This bachelor’s thesis deals with neural network and their use in speech recognition. Firstly, there is some theory about speech recognition, afterwards we show theory around neural networks in connection with connectionist temporal classification method. In next chapter we introduce toolkits, which were used for training of neural networks and also experiments done by them to find out impact of connectionist temporal classification method on precisionin phoneme decoding. The last chapter include summarization of work and overall evaluation of experiments.
Security Integration Processes in the Post-Soviet Space
Švec, Jan ; Rolenc, Jan Martin (advisor) ; Cibulková, Petra (referee)
The thesis deals with the topic of security integration processes in the post-Soviet space, and focuses on the Collective Security Treaty Organization (CSTO). The aim of the thesis is to specify the character of the CSTO from the perspective of collective security theories. The first part of the thesis studies different approaches to the concept of collective security. The result of the first part is the specification of criterions, which allow to identify collective security organization. The second part describes the characteristics of post-Soviet space and integration processes in the post-Soviet space. The third part analyses the Collective Security Treaty Organization. The conclusion provides arguments, why CSTO is not a collective security organization and why it should rather be considered as a collective defense organization.
The Effect of Bulkiness of Non-peripheral Substituents of Phthalocyanines on their Absorption and Photophysical properties
Paušímová, Zuzana ; Nováková, Veronika (advisor) ; Švec, Jan (referee)
Charles University in Prague, Faculty of Pharmacy in Hradec Králové Department Department od Biophysics and Physical Chemistry Student Zuzana Paušímová Supervisor doc. PharmDr. Veronika Nováková Ph.D. Title of thesis The Effect of Bulkiness of Non-peripheral Substituents of Phthalocyanines on their Absorption and Photophysical Properties Phthalocyanines are planar macrocyclic compounds which absorb the light over 670 nm due to their large system of conjugated double bounds. Peripheral substitution of phthalocyanines significantly affects the position of the main absorption band. Absorption at longer wavelengths is highly advantageous in any biological applications because such light does not interfere with endogenous chromophores. It was discovered that the desired red-shift of absorption band is much more pronounced at non-peripherally substituted phthalocyanines. However, it was recently found out that the presence of bulky non-peripheral substituent shifts significantly absorption maximum back to lower wavelengths. The aim of this study was to explain this phenomenon more in detail. For this purpose, a series of phthalocyanines bearing non-peripheral (alkyl- or arylsulphanyl) substituents of different bulkiness was prepared. Syntheses included nucleophilic substitution leading to appropriate...

Krepsová, Veronika ; Nováková, Veronika (advisor) ; Švec, Jan (referee)
Charles University in Prague, Faculty of Pharmacy in Hradec Králové Department Department of Pharmaceutical Chemistry and Drug Control Candidate Mgr. Veronika Krepsová Supervisor PharmDr. Veronika Nováková, Ph.D. Title of Rigorous Thesis Synthesis and Study of Spectral and Photophysical Properties of a New Water-soluble Azaphthalocyanine Bearing Saccharide Units on the Periphery Azaphthalocyanines (AzaPc), the aza-analogues of phthalocyanines are macrocyclic compounds with large conjugated system giving them interesting spectral and photophysical properties useable in photodynamic therapy. An important condition for their use in this application is to provide solubility in an aqueous medium without losing their photodynamic activity. Water solubility can be achieved by the introduction of hydrophilic substituents on the periphery of the macrocycle. Hydrophilic substituents may be charged (anionic, cationic) or uncharged (e.g. hydroxyl-bearing). The aim of this thesis was to prepare and to study the properties of the water-soluble AzaPc bearing uncharged substituents on periphery, concretely saccharide unit, leading to AzaPc with 32 hydroxyl groups on the periphery. Zinc was chosen as the central metal because the zinc AzaPc are stable and responsible for good photophysical properties....

Structures with influence on melting point of DNA duplexies
Vondrková, Nela ; Miletín, Miroslav (advisor) ; Švec, Jan (referee)
This work is research dealing with binding agents in the minor groove of DNA. Minor groove binders (MGBs) are agents with the ability to specifically bind to specific DNA sequences. Together with DNA they form a stable complex. The degree of stability of this complex can be assessed from the change in the melting temperature of ligand/DNA complexes compared with free DNA. The structure of DNA is described in relation to its function and its importance. The basic properties of MGBs are mentioned and how they bond in the minor groove, i.e. the rules used by these agents for "reading" a DNA sequence. An overview is provided of the types of chemical structures, in particular the natural origin described in literature as MGBs. They are most commonly described with the occurring compounds, mainly those with clinical significance. The MGB-BP3 compound is among the most interesting compounds with antibacterial activity, which is effective against the most common pathogens of nosocomial infections. Further development of the CC-1065 analogues is also beneficial as anticancer drugs. Trabectedin has already been approved for use in cancer patients, and brostallicin and irofulven are undergoing clinical evaluation. It is also worth mentioning the development of furamidine prodrugs with anti-protozoal activity....
Photophysical and Photochemical Study of Metal Free Phthalocyanines and Their Azaanalogues
Ludvová, Lucie ; Nováková, Veronika (advisor) ; Švec, Jan (referee)
Charles University in Prague, Faculty of Pharmacy in Hradec Králové Department: Department of Biophysics and Physical Chemistry Candidate: Lucie Ludvová Supervisor: PharmDr. Veronika Nováková, Ph.D. Title of Diploma Thesis: Photophysical and Photochemical Study of Metal Free Phthalocyanines and Their Azaanalogues This work deals with phthalocyanines and azaphthalocyanines that are macrocyclic compounds with large conjugated system giving them interesting photophysical properties. The aim of this work was to prepare new phthalocyanines and azaphthalocyanines and complete a previously prepared series of compounds. Concretely, the compounds with peripheral substituents connected through oxygen (tert-butoxy for azaphthalocyanines and butoxy and tert-butoxy for phthalocyanines) were intended to prepare. The work was mainly focused on the synthesis of intermediates (4,5-dibutoxyphthalonitrile and 4,5-di-tert-butoxyphthalonitrile), that are precursors for the preparation of phthalocyanines. 4,5-disubstituted phthalonitriles with a substituents bound through heterotom are usually synthesized using nucleophilic substitution of 4,5-dichlorophthalonitrile. However, this procedure was not successful in our case. The method was changed and 4,5-dibutoxyphthalonitrile was successfully prepared from pyrocatechol,...

Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Pharmaceutical Chemistry and Drug Control Student: LukášMaršík Supervisor: Mgr. Jan Švec, Ph.D. Expert adviser: PharmDr. KamilKopecký, Ph.D., doc. PharmDr. Miroslav Miletín, Ph.D. Title of diploma thesis: Derivatives of carbazole and their influence on melting point of DNA duplexes I. This diploma thesis deals with preparation of carbazole derivatives bearing bulky basic groups in positions 3 and 6 and an aliphatic chain terminated with azide group in position 9. In the theoretical part, there are described possible methods, how to create basic functional groups on carbazole and other aromatic structures, further the possibility of establishing an aliphatic chain on the aromatic nitrogen and in the last part creation a basic centres on aromatics with a functional group was explored - mostly bromide derivatives were employed as the starting structure. The experimental part deals with the application of methods described in the theoretical part on the structure of carbazole. As the starting molecule 3,6-dibromo-9H-carbazole was used; methods of exchange bromine by carbon or nitrogen containing groups were tested, followed by the creation of the aliphatic chain on the nitrogen of carbazole terminated with azide group and...
