Influence of working conditions on the detected signal by BSE detector in the scanning electron microscope Vega 3 XMU

This work is focused on investigating of influence of different working conditions in scanning electron microscope to signal detected by backscattered electron detector. In the theoretical part, there is a general description of scanning electron microscope, backscattered electrons issue and also definition of methods of signal to noise ratio evaluation. The practical part is focused on observation of suitable samples in a scanning electrone microscope.

Domain conformations of the motor subunit of EcoR124I involved in ATPase activity and dsDNA translocation

Bacterial type I restriction-modification systems are composed of three different subunits: one HsdS subunit is required for identification of target sequence and anchoring the enzyme complex on DNA; two HsdM subunits in the methyl-transferase complex serve for host genome modification accomplishing a protective function against self-degradation; two HsdR (or motor) subunits house ATP-dependent translocation and consequent cleavage of double stranded DNA activities. The crystal structure of the 120 kDa HsdR subunit of the Type I restriction-modification system EcoR124I in complex with ATP was recently reported. HsdR is organized into four approximately globular structural domains in a nearly square-planar arrangement: the N-terminal endonuclease domain, the RecA-like helicase domains 1 and 2 and the C-terminal helical domain. The near-planar arrangement of globular domains creates prominent grooves between each domain pair. The two helicase-like domains form a canonical helicase cleft in which double-stranded B-form DNA can be accommodated without steric clash. The helical domain, probably involved in complex assembly, exhibits only a few specific interactions with helicase 2 domain. Molecular mechanism of dsDNA translocation, cleavage and ATP hydrolysis has not been yet structurally investigated. Here we propose a translocation cycle of the restriction-modification system EcoR124I based on analysis of available crystal structures of superfamily 2 helicases, strutural modeling and complementary biochemical characterization of mutations introduced in sites potentially inportant for translocation in the HsdR motor subunit. Also a role of the extended region of the helicase motif III in ATPase activity of EcoR124I was probed.

Effect of substrate hydrophobicity on the electrocatalytic behaviour of phthalocyanine

In this work, the electrocatalytic activity of pyridinoporphyrazine mediator deposited on two substrates with different hydrophobicity has been presented. We focused on hydrophobic highly oriented pyrolytic graphite (HOPG) as well as hydrophilic annealed gold electrode (Au111). The immobilization of mediator has been carried out spontaneous adsorption from aqueous solution on electrode surfaces. The different hydrophobicity surface resulted in change the electrocatalytic activities of mediator to hydrogen in aqueous solution. For this purpose, electrochemical, in situ spectroelectrochemical, and ex situ microscopically characterizations have been performed. The detailed comparison of surface morphology, surface coverage, surface roughness parameters, thickness of deposited layers of mediator, and pH solution has been studied.

Experiments of Bubble and Drop Breakup in a Turbulent Flow.

Bubble and drop breakage in a turbulent flow is studied experimentally. In an experimental cell, the turbulent flow (of distilled water) is generated by an array of coaxial nozzles, which allows adjusting independently the turbulence intensity and the mean velocity. Either air or cyclohexane is injected in order to produce bubbles or drops. Their breakage by the turbulent flow is observed using a high-speed camera. The image processing provides information about particles in each image. Tracking software is developed and it allows to identify and characterize the breakup events (their location, number and sizes of daughter particles and also trajectories prior the breakup) and to distinguish them from a simple overlap of multiple bubbles. The parameters characterizing the breakup are then studied in dependence on local flow conditions.

Financial analysis of chosen enterprise

The objective of this Bachelor thesis, which is entitled Financial analysis of chosen enterprise, is finding the financial situation of the company KOVO KASEJOVICE MONT s. r. o. from 2011 to 2014. The emphasis is placed on the analysis of the financial health because of basic methods of financial analysis. This thesis consists of two parts, the theoretical and practical part. The theoretical part is focused on describing the company as a whole, financial analysis and accounting statements. This part presents also the methods and indicators of financial analysis. In the practical part, there is the chosen enterprise KOVO MONT KASEJOVICE s. r. o. This part is devoted to the analysis of absolute indicators, methods ratios of profitability, activity, insolvency and liquidity, as well as creditworthy and bankruptcy models based on corporate accounting data. In the end of my thesis are summarizes the results of the financial analysis and evaluation of the overall situation.

