Separation of selected acidic proteins fractions from potato (Solanum tuberosum L.) tubers by chromatographic techniques
LORENC, František
This diploma thesis deals with acidic proteins contained in the potato (Solanum tuberosum) tubers, or rather about their separation with chromatography techniques. For the analysis were chosen tubers of two czech potato varieties, Adéla and Westamyl. From the concentrated potato juice were eliminated basic and most of the patatin proteins with the gravity column chromatography. In the next step were applied the chromatography techniques on the anion and hydroxyapatite columns by the fast protein liquid chromatography (FPLC) system. On the anion column were separated protein fractions which contained proteins of molecular weight in the range of 15 kDa to 60 kDa (Adéla) and 15 kDa to 100 kDa (Westamyl). The most abundant were the proteins with molecular weight 15 kDa a 20 kDa. In the last step was used the mass spectrometry for the identification of chosen protein fractions.

The reactivity of Na and salt of oxidized cellulose and its oligosaccharides with chitosan will be presented. Hydrolysis of Na and salts of oxidized cellulose was carried out in homogenous system by means with microwave irradiation at constant conditions. The reaction progress was monitored by capillary isotachophoresis and size exclusion chromatography. Reactivity of oxidized cellulose or its hydrolysates with NH2 groups of chitosan was promoted by MW irradiation. Reaction was monitored by infrared spectroscopy.

Možnosti on-line spojení extrakce a chromatografie v analýze složitých vzorků
Hyötyläinen, T. ; Lüthje, K. ; Rautiainen-Rämä, M. ; Pól, Jaroslav ; Ranta-Aho, O. ; Riekkola, M. L.
In this study, on-line methodologies have been developed both for liquid and solid food samples. For liquid samples, microporous membrane liquid-liquid extraction (MMLLE) has been combined on-line with gas chromatography (GC). This MMLLE-GC method has been applied for the determination of pesticides in wine and water. The method has been extended to the analysis of solid samples as well, by combining a pressurised hot water extraction (PHWE) to the MMLLE-GC system. The PHWE-MMLLE-GC has been applied for the analysis of pesticides in grapes. For thermally labile analytes, on-line combination of supercritical fluid extraction (SFE) and liquid chromatography has been constructed and it has been applied for the analysis of lycopene in fruits. All the method developed proved to be linear, repeatable and sensitive
