Attempts to demonstrate and interpret differences between subunits forming biologically active oligomers of low-(calf spleen) and high-molecular mass (E. coli) purine nucleoside phosphorylases (PNP) yielded new crystal structures of both enzymes complexed with ground-state analogue ligands in space groups and with two full trimers and one full hexamer in the asymmetric unit, respectively.

LC-MS analysis based on probabilistic approach

Liquid chromatography (LC) in tandem with mass spectrometry (MS) is a measurement tool for obtain information about the compounds in the investigated extracts. There were already developed methods for processing and analysis of measured data sets. However, only partial problems of processing/analysis task were handled independently. Therefore, the rst part describes existing methods and techniques commonly used in the LC-MS for the processing and analysis today. In this thesis an approach based on the theory of systems is used for description of abstract model above the measured data. This model encapsulated all processing/analysis steps into appropriate and consistent mathematical space. The creation of this model via description of the measurement device and data outputs is introduced. Abstract model of LC-MS data set is used to decompose the measurement into three partial contributions, the analyte signal, the random noise and the systemic noise. The separation process of the signal could be estimated using the probabilistic approach. That probabilistic approach to the LC-MS analysis was implemented in the developed software, which was published in the Bioinformatics Journal.

Two Educational Conceptions for Children And Youth. Scout And Pioneer Organizations at Present.

The work concerns with the description and the comparison of the two biggest civil associations in the Czech Republic which offer a meaningful free time use of children and youth. These associations are {\clq}qJunák`` and {\clq}qPionýr``. The comparison is accomplished by function, principles and methods. An inseparable component of the work is a short view into the history of {\clq}qJunák`` and {\clq}qPionýr``. In these days, a lot of people are mistaking scouts and pioneers. At the first sight it seems that their activity doesn{\crq}t differ contently. It{\crq}s partly because of in that time when our country was under control of the communist ideology the mass children organization was {\clq}qPionýr``, on the contrary the scout organization was forbidden. This work gives us an opportunity to learn both organizations in a deeper way. It makes space for the creation of an own opinion about {\clq}qJunák`` and {\clq}qPionýr``.

Evolution of interplanetary coronal mass ejections

Title: Evolution of Interplanetary Coronal Mass Ejections Author: Andrii Lynnyk Department: Department of Surface and Plasma Science Supervisor: RNDr. Marek Vandas, DrSc. e-mail address: vandas@ig.cas.cz Abstract: This thesis deals with deformation of the Interplanetary Coronal Mass Ejections (ICMEs) and their sub-class Magnetic Clouds (MCs) during their propagation in the Solar Wind (SW). The statistical study of the expanded MCs has shown that expansion greatly affects the MC internal magnetic field. We had shown that this influence is more clear for the MCs observed close to their axes. The study of the stand-off shock distance in front of the supersonic ICME confirms a smooth deformation of the ICMEs along their path from the Sun into interplanetary space. We observed that this deformation is increasing with the velocity of the ICME. This study also confirmed the difference in sheaths that are created in front of expanding and non-expanding ICMEs. We found that velocity distribution inside the MC is not uniform and it has large fluctuations. We found that the MC cross-section is usually strongly deformed. This diploma thesis analyses the media image of political party Sdružení pro republiku - Republikánské strany Československa (SPR - RSČ, The Union for Republic - Republican Party of Czechoslovakia), and the media discourse in 1990s. The thesis is focused on analysis of electronic media such as radio and TV talk shows. The time period of the analysis was chosen to capture the period when this political subject was active on Czechoslovak and later on Czech political scene, i.e. 1990-1998. The author discusses the development of the political communication of the party representatives (most of the space is dedicated to media appetence of SPR - RSČ president Miroslav Sládek) and the general political communication in mass media. The goal of the thesis is to analyse the comparison of the media appearance of the party members during the eight years from when the party entered on political scene to its political decline in June 1998. Noise, Vibration and Electromagnetic eFFects Generated by Semiconductor Converters in Traction Vehicles

New problems emerge in the vehicles of city mass transportation due to application of new power switching devices and their control circuitry. Measurements were performed in a low-floor trolleybus ŠKODA 21Tr equipped with power converters based on GTOs and IGBTs. They were aimed at finding the sources of noise and vibrations in the passenger space, at verifying their influence and finding methods for their limitation. Similarly, The same attention was paid to the problems of electromagnetic interference.

Development of access of women as news sources into the news of the Czech Television

The thesis "Development of acess of women as news sources into the news of the Czech Television" deals with access of women into the news programme Udalosti in 1993, 2001 and 2009 in the context of sociology of sources. In the literature, we can find evidence that men dominate television news because they represent one fifth of all news sources and moreover women are more often presented in connection with less important issues and in passive roles. TV news therefore supports stereotypical reception of women in society. It is valid not only for the Czech mass media but also for the whole world. The aim of the thesis was to capture development of access of women during the years. We assumed that frequency of female sources would be higher in 2009 than 1993 and 2001 because we can also see more women in politics and business. We also observed average length of the talks to understand how much space both women and men get in news coverage. The thesis tried to find out contexts in which women and men appeared (e.g. politics, economy, business, entertainment, family etc.) and roles that they held. Furthermore, we focused on position within news coverage and tried to capture whether female sources get a prominent position which indicates capacity and power.

The study "Overweight and personal space borders (psychological aspects of overweight)" concerns the issue of overweight and obesity from the psychological point of view. The theoretical part deals with the general issue of overweight and obesity focusing on the personality of an obese person and tries to find a connection between overweight and a concept of borders of individual's space. The empirical part is focused on mapping the relationship between the body mass index and the ability to control individual's borders within a group of women, using a questionnaire as a tool of research.

Lateral vibration of rotors is significantly influenced by their supports and by their interaction with the medium in the ambient space. If the disc of the rotor is submerged in a liquid and if it vibrates, the pressure field is induced and the liquid acts by a force on the wall of the disc. It is assumed that the disc performs oscillations only with small amplitudes and it enables to describe the produced pressure field by a Laplace's equation and by the relation for the boundary conditions. The liquid is inwettable and it means that it does not lean to the disc surface and therefore no tangential forces acting on the disc are produced. The resulting force is obtained by integration of the pressure distribution around the circumference and along the height of the submerged part of the disc. Its components are proportional to the disc accelerations and it implies that the negatively taken coefficients of proportionality can be considered as additional masses.

Planet Earth, precisely speaking its surface shell, might be considered as a huge, multiphase, batch, biochemical reactor heated by Sun, whose energy is a unique resource. The Sun ensures our earthly life uncondicionally; moreover, it compensated the heat losses of the land, water and ice by radiation emitted from the Earth surface to its surrounding space at night. It is evident that in this macro-reactor the complex transport phenomena occurs in phases;involving mass, heat and momentum. Obviously such a responsive batch system can be only limited by resouces of carbon materials utilized in energy produstion for industrial and chemical processes.
