Creating a strategic plan for the development of selected municipality

The object of this diploma work is to prepare documents for the strategic development plan of the selected municipality. Comprehensive development concept of the village is an important tool for achieving targets and effective management, because the village is then better utilized from human resources and also from tangible and intangible assets point of view. To determine priorities, wishes and demands of the inabitants was carried out a survey. The data were appraised with the use of internet application vyplňto.cz. Based on the situation analysis, socio-economic analysis and the results of the investigation were determined strategic areas, which will be crucial for the development and renewal of the village in the next 10 years. For each area was formulated SWOT analysis and with regard to the vision of the village have been designed for each strategic area the measures and specific projects that describe the problem sets and require special attention in further work in the village. A proposal of organizational development of the village, organisational structure, development indicators monitoring and updating development plan was determined. Based on the results it can be concluded that village acquired basic conceptual program document will be an important step towards the realization of sustainable development.

Analysis of selected processes and their management in RWE

This diploma thesis is focused on analysis of company internal processes and their management; analysis of a chosen process of a company RWE. The selected processes can represent a threat for the company from the point of inappropriate cyclic setting and ineffective management. This diploma thesis aims to streamline the functioning of key processes maintenance of gas facilities. The theoretical part of this thesis is focused on a problematics connected to the definition of the entrepreneurship, production process, of the company. The thesis also looks at the characteristics of process management, the corporate and production strategy, defines processes and describes analytics, which specialize in topics related to business and the business. The practical part, after specifications of the paticular company, analyzes the external environment, focused on business competitors. Considering the inner environment of the company, the dissertation thesis analyzes selected processes, leadership, procedures of cooperation with analytical method of architecture of a program ARIS by IDS Scheer Consulting company. The results are concluded in a specific proposal of management of the company RWE, which is reconfiguration process of maintenance and management.

Issues of Corn Market development

The diploma thesis deals with one of the most widely grown crop in the world that is corn. The diploma thesis consists of theoretical and practical parts. The theoretical part describes corn as an arable crop, aims at its history and trading patterns. The special attention is devoted to crucial factors which are affecting the corn market. The first subchapter of practical part focuses on analysis factors like production, harvest and planted area of corn in United States of America. Based on the graphic analysis are shown and duly commented on the long-term developmental trends of indicators in the timeframe of last fifty years. The next subchapter of the thesis deals with the analysis of time series of individual determinants affecting maize market. In the last subchapter is built econometric model, which is used to compare predictions of ex -post with reality. The conclusion of the thesis evaluates the results of modelling, describes and critically valorizes the influence of individual determinants on the maize market , and outlines the potential development of the corn market.

Legal issues of waste sorting in the Czech Republic in 2015

To analyze a current legislation of waste management is the main aim of this diploma thesis. It is focused on sorting of municipal waste. Theoretical part deals with historical development of waste management, a present Act on Waste and other special legal regulations. Attention is paid to the Act amendment which includes new obligations about sorting metal and biodegradable municipal waste from January 2015. The next part of this thesis predicts the legislation progress and waste sorting in the future. The results of two surveys are interpreted then. The first one affects public opinion and activity in the sorting of metal and bio-waste. The next one monitors the attitude of municipalities to their new legal obligations in waste treatment. The survey found that one third of citizens does not know new waste sorting methods, so they do not engage in sorting of metal neither bio-waste. There were identified the worst segments, women and young people 18-29 years old. The next detected problem is that the municipalities use a Civic amenity sites in many cases, which are in a great distance from the citizens. There shall be taken new waste-policy measures in the Czech Republic, within this will not be achieved the EU targets for the year 2020.

Marketing communication - product placement

This thesis is focused on the impact of product placement on preschool children. Furthermore, there are investigated public attitudes towards the use of product placement as widely as on television shows, as well as on a specially designated shows for children. The influence of product placement on children's audience was determined on the bases of excerpt from the film Lucky Four Serving the King, directed by Michal Žabka, using marketing methods by expert Martin Lindstrom. Moreover, there was investigated the public attitude towards this form of advertising by the online survey and also by the personal interview in premises of the nursery school. It was found, that the impact of product placement on children is relatively large and the product is towards them more memorable by this way of promotion. The results also showed, that the public thinks that promotion of the product is neutral. However, this does not affect placement in children's programs. According to the result of the public, it is believed, that advertising has nothing to do there. More detailed results are analyzed in own work.