The company in a competitive environment

The Master´s thesis is dealing with characteristic of a firm in competitive environment. The theoretical part describes various forms of markets, both perfect and imperfect competition. Furthermore, in this part will be characterized the concept of marketing mix, internal and external environment of a company, and also selected analyses from both these environments. The main part of the thesis is focused on three media agencies, which will be described in detail. Also, locations where these agencies operate will be described. Locations determine positions of these firms in the market and their respective competitive powers. In the analysis section, I will apply the Porter Five Forces model and the SWOT analysis on these selected firms. Subsequently, I will be dealing with advertisement since it is a crucial part of the marketing mix, very important for comparison of the agencies. The practical part will present a research and a questionnaire. The research is focused on the influence of advertisement on public. The questionnaire is dealing with cultural events. The thesis compares total revenues, analyses results of the investigation, and proposes ways that the selected agencies could achieve better position in the market.

Foreigns nationals interned in the Czech Republic

Diploma thesis describes foreign nationals imprisoned in the Czech Republic with more detailed focus on the characteristics of the selected groups. Based on the analysis, characterization and expert examination gives a comprehensive view on this issue and to help you navigate not only the interested persons. The theoretical part focuses on the description of the Prison service of the Czech Republic as an institution, its role, structure and organization system. It also deals with the history and its evolution to the present time. It provides a comprehensive insight into the issue of foreigners in the Czech Republic, legislative regulation and crime. For purposes of this research work have been selected group of foreign nationals imprisoned in the Czech Republic, which are closer to the theoretical part, characterized, distributed and investigated. Analytical part is focused on analysis of selected data, and layout (for clarity) into several sections, which always include one area of research interest of the issue. This section is arranged graphs providing an overview on the development of individual indicators. In conclusion there is summary assessment and recommendations to individual areas of research.

Flow Parameters Simulation Technique in Annular Turbine Cascade INLET

This article summarizes the concept of equipment which produces the required flow parameters at the inlet of annular turbine cascade. The investigation of inlet flow parameters - the boundary layer on hub and tip inlet channel end-walls and the inlet flow turbulence are part of a project intent on a flow in multistage steam turbine arrangement. This variable inlet can produce the laminar or turbulent boundary layer, respectively the laminar or fully turbulent velocity profile in the inlet channel. The change of inlet turbulence level is controlled by the grid. The detailed measurement of inlet flow: the turbulence and the total pressure distribution across the height of inlet channel were performed by HWA (hot wire anemometry) probe and by total pressure probe, respectively. The measurement results characterize the inlet flow for several inlet channel arrangements and are used for the experimental investigation of the flow field inside the stage and as the inlet parameters for CFD simulations.

Evaluation of Some Social Conditions - October 2016

In October 2016 Public Opinion Research Centre's survey investigated views of Czech citizens on some social conditions. From all specific social conditions included in the survey people evaluate mostly positively the accessibility of education (86% positively, 10% critically), the health care (74% : 24%), and a chance to get a job (52% : 45%). All other monitored areas of social conditions - i.e. a chance to get a flat (36% positively, 57% critically), financial situation to start a family with children (29% : 65%), social security for the elderly (18% : 76%), and life conditions for the handicapped (30% : 56%) - are evaluated mostly critically.

Migration of doctors and health workers in selected countries of Europe since 1990

The main objective of this thesis is the comparison of the position of the healthcare personnel on the labor market and migration in selected countries - Czech Republic, Germany, France and the United Kingdom. In the theoretical introduction it describes concepts such as brain drain, remittances, emigration and others subjects associated with the migration of the workforce. It analyzes the differences in labor market of healthcare personnel in selected countries and the activities of doctors for change of working conditions. The practical part presents and analyzes data from publicly available statistical sources (OECD, Eurostat, IHIS CR etc.) regarding the number of healthcare personnel, the number of migrating doctors and nurses and their destination countries, salary and wage conditions of health professionals. Expenses on health and quality of the health care are compared between selected countries. Finally, recommendations for Czech Republic, that could help prevent excessive drain of medical personnel abroad, are given - an increase in wages and health expenditures, increasing the number of medical personnel and reduction of administrative burdens.
