Bukatová, Helena ; Lázníčková, Alice (referee) ; Kubíček, Vladimír (advisor)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Biophysics and Physical Chemistry Candidate: Helena Bukatová Supervisor: Ing. Vladimír Kubíček, CSc. Title of diploma thesis: Interaction of Tensides and Organic Dyes in Solutions II This diploma work is devoted to examination of the aqueous solutions of surfactants and organic dyes in the solution of Britton-Robinson buffer by two spectral methods: UV-VIS absorption spectrophotometry and fluorescence spectroscopy. There are two different dye-surfactant systems investigated: neutral red with sodium lauryl sulfate and phenol red with hexadecyl-trimethyl-ammonium bromide. The possible interactions were observed in the several surfactant concentration levels of the solutions with increased dye concentration. The task of my work was investigation of changes in dye spectra as a consequence of the dye-surfactant interaction and determination of the possible origin of the spectra changes. The results showed that absorption spectra of solutions containing neutral red itself differ sharply from those taken for solutions of neutral red and the tenside. The different character of the spectra is observable from the concentration of the tenside 6∙10-5 mol/l. Two types of dye-tenside associate are probably formed. The associate...

Molecular mechanisms of apoptosis induction in tumor cells
Koc, Michal
This work has focused on study of processes involved in apoptosis induction by various stimuli in cancer cells. In our case we induced programmed cell death in cancer cells by iron deprivation, commercially available taxanes docetaxel (Taxotere® ) and paclitaxel (Taxol® ) and application of novel porphyrins derivates P(-CD)l and P(-CD)2. From presented publications we can conclude: Studies dealing with iron deprivation clearly demonstrated participation different signal pathways which lead to apoptosis induction in mouse and human cancer cell lines. We used for study of apoptosis induction B lymphom cell line sensitive to iron deprivation in both mouse and human experiment model. In the case of sensitive mouse tumor cells (38C13), we have described a significant portion of the apoptotic pathway leading to apoptosis induction by iron deprivation. Iron deprivation in these cells leads to translocation of proapoptotic Bax protein from the cytosol to mitochondria which is succeeded by mitochondrial membrane potential disruption due to mitochondrial permeabilization, cytochrome c release from mitochondria, activation of caspase-9 and subsequent activation of caspase-3. In the case of sensitive human tumor cells (Raji) we detected as well apoptosis induction by iron deprivation, however, in character of...

Antioxidační kapacita a fenolické látky v révovém víně
Mateiciucová, Petra
This thesis is focused on monitoring the health beneficial substances with phenolic character and antioxidant activity sampled from leaves, lateral shoots, stems, berries, juice and wine. There were 4 interspecific varieties selected for the monitoring. These varieties are characterized by an increased resistance to fungal diseases. The monitoring was carried out for 3 consecutive years. Samples were taken from healthy and infected plants in three sampling periods, so it was possible to assess the effect of maturation on the antioxidant activity and the content of phenolic compounds. Samples were analyzed on EPR (antioxidant activity determination), spectrophotometer (antioxidant activity, total phenolics) and assayed by HPLC (determination of selected phenolic compounds, especially trans-resveratrol). The results show that all samples exhibit antioxidant activity and contain a wide range of phenolic compounds. All the samples were analyzed for trans-resveratrol, which is known for its beneficial health effects. The content of resveratrol ranged from 9.07 to 566.14 mg.kg-1 of lyophilized material. The highest content was found in the stems. Antioxidant activity determined by spectrophotometry ranged from 308.0 to 1357.7 mg GA.kg-1 in berries, and from 44.0 to 839.5 mg GA.l-1 in wines. Decreases of free radicals representing the antioxidant activity, which were measured by EPR, ranged from 1.21 to 57.17 % in the leaves. Blue varieties showed higher antioxidant activity. There was a positive correlation shown between the content of trans-resveratrol in the stems in interspecific and European varieties. It also demonstrated a dependence of the antioxidant activity on the content of phenolic compounds in the samples.

Disordes of speech formation in patients with clefts
Haas, Michal ; Dušková, Markéta (advisor)
The reason for choosing the topic for my thesis is the application of the individual opportunities for communication and exchange of information. Therefore, care about communication disorders one of the priorities of medicine of the 21st century. Also in patients with cleft lip and palate in currently evaluating treatment success based on the quality and extent of speech removing marks from defects in appearance. Cleft lip and palate is one of the most common birth defects (in the Czech Republic on 1 500 births). Their treatment in the Czech Republic has a long tradition associated with the name of academician Frantisek Burian, who many original procedures enriched operational methodology injuries and malformations of the face, especially clefts. In addition to affecting speech has morphological character defect negative functional effects on swallowing, breathing and chewing. Aesthetically it is a facial disability Triangle, which most affects the overall appearance of the face. Full rehabilitation of these patients require multidisciplinary treatment, which starts immediately after birth and continues into adulthood. The complex treatment gradually, depending on the postnatal development, health and growth of the victim, participate in specialized team, which is based plastic surgeon, dentist...

