The thesis is focused on getting acquainted with methods of adapting speech signals in spectrum domain and with formants. Fist part is dealing with definition of formant, the relation between Czech language and formants, performing formant features and describing the use of formants in practice. This part includes preprocessing of signals before analysis on its own. This work contains a list of the most common methods for counting speech signals spectrum focused on estimation of spectrum by linear prediction. Practical part contains a verification of the most common methods for specifying formants, which are linear prediction coding and Fourier transformation. Next part is dealing with the description of methods used in created program. It also contains the description of functions of this program and the manual for it. In the end of this work the function of created program is verified and specified database is described.

Effective Algorithms for Verifying Goals in Computer Games
Suda, Martin ; Hric, Jan (advisor)
In the present work we study non-uniform methods for searching game trees of two player games with perfect information. Particularly the non-uniformity based on threats as realized in lambda search and dual-lambda search algorithms is investigated. Threats, de ned as such attack moves that if followed by a pass from the defender result in his certain loss, allow for a reduction of the search space while guaranteeing correctness at the same time. The work then describes a new method for construction of so called relevancy zones, a list of moves or places on the game desk that can only have in uence on the result of the problem in question. Using these zones it is possible to speed up the mentioned algorithms considerably. In the work there are also described three games, AtariGo, Hex and Go-Moku, and their appropriateness with respect to the studied methods is analyzed. Part of the work is also an implementation of the methods for these games using known techniques (transposition tables, history heuristic).

Preparation of radiolabeled albumin
Suchan, Martin ; Trejtnar, František (advisor) ; Lázníček, Milan (referee)
Objectives: This senior thesis was aimed at determining a method of albumin labelling with a particular radionuclide - technetium 99m Tc. Our task was to review the various possible methods needed for working out a procedure for albumin labelling with the mentioned radioisotope and also to test the behavior of labelled albumin in vitro in experiments with newly-isolated rat kidney cells. The next objectives of this work were to analyze the performed experiments, compare the results with other known procedures of albumin labelling with technetium 99m Tc and cast the results in the form of a scientific statement. Methods: A low pH and high concentration of albumin were useful for the albumin labelling experiments. A solution of SnCl2.2H2O was used for the reduction of pertechnetate. Purification of labelled albumin was performed by gel chromatography with a Sephadex G-50. The locations of albumin peaks in the chromatographic fractions were determined by the bicinchonic method. The location of pertechnetate was found by investigating the chromatographic spectrum of pertechnetate. Investigation of the elution spectrum was used to check for proper labelling. The radiochemical purity of labelled albumin was verified by paper chromatography and subsequent detection. Newly- labelled albumin was used in the...

Actual health status of red deer (Cervus elaphus) population in KRNAP region
Bártl, Josef ; Lamka, Jiří (advisor) ; Ducháček, Lubomír (referee)
This study took place in 2006 and 2007 and includes the results of the 2005, is aimed at verifying the health status of wild stocks of Red Deer (Cervus elaphus) in KRNAP in the Czech Republic. The incidence of parasitic infections was investigated by coprological methods (qualitative ovoscopic method and quantitative larvoscopic method) on the Pharmaceutical Faculty of Charles University in Hradec Kralove, Department of Pharmacology and Toxicology. In all yards were from parasitic nematodes most evidenced by the frequency Elaphostrongylus cervi, with less frequency in other nematodes Strongylida family and genus Capillaria. Mycobacterial infection caused by Mycobacterium avium subsp. paratuberculosis and Mycobacterium bovis were investigated by the culture method of mycobacteriosis and serological methods immunodifuse test (IDT) and complement fixation (RVK) on Research Institute of Veterinary Medicine, Center Laboratory, accredited testing laboratory in Brno (registration number: 1354). In this study was not discovered any positive samples. The study shows that mycobacterial infection not occur in wild population of Red Deer, and the parasitic infections in gastrointestinal tract are very low.

The psychology of story
Pithartová Slachová, Helena ; Bahbouh, Radvan (advisor) ; Uhlář, Pavel (referee)
In the theoretical part of dissertation main ideas and concepts of narrative psychology are reviewed. The notion of both narrative and paradigmatic knowing coined by Jerome Bruner is explained. The importance of story and narrative for organizing and understanding human experience is discussed. Other psychological functions of both story and narrative metaphor are mentioned. In the empirical part selected stories that could be considered as a part of European cultural heritage (cultural meta-narratives) are analysed. The methods used for story analysis are TAT (ThematicApperception Test) and CCRT (Core Conflictual Relationship Theme, Luborsky et al.). The aim of the empirical part is to verify the construct of "a latent story". Another goal is to identify repeating motives in the selected stories, that could be used as a form of projective method. Keywords Story, narrative, latent story, narrative psychology, myth.

Rusiňáková, Nikola ; Svobodová, Radmila (advisor) ; Holemá, Irena (referee)
1 Abstract: This thesis is entitled "Selected topics from the educational theme A human and his world; explored employing the methods and techniques of Drama In Education." The first section explores the theory. It not only deals with Drama In Education; its content, principles, form, methods, and planning; but also what a teacher of drama should be; the characteristics of children in the early years of education; the assessment of drama; and of course the theme - A human and his world. The practical section describes the elements of a range of methods and contains individual lesson script. Each lesson is underpinned by a particular method. This section goes on to deal with assigned goals; their verification in practice; and finally, conclusions drawn from the implementation of the planned lessons. In designed lessons I used selected methods and techniques of structured drama and preparatoried games and exercises. I found out that very different topics from this educational theme A Human and his world can be worked up by methods and techniques of drama education because I suggested different topics from this educational theme and I verified them in practice.

Advection character and timing of climatic seasons
Pekař, Bronislav ; Novák, Martin (referee) ; Sládek, Ivan (advisor)
Advection character and timing of climatic seasons Thesis deals with the timing of climatological seasons depending on the temperature and wind direction at 850 hPa level. The theoretical part deals with defining seasons in different ways and climatological processes in Europe. The practical part deals with the verifying of basic hypothesis and the individual steps of the procedure, leading to the timing of climatic seasons depending on the temperature and wind direction. The main method is the method of cumulative series, devised by RNDr. Ivan Sládek, CSc. Western stream over the Bohemia is mainly characterized by cold advection from April 9 to October 21 (advection summer), by warm advection from October 20 to April 9 (advection winter). Eastern stream is mainly characterized by warm advection from April 9 to September 17 and by cold advection from September 17 to April 9. Keywords Advection, circulation processes, climatological seasons, method of cumulative series, timing of climatological seasons.

Vrbata, Petr ; Macháček, Miloš (advisor) ; Vinšová, Jarmila (referee)
Synthesis of Xanthone Derivatives by Microwave-assisted Methods Petr Vrbata This diploma thesis deals with preparation of xanthone derivatives as inhibitors of growth of human cancer cell lines. A new methodology for synthesis of dihydropyranoxanthones 1 and 2, based on the usage of heterogeneous catalysis combined with microwave irradiation, was optimized. This allowed decreasing significantly the reaction time and improving yields. Along with this the Grover, Shah and Shah method (J. Chem. Soc. 1955, 3982) for synthesis of xanthone derivatives was improved due to usage of microwave heating. Using this method a new xanthone derivate 5,8,10-trihydroxy-7H-benzo[c]xanthen-7-on (3) was prepared. Its structure was verified by IR, 1 H NMR and 13 C NMR spectra.
