Chosen problems of genetic diversity of dog
CHLUPATÁ, Lucie
The bachelor thesis is worked out in the form of the research on the theme "Selected questions from the genetic diversity of dogs". The aim of this work was processing of the current situation of scientific knowledge in the field of genetic diversity of dogs. Firstly, the origin is described in the thesis, domestication of dogs, an organisation dealing with dogs breeding and selected breeds of dogs. From the genetic site point of view, the basic terms in genetics are described genetic markers, DNA profile of dog and the methods of evaluation of genetic diversity. In the end of my bachelor thesis I outlined possible recommendations for dog breeders.
Genetic diversity of cattle in Czech Republic
ČERVINKA, Václav
The thesis deals with genetic diversity in the Czech Republic. Firstly domestication of livestock in general and then domestication of cattle is decribed. History of cattle in the Czech Republic, the importance of cattle breeding and last but not least the condition of cattle in the Czech Republic is also introduced. The thesis is also focused on biodiversity, threats to biodiversity and its conservation and the levels of diversity are also specified. Genetic markers, microsatellites, minisatellites, SNP, polymorphic structural genes and also mutations are described. The cattle breeds farmed in the Czech Republic are specified next. The National Programme, genetic resources and genetic resources in the Czech Republic are evaluated. Finally genetic resources of " česká červinka" and "český strakatý skot" are described.
Educational programme about horses for horse rider
JAVORSKÁ, Jana
Javorská, J. 2014: Educational programme about horses for horse riders. Bachelor thesis. Faculty of Education, JU, České Budějovice, 62 p. The educational programme "How to understand horses and how to make horses understand you" is an opportunity to apply essential knowledge of ethology and evolution of the horses in the riding club environment. This bachelor thesis concerns formation an educational programme to enable riders to understand the basic aspects of the evolution and ethology of horses. Its theoretical part works out an overview from the fields of evolution, domestication and ethology of the horses and the problems which are responsible for the prevailing current method of hobby and sport horse breeding. Its practical part contains a description of a proposed all-day education programme, which includes the above mentioned topics placing emphasis on acquired knowledge into practice at the riding club. The programme is in the form of an education project in which the participants become scientists for a whole day at the conference of ethology of horse. A set of worksheets has been created for the programme. The outcome of this work is an implemented educational programme. Its discourse part contains proposals for further improvement of the programme, which followed the pilot project.
Components of wolf behaviour (Canis lupus) relate to the native behaviour of German Shepherd dog (Canis lupus f. familiaris).
ZUNKOVÁ, Karolína
The aim of my study was to compare similarities and differences in behavior among the general wolf (Canis lupus) and German Shepherd dog. My thesis is recherche. Behavior data and a comparison I gained from the literature and created questionnaires designed for breeders of German shepherds dogs. The available sources indicated that the differences between the general behavior of the wolf and German shepherd dog are given by the German Shepherd Dog domestication , environment and human impact and body shapes. ?
The polymorphism of sericin 2 gene in silkworm (Bombyx mori)
TYLLEROVÁ, Helena
In our study, we examined the polymorphism of gene Ser2 from domesticated silkmoth Bombyx mori and its closest wild relative B. mandarina. As a starting material for our work, we used the restriction map of allele C isolated from hybrid lineages 200 and 300 of European silkmoth B. mori (Michaille et al. 1990a). We also used the published sequence of allele D which was isolated from {\clq}qDaizo`` p50 strain of B. mori (Kludkiewicz et al 2009). Based on the published sequence, we designed PCR primers and performed sequence analysis of 4 different alleles of gene Ser2, including the almost complete genomic sequence of allele C (except for two short regions containing repetitive DNA). We then compared the sequences and found that the alleles differ significantly not only in intron lengths and the sizes of exon 9, but also in exon arrangements. While the allele C contains 12 exons (1, 2, 3, 4, 5, 6, 7, 8a, 9, 10, 11, 8b), the alelle D and other alleles analysed have 13 exons (1, 2, 3, 4, 5, 6, 7, 8, 9a, 10a, 9b, 10b, 11). Allele C showed very unique arrangement of exons, which suggested that it may represent an ancestral form of Ser2 gene. We analysed the arrangment of exons located at the 3´end of the Ser2 gene by PCR and electrophoresis in 70 available strains of B. mori and B. mandarina. We also tried to find a similar allele to the allele C and we amplified and sequenced 2 kb region from 55 Bombyx strains. Phyllogenetic analysis of gene Ser2 gene suggested that the C allele of now probably extinct European silkmoth hybrids 200 and 300 does not seem to be an ancestral form of Ser2 gene, but it rather seems to be the result of complex secondary rearrangements.
Spawning of domesticated eurasian perches in controlled conditions of fish breed
TRNKA, Petr
The eurasian perch (Perca fluviatilis Linnaeus, 1758) is considered as very perspective species for breeding at intensive aquaculture, where was successful domesticated. Consumption of eurasian perch is increased at present. The greatest grow of consumption is mainly in countries of alpine region and in Scandinavian countries. The aim of experiment was monitor and character spawning of domesticated eurasian perch in controlled conditions of breed. It was determinated fertility and then fertilization of eggs. The next aim was make incubation of fertilization eggs and determinate hatching. After hatch was determinated total length of larvae and its quality by the help of osmotic shock. Domesticated eurasian perch is spawned without problems in controlled conditions of breed. The resulting data of domesticated perch are worse than date of non-domesticated perch in some spawning characteristic. This problem will resolve with gaining knowledge about spawning of eurasian perch. The greatest problem is hatching of eggs, which is very low during spawning of domesticated perch.