Molecular detection RYR1 gene mutation causing malignant hyperthermia in dogs

The theme of this work are methods for molecular detection of mutations in the dog genome causing malignant hyperthermia. The work is divided into two main parts. The first part is a literary research summarizing knowledge about malignant hyperthermia from the human and veterinary anesthesiology molecular biology and genetics. The second part of the work describes the methodology and conclusions drawn from the molecular detection of above mentioned mutation which was conducted in the laboratory of the Department of genetics. The literary research deal with malignant hyperthermia syndrome. The aim is to familiarize with symptomatic manifestations triggers the mechanisms the development of acute malignant hyperthermia and its clinical diagnosis. The first cases of observation of malignant hyperthermia syndrome are also described as well as the history of the research which led to a detailed description of this disease in humans dogs and other species bearing malignant hyperthermia causing mutation. The next part of literary research is a summary of knowledge about the mechanism of mutation the detection of sequences containing this mutation and influence on the arrangement of proteins in the ryanodine receptors. For the last part of literary research commonly used laboratory techniques of non invasive mutation detection were chosen. These techniques are mostly used by accredit genetics laboratories. Listing of Czech laboratories performing diagnostic of gene RYR1 mutation is also contained. Experimental part of barchelors thesis is focussed on molecular marker optimalization for casual mutation in RYR1 gene detection by technique of restriction fragment length polymorphism (PCR RFLP). This technique was apply to 338 deputies of different dog breeds. Accomplished analysis did not confirm presence of the mutated allele in assessed breeds population. Result of PCR RFLP technique was confirmed by sequence analysis. Results were discussed with literary sources which did not judge real breeds but are developing a model special bred line with presumed mutated allele segregation. The results indicated that the mutation of gene RYR1 do not represent an influential disorder in breeds harbored in Czech Republic. Sporadic presence of this mutation was consulted with commercial genetic laboratories.

Reflects chemical communication of Eurasian beavers density-dependence?

Reflects chemical communication of the Eurasian beavers density-dependence? Almost all studies about population density describe certain dependency: with higher population density increased intensity of chemical communication. This is provided by many authors on different animal species. However this study showed that chemical communication does not reflects density-dependence of the Eurasian beaver. Explained variability of final model was r2 = 0.02586101, but with no significance for both variables. However some relationships were tested. Correlation between biomass (amount of consumed woody resources) and number of shelters (beaver nests) was not significant, but correlation between number of shelters and distance was positively significant. This lead to test the alternative special model (mc) where dependency of number of lodges and its changing distance was tested. This model was significant (p = 0.007182) and was discovered new relationship in population parameters of beaver populations: with increasing distance increased number of shelters. Main focus of thesis was to study how the distribution and intensity of the chemical communication is related to parameters of population density. The result was: with increasing distance the intensity of chemical communication doesnt increase.

Toxoplasma gondii manipulation in the host

The aim of this bachelor thesis is to collect so far discovered findings about manipulation of behaviour caused by Toxoplasma gondii. The first part of this thesis introduces basic characteristic of the parasite and toxoplasmosis, which is a disease caused exactly by this parasite. In the second part we summarize outcomes of many different scientific essays that deal with manipulation of intermediate host behaviour. Toxoplasma gondii is a widespread parasite that can modify the behaviour of the intermediate host, which can lead to serious changes in psychological profile of infected people. Toxoplasmosis is one of the most common zoonosis in the Czech Republic, the prevalence reaches 25-30 %. Toxoplasmosis is usually an asymptomatic infection, however it modifies the behaviour and acting of the intermediate host in some special situasions, it also changes excretion of important substances and affects the secondary sex ratio too. Toxoplasmosis is a very important risk especially for imunodeficit people and pregnant women. The principle of prevention is very strict personal and food hygiene.

