Auxin Transport in Arabidopsis thaliana: From the whole plant to suspension cultured cells

in English Plants with their sessile life-style are exposed to many stimuli from environment. They have developed mechanisms how to coordinate their growth and development, which allows them to survive sometimes in very difficult conditions. Plant hormones are one of the most important regulators of this signal transduction. Auxins, as the oldest known group of plant hormones, play important role in many physiological processes in plants. To allow perceiving the information in every single cell, auxin molecule is transported by cell-to-cell manner. Auxin molecules enter the cell by passive diffusion or by active uptake by auxin influx carriers. To reach the next cell, they are transported actively out of the cell by auxin efflux carriers. Athough active auxin transport has been studied for almost four decades, past two decades contributed to the identification and characterization of particular auxin carriers remarkably. This thesis contributes to the knowledge on the auxin efflux and influx carriers and their involvement in the processes occurring from a single cell level to the level of a whole plant. Firstly, it brings detailed description of auxin transport characteristics in Arabidopsis thaliana cell suspension cells (Ath cells). Organization of pre-hospital urgent care upon transport accident with mass health disability

This thesis deals with the organization of pre-hospitalization emergency care in the event of a traffic accident involving multiple human injuries. It characterizes the activities of the emergency medical service. The objective of this thesis is to map in detail the organization of pre-hospitalization emergency care in the event of a traffic accident involving multiple human injuries. However, the established structure of the organization applies to any emergency involving multiple human injuries. Two research questions were defined for the research part of the thesis; they focused on the Emergency Medical Service of the Central Bohemia Region and the Emergency Medical Service of the South Bohemia Region in order to obtain research value. The first research question focused on comparing the ways in which paramedics were prepared to deal with emergencies in traffic accidents involving multiple human injuries at the Emergency Medical Service of the South Bohemia Region and the Emergency Medical Service of the Central Bohemia Region. The second research question was intended to map the solutions of traffic accidents involving multiple human injuries at the Emergency Medical Service of the Central Bohemia Region and the Emergency Medical Service of the South Bohemia Region from 2002 to 2012. The methodology selected for the research part of the thesis was a method using a qualitative research conducted through interviews in the form of semi-controlled interviews with paramedics with the Emergency Medical Service of the South Bohemia Region and the Emergency Medical Service of the Central Bohemia Region. The research group was composed of paramedics from two emergency medical services from each of which five respondents were selected randomly. Processing of the results relating to the first research question, which dealt with comparing the ways in which paramedics are prepared to deal with emergencies in traffic accidents involving multiple human injuries, showed that although the structure of the training is largely determined by law and, therefore, it is very similar at both the emergency medical services, there is a striking difference in the access and participation of paramedics to the training organized. The paramedics with the Emergency Medical Services of the Central Bohemia Region do not participate in theoretical preparations regularly, compared to their counterparts, even if they are organized every year. Processing of results relating to the second question, which focused on mapping the solutions of traffic accidents involving multiple human injuries, showed that the Emergency Medical Service of the South Bohemia Region participated in a much more serious traffic accidents involving multiple human injuries than the Emergency Medical Service of the Central Bohemia Region. The description of the procedures selected by paramedics at the scene of a traffic accident clearly shows that they had to use disaster medicine procedures, such as triage using sorting identification cards, for rescue and disposal operations. It can be clearly concluded that they are an emergency medical service far more experienced in the procedures for dealing with traffic accidents involving multiple human injuries are. When examining the organization of pre-hospitalization emergency care in the event of a traffic accident involving multiple human injuries I found out, using the methods of qualitative research conducted through interviews in the form of semi-controlled interviews carried out at two emergency medical services, some differences concerning theoretical training and actual procedures of dealing with traffic accidents involving multiple human injuries. The cause of these differences can be seen mainly in the frequency of occurrences of emergencies in the South Bohemia Region. For this reason, I see the paramedics with the Emergency Medical Service of the South Bohemia Region as more experienced.

