Evaluation of soil quality by some biological parameters

The methodology summarizes the basic information on applicability of biological methods for evaluation of soil quality. Selected biological methods are recommended as an early and sensitive indicators of contaminated soils and also as indicators with high sensitivity to long-term soil changes originated by management and environmental factors.

The methodics summarizes the basic information on applicability of beneficial soil bacteria Azotobacter spp. for inoculation of field crops. Specific procedures of isolation, screening, selection and maintaining of bacterial strains are described. The technology of inoculant production, and application techniques are involved. The methodics includes evaluation and recommendation of specific farming practices supporting development and activity of Azotobacter spp.

Preconditions for innovative technologies of stand establishment of field crops

This methodology includes the contemporary knowledge on the newest developmental directions of stand establishment of field crops. The attention is focused on minimization and conservation soil tillage and on their influence of the soil properties. Site, agronomical and machine preconditions are evaluated in details for new unconventional technologies of stand establishment of field crops in different soil and climatic conditions of the Czech Republic.

Methodology of ApMV, ACLSV and ASGV diagnostic in apple and pear cultivars and rootstocks by ELISA

The presence of Apple mosaic virus (ApMV), Apple chlorotic leaf spot virus (ACLSV) and Apple stem grooving virus (ASGV) is tested in apple and pear cultivars and rootstocks according to the EPPO recommendation by evaluating symptoms on tree indicators after grafting. This method requires two or three years of planting. For that reason it was prepared and optimised serological testing the presence of ApMV, ACLSV and ASGV by ELISA as a reliable specific methodology. Research results obtained in Crop Research Institute, Prague approve suitability of ELISA for the detection of these viruses in a specific tree tissue during a specific vegetation period. Developed methodological procedures are appropriate especially for state control and supervision laboratories and they are necessary for the certification scheme of fruit trees.

The methodics of plant nutrition and fertilization by potassium

Methodics is found on utilize analytical procedure for determination of content available potassium (Mehlich 3), whose it is possible subsequently use for calculation optimal fertilization.

Principles of cultivation and possibilities of utilization of crambe

Methodics presents overview on botanical characteristics of the plant, requiremenf of the plant on site conditions, farming techniques including fertilization, crop establishment, plant protection till harvest and postharvest treatment. Methodology of detection Penicillium expansum, Monilia frutigena, Botrytis cinerea and Neofabraea alba using multiplex PCR

Multiplex PCR for rapid and reliable detection of species causing rotting apples: Penicillium expansum, Monilia fructigena, Botrytis cinerea and Neofabraea alba is described here. Reaction is optimized for using in a common molecular laboratory. Described method allows both detectino of the pathogens from pure mycological cultrures, and their detection directly from the rot apple tissues. Cultivation of the micromycethes on cellophane surface is also described. References for mycological determination literature are listed, and can be helpful for determination in case of negative results of PCR.

Alternative phosphorus nutrition of plants

The methodology summarizes the basic information on applicability of phosphorus solubilizing bacteria for inoculation of field crops. Specific procedures of isolation, screening, selection and maintaining of bacterial strains are described. The technology of inoculant production, and application techniques are involved. The methodology offers the use of inoculants as an alternative plant nutrition. Results of practical experiments show that P-solubilizing bacteria increase the utilization of phosphorus. Plant growth promoting microorganisms in the inoculant also support the root system and consequently uptake of all nutrients by plants.
