advisor) ; Fischer, Jan (referee)
Propyl gallate (PG) is a significant synthetic antioxidant and preservative. Its determination has been studied at carbon paste electrode (CPE) using differential pulse voltammetry (DPV) and high performance liquid chromatography with electrochemical detection (HPLC-ED) and with UV spectrometric detection (HPLC-UV). Concentration dependences were measured in the media of Britton-Robinson buffer (pH 5) and methanol (20 %, v/v) by DPV and the limit of detection 0,6110-7 moldm-3 was obtained. Using HPLC with a mobile phase consisting of 0,01 moldm-3 phosphate buffer (pH 4) and methanol (50 %, v/v) with potential of working electrode E = +0,8 V and detection wavelength λ = 280 nm, concentration dependences were measured. Limit of detection was determined to 0,39 moldm-3 for HPLC-ED and 4,95 moldm-3 for HPLC-UV. After verification of the extraction procedure PG was determined in vegetable oil. The resulting value of 3,2 mgkg-1 corresponds with permited limits.
Electrochemical detection of n-propanol on Au and Pt electrodes modified with polyaniline-based composites
Pejzlová, Michaela ; Fischer, Jan (advisor) ; Vyskočil, Vlastimil (referee)
The electrochemical oxidation of aliphatic alcohols is problematic on conventional electrode materials and to develop methods for their detection, the catalytic properties of electrode surfaces need to be exploited. This thesis is concerned with the study of the electrochemical deposition of polyaniline (PANI) in acidic media on three different electrodes, platinum, gold and glassy carbon electrode. The properties of the PANI modified electrodes were tested in comparison to the unmodified electrodes. The electrooxidation of aliphatic alcohols n-propanol and 2-propanol in alkaline medium was studied by cyclic voltammetry. Subsequently, the effect of electrodeposited noble metals (palladium and gold) on the surfaces of unmodified and PANI modified electrodes on the electrooxidation of propanol was investigated by using the same procedures. From the linear part of the concentration dependence of n-propanol on the chosen electrodes, the limit of detection was determined. The lowest value was calculated for the platinum electrode with electrodeposited gold (LOD = 0.2 mmol l−1 ). PANI modified electrodes showed wider linear dynamic range than electrodes without this modification. Keywords: cyclic voltammetry, electrooxidation, glassy carbon electrode, gold electrode, metal electrodeposition, n-propanol,...
Electrochemical behaviour of {110}-oriented single crystal boron-doped diamond electrode for electroanalysis
Poláková, Jana ; Schwarzová, Karolina (advisor) ; Fischer, Jan (referee)
This bachelor thesis deals with the study of the electrochemical behavior of the neurotransmitter dopamine on mechanically polished single crystal boron-doped diamond electrodes with crystal orientation {110} and the boron-to-carbon (B/C) ratio in the gas phase during the chemical vapor deposition process at 250, 500, 1000 and 2000 ppm. Measurements were carried out on mechanically polished, O-terminated and H-terminated electrodes. The redox inner-sphere markers [Fe(CN)6]3−/4− and dopamine/dopamine-o-quinone and the outer- sphere marker [Ru(NH3)6]3+/2+ were used for electrochemical characterization of the polished electrode surface by cyclic voltammetry. As the B/C ratio increases for inner-sphere redox systems, ΔEp values decrease due to the acceleration of heterogeneous electron transfer kinetics. The high values of potential differences indicated that DA behaves quasi-reversibly at BDD electrodes. The ΔEp values for [Ru(NH3)6]3+/2+ showed no trend, confirming the independence of this marker at the doping level. Repeatability was further investigated when several measurements are performed in succession, with the DA solution being stirred before each measurement. It was found that when the solution is stirred, the current response does fluctuate (sr = 9.20), but the potential shows very low...
Voltammetric determination of sulfathiazole on boron doped diamond electrode
Zich, Mikuláš ; Fischer, Jan (advisor) ; Schwarzová, Karolina (referee)
This bachelor thesis is dedicated to study of the electrochemically active substance sulfathiazole. Sulfathiazole is an antibacterial agent added to animal feed. As a result, it contaminates soil, water bodies and food. A boron-doped diamond electrode was chosen as the working electrode for voltammetric determination. First, the optimal pH for the study of sulfathiazole in the range of 2 to 12 by the technique differential pulse voltammetry was determined and the optimal pretreatment of the electrode before measurements. Subsequently, the calibration dependence was measured. Sulfathiazole was successfully dissolved in water at a concentration of c = 1·10-3 mol l-1 and Britton-Robinson buffer was used as the electrolyte. As the optimal pH was determined pH 6,0. At this pH, a relative standard deviation of sr = 5% was achieved. Standart deviation of 5 % has been achieved when the working electrode was polished before each measurement. Under these conditions, was calibration dependence measured in concentration range from 0,1 to 100 μmol l-1 . Linear dynamic range was calculated between 0,1 - 20 μmol l-1 . Limit of quantitation was found to be 97 nmol l-1 .
