Biosensors for Environmental Monitoring and Biomedical Applications

Study of biosensors has become an essential part of research in biotechnology. Biosensors as fast, portable, highly sensitive, and low-cost bioanalytical detection devices have been utilized in many fields of human activity. The first part of the presented work focuses on electrochemical biosensors for rapid environmental screening of herbicides as water pollutants. A sol-gel immobilization method for a photosystem II (PSII) complex is studied in order to enhance the sensitivity and the signal strength and stability of a PSII-based biosensor. Computer simulations of a PSII biosensor are employed with the aim to find out how the immobilization membrane properties influence the biosensor parameters. Newly developed immobilization by a thin-layer membrane based on the results of computer simulations and revised measurement protocols are presented. The second part of the work is devoted to synthesis and electrochemical detection of newly developed metal labels for electrochemical immunosensors. The synthesis of dendrimer-encapsulated silver nanoparticles and biorecognition properties of biotin-nanocomposite conjugates are discussed. For detection of synthesized labels, a microfluidic detector was manufactured and tested and different approaches to packing of a microfluidic chip employing polydimethylsiloxane (PDMS) were investigated. Newly designed microstructures for a microfluidic separator of magnetic beads (MBs) were studied by computer simulations. The separator was made and trapping of MBs for the further employment in MBs-based immunoassays are presented

This diploma thesis offers to the reader the basic overview of the most used methods of computer simulation and accessible software which can be used for this purpose. The aim of the thesis is not a detailed description of individual procedures, but rather a creation of overview text which can be used by the reader as a material for the primary orientation to the given problems. The thesis is outlined as a proposal of the themes of lectures of the subject ?Computer simulation and calculations in technology?. The thesis is divided into three thematic blocks. In the first block, the reader is acquainted with the programming language MATLAB, with its possibility of application in study as well as in practice. The second block acquaints with the CAD systems and the last block presents a nearer usage of individual software applications during the solution of selected model situations.
Comparison of Simulation Software and Areas of Its Application

This diploma thesis deals with computer simulation. In the opening, there are descriptions of theoretical aspect of computer simulation, simulation model structure and individual phases of simulation process. After that follow areas of application, characteristic features of each area and classification of simulation models, that are used in those areas. In this thesis only 4 areas of application were chosen: automotive industry, logistics and transportation systems, healthcare and military simulation. Each application area contains descriptions of a few chosen simulation software products that are used in this area. At the close there is a price comparison of described simulation products, their usage in Czech Republic and their development evaluation.
Simulation of hospital evacuation

This diploma thesis deals with security in the hospital enviroment. The work creates a new evacuation plan for large-monoblock Hospital for adults in FN Motol. Evacuation plan covers all activities of the announcement of evacuation after the removal of the last patient from the hospital. For developed evacuation plan at workcompiled four simulation models in SIMUL8. With these models, theevacuation plan and tested models provide a quantitative value, according to which it can better prepare for possible evacuation. This thesis has fulfilled its purpose -- was created functional evacuation plan, and simulation models provide additional information as to which should be ensured smoother running of the evacuation.
Methods computer simulation

This text includes analyses of fundamental techniques that are often used for creating mathematic models in the computer physics. The text is divided into four basic chapters. The first chapter brings the definition of computer models and of computer simulation. The following chapter deals with particle, linking and hybrid techniques of the computer modeling. In the end the text describes the most used methods of the computer simulation, i.e. the method of molecular dynamics and the Monte Carlo method.
Computer simulation of up-hill diffusion in ČSN 17 242/P91 welded joint

The DICTRA software is used for simulating diffusion in multiphase diffusion couple ČSN 17242/P91 at temperature ranging 500-1100 C. The examples of calculated phase profiles, activity and concentration profiles of all species at selected condition are shown.
Analysis of computer simulation of sound and its limits

This thesis is focused on the question whether it is possible to fully simulate the sound of real musical instruments on the personal computer. The opening chapters deal with basic characteristics of sound information, its perceiving and also its digital form. Futher there is an elaborate analysis of computer simulation in comparison with legitimate records. The output of this part are worded assumptions. The next parts offer a detailed look at programming approaches and methods of computer sound creation. The research leads to a hearing test in which author tries to confirm the hypotheses as well as to answer the primary question.
