Immobilization of protease V8 on magnetic particles for application to proteolytic cleavage of pepsin A

This thesis is part of a long-standing research in the field of diagnosis of the stomach diseases, which is based on the gastric enzyme pepsin A mapping. It was found that a phosphorylation in the primary structure of this enzyme may serve as a marker of incipient stage of carcinogenesis. This thesis is focused on the immobilization of protease V8 isolated from microorganism Stafylococcus aureus to magnetic agarose beads. Protease V8 is a promising candidate for producing peptide maps of pepsin A. The influence of pH, temperature and reaction time on the enzyme to activity has been studied and the optimal conditions for hydrolytic catalysis of formation of peptide fragments of pepsin A.

Development of GC Metod for the Determination of Vitamine E Acetate in Nutritive Supplements

Title of Thesis: Development of GC metod for the determination of vitamine E acetate in nutritive supplements A GC method was developed for the determination of content of vitamine E acetate in nutritive supplements. The method was optimised and validated. The column Capillary GC, Equity Columns, Equity TM - 5 poly (5 % bifenyl / 95 % dimethylpolysiloxan); 0,32 mm ID x 30 m; 1,0 mm dr was used for the analysis. Helium was used as a carrier. Split was 1 : 100. Flame-ionization detector was used for detection. Optimal conditions for analysis: temperature of column 340 řC, temperature of injection 340 řC, temperature of detector 320 řC, flow-rate 2 ml/min. Fenoxycarb was chosen as an internal standard. The method was used for an analysis of vitamine E acetate in nutritive supplements Geladrink Forte pulverized drink - pineapple and Chondrotin MSM 2600. Determined concentration of vitamine E acetate was re-counted to the content of vitamine E declared by producer.

In this thesis the analysis of second phase particles influence on recovery and recrystallization processes in twin-roll cast sheets of AA 8011 aluminium alloy based on Al-Fe-Si is studied. The role of arrangement of impurity atoms and the type of thermomechanical treatment is investigated. Unbalanced crystallization of solid solution, which occurs during twin-roll casting, results in the creation of complicated structure of second phases, which isn't as a rule homogenously distributed in the volume. Therefore these materials exhibit poor mechanical properties in comparison with conventionally cast alloys. Important question the materials science deals with in the last years, is to find such a thermomechanical treatment, which results in balanced phase composition and optimal mechanical properties, comparable with properties of products produced by conventional methods. Such properties are especially high formability combined with sufficient strengths. To study physical properties of the alloy the resistometric measurement during linear heating, thermo-electric measurement and hardness measurement were used. Evolution of structure and microstructure in the alloy during heat treatment was studied by means of optical microscopy and transmission and scanning electron microscopy.

Determination of the kinetics of crushing industrially produced tablets

7.2.Summary Determination of the Kinetics of Crushing of Mass-Produced Tablets The degree paper aimed to determine the optimal parameters of the devices for the evaluation of the strength of tablets. Rates of preloading from 0.01 to 0.1 mm/s, preloading from 1 to 4 N, and rates of the cycle from 5 to 20 N/s were tested. These parameters were evaluated on the basis of the records of the force-course. Deformation curves were employed to calculate the course of crushing in mm, crushing force in N, and energy of crushing in N.mm. The reading of the curves was evaluated as well. The obtained results were evaluated using the method of the analysis of diameters following Otto. The results show that for the standard determination of mechanic resistance of tablets, the following adjustment of the device is suitable: rate of the cycle, 0.01 N/s, preloading, 4 N, rate of preloading, 5.mm/s.

Analysis of anthraquinone secondary metabolites produced by Geosmithia spp.