Biosystematic studies in the family Cyperaceae
KOŠNAR, Jan
The thesis was focused on the microevolutionary mechanisms that contribute to morphological diversity in selected members of the sedge family (Cyperaceae). Natural hybridization, evidenced from both morphological characters and molecular markers, was revealed to be a potentially important source of diversification in the tropical spikerushes of Eleocharis subgenus Limnochloa. High levels of phenotypic plasticity of clonal growth but rare genetic (ecotypic) differentiation among contrasting morphotypes were found in the polymorphic species Carex nigra, which implied that taxonomic splitting of the species was unreasonable.

Self-image of Women Undergoing Cosmetic Surgery
Karalová, Lucie ; Chloubová, Helena (advisor) ; Marková, Eva (referee)
The submitted diploma thesis deals with the issue of higher psychosocial needs and particularly focuses on the need of self-conception. The aim of the thesis is to try to find out, why women undergo plastic aesthetic operations, what their motives are, what they are led by, mostly influence and attempt describe their character. The thesis is divided into two parts, theoretical and practical. In the theoretical part, basic aspects of human needs are generally characterized and the issue of higher psychosocial needs with respect of selfconception is specified. Next, the terms of aesthetics, beauty and the ideal of womanly beauty are dealt with. Psychotherapeutic approaches, as one of the potential ways how to solve one's dissatisfaction with oneself and, another potential way, aesthetic surgery were taken into account. For the practical part, the research method of a survey/questionnare was chosen. It comprises 66 interviewees staying in hospital at plastic surgery clinic at FNK.V in Prague and the private plastic surgery sanatorium in Kolin. The researched sample of respondents was chosen in terms of receiving as wide view of the whole issue as possible. The aim of the research was to validate or invalidate the set hypothesis. Although the conclusions cannot be generalized on basis of such a small sample of...

Molecular identification of purebredness and kinship of the Philippine crocodile (\kur{Crocodylus mindorensis}) and the Cuban crocodile (\kur{C. rhombifer}) for \kur{ex situ} conservation management
STRAKOVÁ, Helena
The current status of crocodilians recognizes them as a group under serious threat due to their habitat destruction and illegal poaching for their lucrative products. In addition to these threats, the elimination of spatial and temporal boundaries through modern anthropogenic pressures has facilitated hybridization in crocodiles by bringing together species that would otherwise not breed due to lack of opportunity. Here analyzed crocodiles, the Philippine crocodile (Crocodylus mindorensis) and the Cuban crocodile (Crocodylus rhombifer) are critically endangered and listed in CITES Appendix I. This study deals with a significant portion of the Philippine and Cuban crocodile captive population in Europe based on mtDNA, nucDNA and microsatellites. The species genetical purity of 13 specimen of C. mindorensis was determined on the ground of testing maternally inherited mitochondrial gene cytochrome b and and D-loop loci from the mitochondrial control region as well as two nuclear markers, LDHA and C-mos. Also the purity of 11 out of mentioned 13 individuals on the basis of several (7) microsatellite loci, while possibly confirming a supposed hybrid origin of two crocodiles with mixed morphotype. Then a purity of 4 samples of the Cuban crocodile was verified with cyt b gene and nuclear DNA purity of all 7 tested individuals with LDHA gene and 13 microsatellites. Based on the obtained genetic characters I proceeded a likely kinship of the two groups of crocodiles and subsequently suggested an optimal breeding management within the ex-situ conservation

Optimization of conditions and procedures for plant extraction.
SMUTNÍKOVÁ, Kateřina
The thesis deals with the content of selected phenolic compounds in some species of the genus Amaranthus, in black elderberry (Sambucus nigra L.) and buckwheat (Fagopyrum esculentum M.). Phenolic compounds are a group of natural compounds exclusively vegetable character. Flavonoids represent only one group of phenolic compounds. Flavonoids show many positive biological effects, in particular act as antioxidants. Natural flavonoids may cause to prevent from coronary- heard diseases and other diseases associated with older age. In recent years the increased attention is paid to flavonoid investigation due to its biological effects. For the determination of phenolic substances there were used two independent analytical methods. There are the high-performance liquid chromatography (HPLC) and micellar electrokinetic capillary chromatography (MECC). The MECC method was used for determination rutin and free quercetin. The highest content of rutin was found in leaves of buckwheat (76,400 mg/kg of dry weight) and the lowest content of rutin was determined in buckwheat hulls. The highest content of rutin was observed in teas from buckwheat leaves and inflorescence. This amount of rutin corresponds with rutin content in more than two pills of Ascorutin (the most favourite flavonoid medicament in the Czech Republic) The HPLC method was used for quantitative determination of phenolic acids. The content of free quercetin was monitored in all samples. No free quercetin was found both in plant material and in samples of teas. The ethanolic extract from the elderberry inflorescence didn´t contain any free quercetin. Free quercetin wasn?t found in any further samples of teas, which were prepared by described methods.
