Cellular senescence and ageing-associated diseases

In the last years more evidence supports the role of cellular senescence in ageing-associated diseases. Cellular senescence plays a role in these diseases on multiple levels and often directly supports their pathogenesis. The goal of this bachelor thesis is to summarize latest knowledge regarding the mechanism and the efect cellular senescence has in ageing-associated diseases, namely cancer, atherosclerosis, osteoarthritis, neurodegenerative diseases such as Alzheimer's disease, Parkinson's disease and multiple sclerosis, obesity and diabetes mellitus type two.
Biological effects of edible algae.

Nutritional factors with antioxidant properties, such as those contained in edible algae or green plants, might be implicated in protection against cancer development. Chlorophyll and other tetrapyrrolic compounds, structurally related to heme and antioxidant bile pigment bilirubin, belong to important candidate molecules, which might be responsible for these effects. Based on our studies demonstrating antiproliferative effects of S. platensis edible alga extract on experimental model of human pancreatic adenocarcinoma we investigated in detail the effect of chlorophyll occurring abundantly in this alga. Since only scarce data exist on the antiproliferative effects of chlorophylls, the aim of our study was to assess these effects. The study was performed on experimental models of human pancreatic and prostate cancer. The inhibitory effects of chlorophylls (chlorophyll a, chlorophyll b, chlorophyllin and pheophytin a) on cell proliferation and cell viability were investigated in in vitro studies. Chlorophylls reduced the mRNA expression as well as activity of hemeoxygenase in tested pancreatic cancer cells. Simultaneously, chlorophylls played an important role in redox environment of studied cancer cell lines including modulation of mitochondrial membrane potential, reactive oxygen species (ROS)...

The aim of this thesis was to summarize the role of innate immunity in antitumor immunity and to define mechanisms by which cells of innate immunity (especially neutrophils, macrophages, NK cells and dendritic cells) recognize and eliminate tumor cells. Finally, I suggested methods of tumor immunotherapy using cells of innate immunity.

Adenosin a nádorová imunoterapie

The aim of this bachelor thesis was to study the immunosuppressive effects of adenosine and the use this knowledges in cancer immunotherapy. The impact of enzymatic removal of adenosine on efficacy of cancer immunotherapy was examined using murine pancreatic model Panc02.

The aim of this thesis was to study the possibilities for targeting in tumor immunotherapy. We studied the expression of phosphatidylserine in melanoma B16-F10 in vivo and in cell culture B16-F10 in vitro. We analyzed conditions resulting in higher expression of phosphatidylserine on tumor cells surface.

The aim of this diploma thesis was to assess the current knowledge of the pupils of 8th and 9th grades of primary schools about selected chronical non-infectious diseases (known as civilisation diseases). The pupils of rural and urban types of primary schools have taken part in the questionnaire survey. The theoretical part is focused on the characteristics of selected diseases, their causes and prevention. In the practical part, the results of the survey are processed and evaluated.
The role of galectins in cancer cell invasiveness

Galectins are family of β-galactosidase binding proteins that serve many functions in all kind of mammalian cells. In the past years galectins, namely galectin-1 and galectin-3, have been revealed to play a major role in various cancer processes including cancer cell invasiveness, a process indispensable for the formation of metastasis. Both extracellular and intracellular forms of galectins modify the process of invasiveness in various ways, through interacting with different components of the cell or of the cell signalling pathways. The aim of this bachelor's thesis is to summarize mechanisms by which galectins promote cancer cell invasiveness. Keywords: galectins, galectin-1, galectin-3, invasiveness, cancer, metastasis