Analysis of anthraquinone secondary metabolites produced by Geosmithia spp. Zdena Křesinová Geosmithia species are little known fungal symbionts of bark beetles. Secondary metabolites from lilac colored species G. lavendula (strain MK 1008) and nine other Geosmithia species were investigated in order to elucidate their structures and quantify their production during submerged cultivation. Five hydroxylated anthraquinones (HAs) were isolated from culture media during submerged cultivation of the fungi and three of them were identified using NMR and MS techniques as 1,3,6,8- tetrahydroxyanthraquinone (1), rhodolamprometrin (1-acetyl-2,4,5,7- tetrahydroxyanthraquinone; 2), and 1-acetyl-2,4,5,7,8-pentahydroxyanthraquinone (3). Preparation, quantification and identification of HAs in fungal samples involved a SPE step, semi-preparative HPLC/UV and UPLC/UV methods. For optimization of analytical methods, separation qualities of two types of reversed phase sub-2-micron particle sized columns and one 5-micron particle sized column were tested. The most efficient Sheild RP C18 column filled with 1.7 µm particles was then used for quantification of HAs production during the cultivation period. Recombinant preparation of isotopically labeled receptor rNKR-P1A for NMR studies

anglicky NK cells belong to the population of cells which are able to cytotoxic kill certain tumor and cells infected by viruses. This bachelor thesis focuses on the rat's NK cell receptor, especially rNKR-P1A. This protein belongs to the activating receptors and is able to activate the cytotoxic function of NK cells. Physiological ligand and three-dimensional structure of this protein has not been resolved yet. The aim of this work was optimalization of production rNKR-P1A in minimal medium and thereafter producing isotope-labeled proteins. Isotope- labeled proteins could contribute to solving the three-dimensional structure of this rat receptor by using NMR methods.

Chromatography method optimization for HPLC determination of carotenoids.

This thesis deals with searching for optimal conditions for determination of carotenoids lutein, betacarotene and zeaxanthin. High performance liquid chromatography - HPLC was used for determination. During searching for optimal conditions many various columns and mobile phases were tested. Primary information about carotenoids separation was obtained from scientific literature. There were tested both types of chromatography - normal phases chromatography and reversed phases chromatography. The most frequently tested columns were C18 columns from different producers. The biggest problem for HPLC determination and separation of these three substance were position isomers of lutein and zeaxanthin. Separation of these substances is relatively difficult and it wasn't completely solved in this thesis. In spite of it new metod for determination and separation of three carotenoids was developed. This method used isocratic elution at column Zorbax SB C18 (50 x 4,6 mm, 2 μm), UV detection at 450 nm, mobile phase of methylene chloride:hexan:acetonitrile (2,5:2,5:95), injected volume 1 μl and flow rate at 1 ml/min.

Photochromic effect in nanoporous titanium dioxide film with Ag nanocrystals

The diploma thesis is devoted to multicolour photochromic effect in nanocomposite material Ag-TiO2, which is caused by a spectrally selective photoinduced decrease in extinction. The Thesis introduces briefly the theory of particle plasmons which are of fundamental significance for the investigated phenomenon. A short summary of the models and results published on photochromic effect in Ag-TiO2 is than given. The core of thesis deals with the optimization of this nanocomposite material towards a high photochromatic response. The influence of concentration of the TiO2 solution and of the speed of its spin coating deposition on a substrate was measured. Also the effect of AgNO3 quality used for sample preparation was monitored. A change in the extinction was found to depend on parameters of the excitation laser beam too. As a part of the optimization process, also the alternative TiO2 matrixes produced on basis of lamellar micelles were tested. The photochromic effect was monitored by measurements of the light extinction and scattering in dependence on the sample photoexposition. Juhaňáková Jana - diplomová práce - 2006 The diploma theses deal with the measurement of mechanical resistance of tablets on basis of force - displacement record. The new method compared to the classical method characterizes mechanical resistance by means of destruction and destructive energy. Theoretical part of the work describes characterization and distribution of granules and tablets, characterization of additives used for production of tablets. It deals with the starch group in detail as well. The aim the experimental work was the determination of optimal parameters for measurement of the kinetics of destruction of tablets, preloading, rate of preloading a cycle. Commercially produced tablets of Paralen 500 were evaluated. The main goal was to analyze the detection mechanisms and to find optimal system setting for detection of the given part of signal electron spectrum. It is used for maximization of the material or topographical contrast.
