Utilization of confocal microscopy to study of degradation of organic dye in cell biology

This thesis focuses on the study of the life of a fluorescent dye. It also examines changes in its emission spectrum determined with the aid of development of fluorescence intensity over the long term sensing in several kinds if of cella. The general part of the thesis with the issues of confocal microscopy, fluorescent radiation and maks. Furthermorer, an analysis of methods for measuring fluorescence lifetime is done. In the thesis there is elaborated a description of the program, whitch was designed for the analysis of the collected output of the used confocal microscope. Sebsequently, the results are evaluated.

A scanning low energy electron microscope (SLEEM) was realized with a cathode lens in which the negatively biased specimen is used as the cathode. In this arrangement, electrons pass through the microscope at high energy and are decelerated to low energy before landing on the sample. This design brings plenty of signal even in the landing energy range of tens or units of eV. However, the use of the primary electrons with low energy brings some problems e.g. low source brightness, increased aberrations and sensitivity to stray fields. The scanning Auger microscopy (SAM) is well-established experimental method. A combination of SAM and SLEEM in one device would therefore provide a sufficient tool to solve the problems inherent in these individual methods. Although much has already been achieved in the area of detection of slow electrons, none of the methods currently known is suitable to be built into a scanning illumination column for Auger microprobe analysis

Fabrication of Nanowires into Porous Alumina by Electrolysis

This thesis deals with preparation of metal nanowires based on electrodeposition from aqua solution of metallic salts into porous dielectric aluminium oxide templates. The theoretical part covers various bottom-up methods of nanowire preparation using a template, electrochemistry and electrodeposition overview. The Experimental part is focused on the nanowire (Ag, Cu, Ni-H) preparation by means of direct current and alternating current electrodepositon and geometrical characterization of fabricated nanowires by scanning electron and atomic force microscopy. Defects of commercially available and prepared porous alumina templates are mentioned. Branching and coalescence of nanowires are briefly discussed as well.

New diagnostic and therapeutic aspects of inflammatiory cardiomyopathy

Introduction: Inflammatory cardiomyopathy (DCMi) represents a non-familial form of dilated cardiomyopathy (DCM) and endomyocardial biopsy (EMB) is crucial for its diagnosis. Aims: To assess the prevalence of DCMi in patients with DCM of unclear origin, to evaluate the significance of serological tests for antibodies against infectious cardiotrophic agents and to analyze the effect of specific therapy guided by EMB results. Methods: EMB was performed in 56 subjects (mean age 52 ± 10 years) with DCM of unclear etiology and left ventricular (LV) ejection fraction (EF) < 40% with a history of heart failure less than 1 year. EMB samples were analyzed by immunohistochemistry, polymerase chain reaction (PCR) and electron microscopy. Results: Immunohistochemical examination revealed myocardial inflammation in 26 patients (46%), the PCR method detected genome of microbial agents in 32 patients (57%). Electron microscopy showed the presence of particles of microbial agents in 41 patients (73%). Serological blood tests found no IgM antibody positivity against any of the investigated microbial agents. Targeted antibiotic therapy in patients with evidence of Borrelia burgdorferi (Bb) genome in the EMB led to a reduction in LV size, improvement of LV EF and alleviate symptoms of heart failure. Mechanical properties and structure of the aluminum alloy after heat treatment

The presented bachelor´s thesis deals with the influence of heat treatment especially artificial aging on mechanical and structural properties of AlSi7Mg0.6 alloy. Cylindrical test bars of diameter 6 mm and 9 mm cast into a self-supporting ceramic shell made by plaster investment technology were used as experimental samples. These test bars were cast by Alucast whereas the patented technology of casting- JF casting was used. On the test bars was carried out the T6 heat treatment according to the Standards AMS 2771C. After the solution annealing were test bars divided to the six groups and put through various time of artificial aging, namely from 2 to the 12 hours. In order to evaluate the effect of the time of artificial aging on the structure and mechanical properties of both diameters of the test bars, the method of light microscopy, Brinell hardness test and static traction test were used. Also SDAS quantification was carried out by means of image analysis. On the grounds results of experiments it is possible to state, that the best values of mechanical properties of both diameters of the cylindrical test bars were reached after 10 hours of artificial aging.


Effect of preparation conditions on the structure and properties of hybrid light curing composites

The purpose of this thesis was to implement and evaluate series of experiments focused on the impact of mixing conditions and component dispensing on the structure and properties of hybrid polymer composite materials for resistant dental fillings. There were suggested some preparation methods and subsequently some mechanical properties of prepared samples were measured. Short non-formable polymer fibers in combination with filler particles of various particle sizes were added into the matrix. As matrix a polymer, light curable monomer mixture based on urethandimethacrylate and dodecandioldimethacrylate was used. Tensile deformation curves of prepared samples were measured. Morphology of prepared composites was observed by surface fractography using scanning electron microscopy (SEM). Samples were evaluated by comparing of tensile curves and morphology observed on SEM slides. The best way of component mixing and dispensing was evaluated based on comparison of experiment results and properties predicted by existed models.