Biotransfer of selected risk metals into plants and their accumulation and distribution in plant organs

Contamination of soils with heavy metals is one of the serious environmental problems threatening human being. Heavy metals are considered as the special hazard of soil pollutants because of the adverse effects on the plant growth, the amount, activity of useful microorganisms in soils and the quality of food. Regard to the persistent and toxicity, the heavy metals are toxic when we consider different kinds of pollutants in soils. In the soil, zinc (Zn), cadmium (Cd), lead (Pb) and mercury (Hg) toxicities frequently occur than the other metals because of their precipitation and sorption by the soil. It is a very dangerous situation because when these metals are taken up by plants, they can be transported to the food web and food chains. In the present study, the accumulation of four heavy metals (mercury, zinc, lead and cadmium) in the whole grain of spring accessions of emmer, einkorn and common spring wheat cultivars and potato (Solanum tuberosum) is reported. Heavy and essential elements were monitored in potato cultivars in the exact field experiments and in hydroponically grown plants. The elements were determined by methods FAAS, ET AAS, and AMA (Advance Mercury Analysis). Statistical analyses were performed using SPSS 9.0 with the Tukey HSD (Honestly Significant Difference) test (alpha equal to 0.05). In our study, the concentration of heavy metals decreased in the order zinc (Zn) > lead (Pb) > cadmium (Cd) > mercury (Hg) in the wheat grain. The comparison between three varieties of investigated wheat revealed that the emmer wheat was rich in zinc content (62.12 mg kg-1 dry matter), while the spring wheat had the lowest average concentration of zinc in the grain (40.99 mg kg-1 dry matter). Generally, the values of lead concentration in grain wheat varieties were low (ranging from 0.1268 mg kg-1 dry matter to 0.2950 mg kg-1 dry matter). The concentrations of mercury in four typical growth stages of wheat (boot stage 10, heading stage 10.2 1/4 of head emerged, leaf-stage 10.2 and stage ripening 11 according to Feekes) were also determined. It has been shown that the concentrations of mercury in different wheat varieties were absorbed differently at different growth stages of plant. Stage 10.2 and leaf stage 10.2 showed the high mercury content (0.0152 mg kg-1 dry matter and 0.0214 mg kg-1 dry matter, respectively). Among individual varieties significant differences were determined. Amounts of toxic and potentially toxic elements detected in investigated potato tubers are characterized by a large variability within investigated groups. Performing statistical analysis (one way ANOVA) showed that there were no significant differences between two investigated groups of samples (samples from Uhříněves and Valečov in the year 2013 and 2014) considering either one of investigated metals. Measurable levels of mercury were found in smallest amounts in all investigated potato samples comparing to other metals (Cd, Pb). Plant cells compared to animal cells are characterized by the formation of cell walls. Plasma membrane or cell membrane is a biological active membrane separating the interior of cell from the outside environment. An adjusted method for isolation of protoplasts was developed and adapted for isolation of protoplasts from plant material (potatoes). In our experiment, the plants were grown hydroponically in the Research Institute of Plant Crops Prague-Ruzyně. If we examine the plant membrane, one option is to remove the cell wall by means of special mixture enzymes. Protoplasts were released in the dark at 25 degrees of Celsius for 18 hours. The 70 and 90 microns sieve was used to filter and then centrifugation for 5 minutes at 100 x g. All the steps were carefully carried out to prevent the damage or breakage of protoplasts.

Kinematics and elastokinematics of independent multi-link suspension of axle using flexible supporting elements

This thesis deals with elasto-kinematic properties of multi-link rear suspension system, based on simulations. The creation of computational model and simulation of elastokinematics are based on specialized modules of the HyperWorks software. The thesis idea is to create new models and improve their properties. The chapter introduction presents and discusses the current knowledge state in the issue of elasto-kinematic characteristics of the vehicle suspension. Previously published works are presented in this section. The following section defines basic terms, features and design elements concerning the topic vehicle suspension and vehicle axle, which can help readers to orientate in this field. The first part of the thesis is focused on the collection of high-quality input data to create the MBS computational model based on the real suspension components. The determination of kinematic suspension points, experimental measurements of inertia moments of supporting elements of the suspension and measurements of deformation characteristics of rubber-metal bushings are presented in the thesis. There are also measuring characteristics of force elements such as springs or shock absorbers. Then the proposed method of measuring elasto-kinematic characteristics of the suspension using testing machine for model validation is also introduced. The second and main part is devoted to creating a new MBS simulation models of multi-link rear suspension using HyperWorks system. The suspension model is improved by successive steps, from kinematic model, through model with flexibility of the bushings, to the complex model in which the flexible properties of all supporting elements are reflected. The properties of the used models are described together with arising calculation problems. The results discuss the impact of elements flexibility and individual structural alternatives on elasto-kinematic characteristics of the suspension system.
