Utilization of Particular Leisure Time Establishments for Children and Youth in Chosen Districts of South of Bohemia

Institutions for leisure time activities focused on children and youth are divided into two basic groups in this thesis. First group is represented by educational institutions whose activity is arranged by Act No. 561/2004 Coll., on pre-school, basic, secondary, tertiary and other kind of education (School Act). These institutions concern basic art education, language education and free-time activities. Non-state and non-profit organizations concerning upbringing and education of children and youth in their leisure time belong to the other group. Mostly, they are civil associations whose activity is arranged by Act No 83/1990 Coll. on gathering of citizens. For example, association Junák {--} union of scouts of the Czech republic, civil association YMCA (Young Men´s Christian Assiciation) or civil association Duha (Rainbow) belong there. The aim of this thesis is to describe the problems of institutions of free time activities in short and then bring information on particular organizations functioning in regions Český Krumlov and Prachatice. For this purpose the database of subjects functioning in these regions was created. From them, twenty organizations were chosen, investigated and described more detailed. Because of the aim, the method of qualitative research was used, to be specific, the technique of interview and technique of documents analysis were chosen. Recognized information is divided into six spheres, e.g. legal norm, financing, membership, focused activity, presentation for the public, staff and material funding. According to acquired information from these thematic spheres, separate institutions are compared and their common characteristics or the specifics of single respondents are checked. This work is established for everybody who wants to acquire basic information on free time institutions and know more about some of these organizations.

NOURISHMENT OF TEENAGE AND YOUTHFUL, POSSIBLE RISKS

Abstract: Nutrition of adolescents, possibly hazards My bachelor work draws attention to possible hazards on inappropriate nutrition of adolescents especially on obesity risk, mental anorexia and mental bulimia. The theoretical section is focused on fundamental information about rational nutrition, alternative nutrition and problems with occurence of obesity, mental anorexia and mental bulimia. Rational nutrition is characterized by suitable and adequate food input, ideally qualitatively and quantitatively composed. Standard for rational nutrition is so-called Nutrition pyramid. Alternative nutrition hazards are escalated by stage of strictness and quantity of food excluded. Among most frequent diet methods belongs to vegetarian and macrobiotic diet, organic nutrition and separated diet. Obesity is caused by excessive energy input and insufficient energy output. Mostly Fast Food dishes take part on obesity beginning. Regarding rational nutrition these dishes do not fulfil recomeded nutrition value of standard lunch, therefore it is possible to accept them only occasionally. Mental anorexia and mental bulimia is mental defect which is characterized namely by intentional reducing of physical weight. Patient suffered by mental anorexia can achieve of weight decrease by avoiding of dishes classifiable into foodstuff group, after which patient puts on weight. Patient suffered by mental bulimia repeatedly overeats and subsequently evokes vomit. In the practical part there are presented research results focused on issue mentioned above. Investigative section of the work is realized in a form of questionnaire, which specializes in valuation of way of adolescent catering. These questionnaire are performed for two adolescent groups. The first group consists of high school student and the second one consists of educational establishment students.

Analysis of selected polar compounds in HILIC mode

Nowadays High Pressure Liquid Chromatography is the most important and the most frequently used separation technique that enables qualitative and quantitative analysis of the complex mixtures. One of its variants is Hydrophilic Interaction Liquid Chromatography. Lately this technique has experienced an increasing boom particularly, due to its ability to improve separation of very polar and hydrophilic compounds. The adeninribonucleotides (ATP, ADP, AMP, cAMP), creatine and phosphoceratine are among these highly polar compounds that are of great importance not only for human body. Number of analytical methods has been developed and published for the analysis of these compounds; however HPLC methods using the reverse phases have been dominated. The basic overview of these methods is a part of this diploma thesis. The experimental part of this thesis is focused on the development of chromatographic conditions suitable for the simultaneous analysis of ATP, ADP, AMP, cAMP, creatine and phosphocreatine in HILIC mode. The best separation was achieved on ZIC®-HILIC (150 x 2,1 mm; 3,5 μm; 200 Å) column using a binary mobile phase and gradient elution. Použití metody Sherlock MIS System pro identifikaci bakteriálních izolátů z lesního opadu – předběžné výsledky

This work has been related to the effect of climatic changes (especially warming) on communities of forest litter bacteria. We simulated this effect with use of litter bags with beech and/or oak litter transported to beech and spruce forest. After 2, 6 and 12 months of exposition we collected and analysed the samples. We measured quantitative and qualitative characteristics of bacteria and micromycetes community structure in all variants of forest/litter combinations (number of CFU, description of bacterial morphotypes, identification of dominant bacterial morphotypes with MIDI Sherlock Microbial Identification System). We found out that there were differencies in diversity in all variants in exposition time. From the first to the third sampling the CFU bacterial number has decreased whereas CFU micromycetes number has increased. Multivariate comparison (PCA based on relative distribution of bacterial morphotypes in samples) separated the oak litter bacterial community in spruce forest soil as the most different variant among the others.

