Symbolic Production of Papal Institution
Gerbery, Juraj ; Češka, Jakub (advisor) ; Marcelli, Miroslav (referee)
The topic of the submitted thesis is a symbolic performance of the papal institution. We are laying stress on the form of communication of the papal institution and design of its legitimacy within the medial discourse. In the first and second chapter, we summarize the definition of papal institution as such. First of all, we investigate papal institution from the point of view of its historical development, eventually of its rich structure. Third part is defining the basal papal symbolism by means of discussing the principles of clothing and papal insignia. The key parts are composed by analyzing of both, the Pope election according to the Constitution Universi Dominici Gregis and the analysis of the Doctrine of impeccability of the Pope. The first of the phenomenon, being analyzed using the theory of modern myths by Roland Barthes and which is being derived from traditional conception of investigation of symbols performed by Ferdinand de Saussure. The second semiotics analysis is focused on the doctrine of impeccability of the Pope stressing the Theory of performatives of John Langshaw Austin with remarkable contribution of Pierre Bourdieu and his conception of the social conditions for the effectiveness of ritual discourse. Our hypothesis is, that investigation of instruments of symbolic performance of...

Electrochemical determination of 2-nitrofluorene and investigation of its interaction with DNA
Skalová, Štěpánka ; Stávková, K. ; Barek, J. ; Vyskočil, V.
2-Nitrofluorene (2-NF) belongs to the group of nitrated polycyclic aromatic hydrocarbons. These compounds are categorized as environmental pollutants and they can manifest mutagenic and carcinogenic effects. A differential pulse voltammetric method at a glassy carbon electrode (GCE) was developed for sensitive determination of 2-NF. Under the optimum conditions found, a linear calibration dependence was obtained in the concentration range of 2 x 10(-7) - 1 x 10(-5) mol L-1, with the limit of quantification of 2 x 10 mol L-1. The practical applicability of this method was verified on the direct determination of 2-NF in a model sample of sand. Moreover, the mutual interaction between 2-NF and DNA was investigated using an electrochemical DNA biosensor (DNA-modified GCE). Cyclic voltammetry, electrochemical impedance spectroscopy, and square-wave voltammetry were employed in this study, confirming the formation of a 2-NF-DNA complex.\n

Analytical evaluation of drugs and novel drug candidates using HPLC
Kovaříková, Petra ; Klimeš, Jiří (advisor) ; Nobilis, Milan (referee) ; Blešová, Marie (referee)
High performance liquid chromatography (HPLC) is one of the most frequently used analytical techniques for the analysis of drugs. HPLC methods are widely employed in all fields of the modern pharmaceutical analysis - new drugs development, quality control and assurance, the analysis of drugs and metabolites in a biological material (e.g. therapeutic drug monitoring, bioequivalence). The theoretical part of this thesis deals with the main specifics and aspects associated with HPLC analysis of drugs. The experimental part is consisted of six original research papers with appropriate comments divided in to two sections (The analysis of the drug candidates from the group of aroylhydrazone iron chelators and The stability study on selected drugs). The first section concerns with development, validation and application of new HPLC methods in the analysis of drug candidates from the group of iron chelators - pyridoxal isonicotinoyl hydrazone (PIH), salicylaldehyde isonicotinoyl hydrazone (SIH) and pyridoxal 2-chlorobenzoyl hydrazone (o-108). Aroylhydrazone iron chelators are under the investigation as promising drug candidates. Despite these chelators have demonstrated number of interesting pharmacological effects, in fact there were no modern analytical techniques suitable for the analysis of these drug...

Stress in working environment

Highway multispan bridge over wide wally
Bobek, Lukáš ; Daněk, Ing Stanislav (referee) ; Klusáček, Ladislav (advisor)
The aim of this diploma thesis is to design and assess highway bridge. The structure is located on the D1 motorway section bridging a wide valley between the Slovak villages Doľany a Klčov. Three variants have been created – box girder bridge with transverse overhangs, a pair of girder bridge and a pair of box girder bridges. The various proposals were compared with each other. For the most valuable option is selected prestressed box girder bridge with transverse overhangs, which are supported by prefabricated concrete struts. The selected proposal was subsequently elaborated in detail, the load-bearing structure is analyzed using Scia Engineer 16. In calculating the internal forces is adjusted for the effects of construction methods, even as time-dependent analysis TDA. When designing the load-bearing structure it is considered the action of permanent load, also loading from transport and temperature. The structure is assessed for serviceability and ultimate limit states according to current standards. The bridge deck is built by incremental launching method. The principle of this method consists of building the segments in a casting yard located behind the bridge abutment. Each segment is matchcast against the previous one and prestressed to the section of structure already built. The whole superstructure is then jacked forward a distance equal to the length of this segment. This process is repeated until the bridge deck is in its final position. Chosen method of construction is very fast and efficient, to the country in the valley isn´t damage during construction.
