Role of FtsH proteases in the cyanobacterium Synechocystis sp. PCC 6803

This thesis focuses on the functional and structural characterization of FtsH proteases in Synechocystis PCC 6803. One of the aims was to determine localization and subunit organization of FtsH homologues in Synechocystis cells using GST and GFP tagged FtsH derivatives. The main result of the thesis is identification of two FtsH hetero-oligomeric complexes and one homo-oligomeric complex in Synechocystis cells. The large part of the thesis is aimed at establishing the role of the first hetero-oligomeric complex, FtsH2/FtsH3, in quality control of Photosystem II and at identification of a mechanism, how its substrate proteins D1 and D2 are recognized. Another part is dedicated to characterization of the second hetero-oligomeric complex, FtsH1/FtsH3, which consists of two essential FtsH homologues and which is here identified as an important regulatory element in maintaining iron homeostasis.
Centre of rotation stability setting for the INFINIA camera

My bachelor thesis focus on Centre of rotation stability for Infinia cameraAt my work I got closely familiarize with scintillation camera Infinia Hawkeye, which is located in the department of nuclear medicine in the České Budějovice. The aim of my work was to determine whether the centre of rotation (C.O.R.) takes place around a fixed axis, and thus whether there is not movement. The method that I chose to accomplish my goals, was to test the camera , which was performing by 360° acquisition around a point source 99mTc. The point source was fitted to a rod and placed on the Infinia camera on patient's pallet. Testing was carried out with two collimators LEHR and LEGP in modes H and L. The testing was performed by radiological physicist, always in the afternoon to ensure smooth running of the workplace. The test confirmed my hypothesis that the value will be within the values specified by the manufacturer. Testing was performed once a month for half a year. In In this days because we are apperceiving tremendous technological progress, and devices which allow us to produce more perfectly and detailed images and contribute to better diagnosis and treatment are created it is very important to ensure the quality control. Over the last few years progress in gamma camera designs , have improved all aspects of image quality, particularly in tomographic images. As the system complexity increases, it becomes more important that all nuclear medicine employees are able to recognize the various types of aberrations or artefacts that can occur in nuclear medicine imaging systems. The primary purpose of the quality control program in department for nuclear medicine is to verify that the images accurately reflect distribution of the radiopharmaceutical within the patient. The purpose of quality control is detect changes of gamma camera system that may adversely affect the interpretation of clinical studies. There are large number of factors that contribute to the final quality of the image ( including uniformity, resolution - both intrinsic and energy, pulse frequency, centre of rotation system, rotational stability of the detector head). On a everyday examinations there is a limited amount of time that can be reasonably devoted to quality control system. Hence the main goal of quality control program should be to monitor those parameters that are most sensitive to changes in system performance and are most likely to impact clinical studies. Therefore , the main objective of quality control program should be monitor the parameters that are most sensitive to changes in system performance and most likely have a impact on the clinical studies. Specials series of quality control tests were designed to ensure the optimal performance of the camera. Over the whole time of the entire period of clinical operation, technicians or physicists regularly perform various quality control tests, some daily, others weekly or monthly. The purpose of these tests is monitor day to day camera performance and detect changes from the baselines values. Additional quality control test may be performed to compare characteristics of different systems and to optimize the quality of clinical studies.

Quality control and homogeneity testing of daily time series of eca ECA&D

For any meaningful climate analysis, it is necessary for analysed time series to be homogeneous, which means that their variations are caused solely by variations in weather and climate (Conrad and Pollak 1950). Th us, prior to any analysis, the need to homogenize data and check their quality arises. Unfortunately, most of the climatological series that span over decades, to centuries, contain inhomogeneities caused by station relocations, exchange of observers, changes in the vicinity of the stations (e.g. urbanization), changes in instruments, observing practices (e.g. diff erent formulas for calculating daily means, diff erent observation times), etc. In this work we focused on testing the quality and homogeneity of daily data produced by ECA&D. Th is is a free available dataset of daily meteorological elements from the European Climate Assessment & Dataset (http://eca.knmi.nl/). Th is database was used to create a regular grid of EOBS points, which are oft en used to validate climate models.
Inspesting the sensitivity and resolution of the CT-part of the Infinia-Hawkeye

The medical discipline of nuclear medicine is characterised by the constant development and perfection of the appliances used and thus, also higher diagnostic yield. The most frequently used equipment currently includes hybrid appliances enabling image fusion, e.g. SPECT/CT. The result is a recording combining anatomical and functional information allowing for more accurate localisation of the lesion and better spatial orientation. Equipment in nuclear medicine is subject to inspection under the regulation of the State Office for Nuclear Safety No 307/2002 Coll. On radiation protection and as amended under Regulation No 499/2005 Coll. When inspecting the characteristics of an appliance, procedures specified by the manufacturer are met, and each workplace performing nuclear medicine must follow a sophisticated system of quality control stipulating the individual tests and duration of the cycle when they shall be performed. My work consisted of performing quality tests on the CT-part of the Infinia-Hawkeye system. The testing was performed once every three weeks over a period of seven months at the Department of Nuclear Medicine in České Budějovice hospital. The results of individual readings were recorded in overview charts and compared with the parameters provided by the manufacturer. The performance of the comparison confirmed the defined goals, all the recorded values agreed with the limits set by the manufacturer. These values were never exceeded during the entire testing period. No short-term changes of parameters were recorded. The bachelor?s thesis can be used as an overview of quality tests of the CT-part of the Infinia-Hawkeye system and can also serve as instructions for performing these inspections at all workplaces that are equipped with the Infinia-Hawkeye hybrid system.