The alternative stationary phases in HPLC analysis of drugs

Petra Holásková Charles Univerzity in Prague, Faculty of Pharmacy in Hradec Králové, Department of Pharmaceutical Chemistry and Drug Control The solubility of silica gel at high pH is the cause for developing alternative HPLC columns based on metal oxides, such as titanium oxide, aluminium oxide, and zirconium oxide. Advantages of these columns are particularly high chemical stability and separation efficiency. In our project, zirconia-based stationary phases were tested in qualitative and quantitative analysis of doxazosine and its five impurities. Polystyrene-coated zirconia was chosen because of its low retention and appropriate selectivity. The separation was done within 16 min. Subsequently, chromatographic conditions were validated.

HPLC analysis of dexrazoxane and its metabolites

High performance liquid chromatography (HPLC) is one of the most used separating methods of instrumental analysis enabling both qualitative and quantitative analysis of substances. Anthracyclines are cytotoxic drugs widely used in clinical practice for treatment of hematological malignancies (leucaemias, lymphoms) and solid tumours (stomach, breast, ovarian cancer). Treatment by these drugs has a serious limiting factor, however. This factor is an anthracycline induced cardiotoxicity, which is caused by hydroxyl radicals and other reactive oxygen forms. The main role in their formation play the complexes with iron. Dexrazoxane is a derivate of bisdioxopiperazine, which is used to reduce the cardiotoxic effect of anthracyclines. It is supposed that it takes effect by the means of its active metabolite ADR-925, which works as a substance chelating iron. This mechanism hinders the iron dependant formation of oxygen radicals. This thesis deals with analysis options of dexrazoxane and its hydrolysis products/metabolites on the stationary phase Ascentis HILIC in conditions compatible with mass spectrometric detection (MS). This thesis investigates possibilities of suppressing the boundary layer separation which is made by controling boundary layer stability. Orr-Sommerfeld equation is numerically solved for this purpose and its solution was tested on Blasius boundery layer which stability is described in many specialized publications. Transition to turbulence is found out by eN method. Single-variable description of velocity pro les on own surface, which is based on the aproximative properties of Falkner-Skan equation, was used. Process of transition on the aerofoil NACA 63-418 was solved as an example of using proposed computational programs.

HPLC analysis of drugs and drug candidates in biological material

High-performance liquid chromatography (HPLC) is currently one of the most progressive separation methods, permitting, simultaneously, both quantitative and qualitative analysis. An important area for the utilisation of HPLC is the identification of drugs and their metabolites in biological materials. This study was focused on the development of chromatographic conditions appropriate for HPLC analysis of two new potential drugs derived from aroyl-hydrazone, HNAF-INH and 4,6DAR-INH, in biological material and its subsequent application in the assessment of the stability of these substances in plasma. Analysed chelators were prepared by the modification of the structure of salicylaldehyde isonicotinoyl hydrazone (SIH) with the purpose of improving the stability of the substance in plasma. As a suitable stationary phase the HPLC column LiChroCART® HPLC - cartridge LiChrospher®100 RP - 18e (15μm) with the pre-column LiChroCART® 4-4 Purospher® RP - 18 (5μm) was chosen. The mobile phase in this case represented by a HNAF-INH compound: phosphate buffer (0,01 mol/l aqueous solution of NaH2PO4 · 2 H2O with 2 mmol/l EDTA, pH 3) : methanol, 42:58 (v/v). Coaching - the part of professional development

This work deals with subject of coaching as a part of the professional development. The first part is devoted to the theoretical initial data of the coaching. There are summarily described definitions that can be found in specialized literature. A strong attention has been given also to historical and philosophical principles of coaching. Having presented this essential information the work defines coaching compared with its related branches as consultancy, training, mentoring or psychotherapy. In a separate chapter, the process of coaching, its principles, stages and basic models have been treated. Two main types of coaching are mentioned in brief. A more extensive chapter has been dedicated to the personality characteristics of a coach and his function depending on his position in the company. Conclusion of the first part is focused on questions of ethics in the coaching process. The second half of this work is represented by a practical part describing two research studies - a qualitative and a quantitative. In the course of the qualitative research there have been obtained information and opinions on coaching in semi-structured interviews from the leading Czech coaches.
