Methods for Analyzing Change From Baseline to Final Assessment

In this thesis, we analyze treatment effect estimate in randomized clinical studies. Treatment effect estimates are constructed on the basis of three models. The first part of this thesis is about the behaviour of these estimates when the treatment effects vary with patients. We find out that all types of estimates are consistent and we derived their asymptotic distribution. The estimates are compared by their asymptotic variances. The theoretical conclusions are confirmed by a simulation study. The second part describes the case where measurements of baseline and final values contain an error. Two estimates are analyzed. We find out that both estimates are consistent. We derive their asymptotic distribution and compare their variances.

Targeted differenciation of limbal and mesenchymal stem cells and their therapeutic application

The research of stem cells slowly transfers from the experimental to the preclinical and clinical level. They are in the centre of interest thanks to their potential to treat many of severe injuries and genetically determined diseases. However, the clinical application of these cells has to be based on a basic research of their characteristics and differential potential. Adult stem cells are in organism in minor populations in unique niches. In comparison with embryonic and induced pluripotent stem cells, the adult stem cells have lower differential potential but they also tend less to making teratomas. The therapeutic use of differential and transdifferential potential of limbal and mesenchymal stem cells is described here in more detail with focus on their use in damaged ocular surface treatment. Limbal stem cells are the only source of stem cells for corneal epithelium regeneration in most organisms. Deficiency of these stem cells leads to severe eye disorders even to blindness. Nowadays, a transplantation of allogeneic limbal stem cells or allogeneic limbus is the only chance for patients with total limbal stem cell deficiency. The subject of this diploma thesis is engineering design of the sediment conveyer. Conveyer is instrumental to total emptying of sediment silo that is situated in a sewage treatment plant. Proposed construction has to fulfill the specified functional requirements and parameters. Parts of solution are also necessary analytical and FEM calculations.

The influence of one-week hospitalization on patients with surgical treatment of biliary paths diseases.

The aim of this thesis is to evaluate how are patients affected by process of hospitalization since they are hospitalized to their discharge. Teoretical part of the study describes anatomy and pathology of biliary paths which is elementary for making context of biliary paths diseases. Further, the process of hospitalization and dietary restrictions that supports the treatment are properly described. Last but not least, psychological aspects of hospitalization are studied. Empirical part of the study is focused on examining of how single parts of hospitalization affect patient's psyche. Forms were used as a researching method. They were filled in by patients at hospital in South Bohemian region.

Immunogenic cell death and its application in imunotherapy protocols

Treatment of tumors by protocols based on combination of surgery, radiotherapy and systemic chemotherapy resulted in the improved prognosis of many human cancers. Despite the continuous introduction of new drugs and further improvements of chemotherapy protocols, it's likely that at some point chemotherapy will reach its limits and clinical efficacy will plateau. Immunotherapy has emerged as another treatment modality with the potential to contribute to further improvements in the survival. The products of advanced cellular terapies must be generated using GMP-approved reagents and number of studies have addressed the need to generate large numbers of DCs for clinical trials according to regulatory authorities and to current legislation. The rising knowledge about dendritic cells (DCs) in the immune response against tumors and the ability to prepaire DCs in vitro leaded to the establishment of immunotherapy protocols. Breakthrough studies that identified markers of immunogenic tumor cell death after chemotherapy treatment challenge the long-time perception of chemotherapy and immunotherapy as opposing and incompatible treatment modalities. Determination of Valproic Acid using Capillary Gas Chromatography with Mass Spectrometry Detection

Valproic acid is a chemical compound with simple structure and wide therapeutic effect. It is primarily used in treatment of epilepsy, bipolar disorder and rarely in treatment of depress. The serum levels of valproic acid have to be determined due to its narrow therapeutic window and risk of drug-drug interactions. Thus a new sensitive analytical method using capillary gas chromatography with mass spectrometric detection has been developed for determination of valproic acid levels in human serum (caprylic acid used as an internal standard). The results showed that the new method was linear within the range of 0,96 - 967,38 µmol/l, which include the therapeutic range (367 - 693 µmol/l) of valproic acid. This method was compared with formerly established fluorescence polarization immunoassay (FPIA) method for determination of valproic acid levels in human serum (FPIA) by means of the Passing-Bablok regression analysis and Bland & Altman Plot. The statistical analysis confirmed that new method is suitable for determination of levels of valproic acid in real human serum samples. The validated method has been applied to the routine analysis of patient's serum samples.

Právní požadavky při nakládání s biodegradabilním odpadem

The first section identifies the types and origins of biodegradable waste. There is biodegradable waste of both animal and vegetable origin. It also analyzes legislation relating to the treatment of biodegradable waste at the national level and the EU. In the European Union and the Czech Republic there are many laws regarding this topic. These laws are all interconnected and they set requirements to be respected not only with regards to the waste, but also incorporating the operators of the facilities for the disposal or use of these types of waste. Furthermore, present legal requirements for technical and the technological level of processing biodegradable waste, they analyze the competence of administrative authorities in the management of biodegradable waste from all sources. In the last section, present and assess the priority targets for the management of biodegradable waste.

The assesment of trombocyte function by optical and impendance agregometry

Introduction: Platelets have an important role in the process of haemostasis in which they form blood clot generation. In some cases it is necessary to suppress activity of platelets by antiplatelet drugs to prevent life threatening states. Acetylsalicylic acid (ASA) and clopidogrel can influence the mechanism of primary haemostasis by inhibition of platelet function. This process has to be controlled. Clopidogrel is used for prevention of thrombotic states. The active form of clopidogrel is formed by cytochrome P450 3A4. Clopidogrel binds to the receptor P2Y12 instead of ADP, so platelets cannot be activated. The effect of clopidogrel is very variable. It is known that there is unresponsiveness to clopidogrel in 5-30 % patients. Acetylsalicylic acid acetylates cyclooxygenase and blocks the conversion of arachidonic acid to tromboxan A2, so platelets cannot be activated. ASA treatment is effective, but in some patients can appear so called aspirin resistance (AR). Possible unresponsiveness to the therapy is the reason why it is suitable to monitor antiplatelet therapy, which may be provided by various methods. Transsexuality and HSR-problem.

This work wishes to present the problem of trans-sexuality and its treatment, especially its most controversial part, i.e. hormonal and surgical reassignment. It brings the information about disease as such, shows treatment possibilities and points out also certain ethical problems which the patient faces, especially when he is a believer. Second part of this work studies the life quality of trans-persons and brings author?s research within a small sample group of transsexual believers. The last part of this work presents summary of results.

Quality of life children with installation ventricle-peritoneal shunt

The goal of bachelor's thesis is to analyze quality of life children wiht hydrocephalus with installation ventricle-peritoneal shunt. In the thesis there are given detais on problems, which arise from leave hospital and treatment at home. Thesis inqure problem 's influence on quality of life illness children and their families. A quantitative oservational study is made of questionnaire that include 9 parts about future children 's life. Physical effort was researched on diseased children from neurosurgery clinic.