The thesis deals with the identification and control of dynamic systems using LabVIEW graphical programming environment. Cascade form of control with PI and PD controllers has been used for regulation and designed in Matlab. These controllers were then tested on real product. Laboratory exercises with model solutions are also included.

Comparison of approaches in determination of transfer pricing in the Czech and Slovak Republics

This thesis deals with differences in approaches of identification and documentation of transfer prices in the Czech Republic and Slovak Republic. The first part determines the need to regulate transfer pricing. The second part is devoted to editing this issue of the OECD and EU directives. The third and fourth section define the framework for transfer prices in the Czech and Slovak law. The last, fifth chapter except the identification of fundamental differences in adjustment in the Czech and Slovak law, thesis deals with the application of the provisions of the judicial decision of the Czech courts, which are supplemented with a solution which would those cases have gone through the litigation in the Slovak law.

Analysis of the training process in the international business

The thesis examines the issue of training and development from a theoretical point of view, then from a practical point of view in two selected international companies (operating in the fields of banking and logistics). The thesis also discusses a problems of European structural funds, especially the request for education projects of an Operational program of the CR "Lidske zdroje a zamestnanost". The theoretical part describes the phases of the educational process: the identification of training needs, education planning, implementation phase and is greatly accentuated phase measurement and monitoring of the effectiveness of training. This work examines new trends in education and development of employees in 2010. The practical part of the bank monitors and evaluates the process of systematic training and use of competency models, practical part in the logistics proposes educational activities under conditions of low input costs - notably through training internal trainers.

Identifikace daňového zatížení příjmů fyzických osob v České a Slovenské republice

Thesis is focused on the identification of the income tax burden of individuals in the Czech Republic and Slovakia . The work deals with the income from employ-ment and income of self-employed persons. Literature review section describes basic concepts of taxation and discusses the individual paragraphs of the Law on Income Tax . In the practical part of thesis are concrete examples showing diffe-rences in taxation in both countries . In conclusion, country which is the favorable tax treatment of taxpayer's income is recommended.

The introduction of diagnosis of new bacterial pathogens Ralstonia a Achromobacter isolated from patients with cystic fibrosis and determining their sensitivity to antibiotics

Cystic fibrosis (mucoviscidosis) is an incurable genetic disease caused by mutations in the CFTR (Cystic Fibrosis Transmembrane Conductance Regulator Gene). The most common causes of increased mortality and morbidity of patients include bacterial respiratory infections which may occur even due to less frequent pathogens. Bacterial species of the Ralstonia and Achromobacter genera are not considered pathogenic for healthy people, but they have been established as pathogens in the sputum of patients with CF. However, due to their phenotypic similarity to other bacterial pathogens encountered in patients with CF, microbiologists often do not pay attention to them. The aim of this thesis was to propose some methods of identification of the genus Ralstonia, to determine both quantitative and qualitative susceptibility towards antibiotics, and to make a bibliographical search focused on the issue of the Achromobacter genus. Strains of Ralstonia spp. were identified using phenotypic and genotypic methods and were tested for the susceptibility towards antibiotics. The thesis also proposes a new method of genotypic indentification of R. respiraculi.

Principy daňových systémů severských zemí v kontextu podmínek ČR

This diploma thesis is concerned with the identification of strong aspects of the tax system of selected Nordic countries and their potential application to the Czech Republic with a target of simplifying and streamlining the Czech tax system and tax collection in general. The theoretical part is concerned with the description of the tax systems of the Czech Republic, Denmark, Finland, Norway and Sweden. Cluster analysis classifies the EU Member States and EEA in individual clusters based on the criteria of tax revenue and transparency of the tax system. The resulting strong aspects of the Nordic tax systems are suggest for application to the Czech tax law.

ANALYSIS OF LOGISTIC PROCESSES IN LOMBARDINI SLOVAKIA, s.r.o. ,

The aim of the bachelor thesis is to describe logistic processes which are running within the particular company and which are inevitable for continuous and effective production in accordance with customers' demands. Further, the purpose is to identify main deficiencies of these processes on the ground of the author's theoretical knowledge in the logistics sphere, and to propose potential improvements. This thesis is divided into two parts. The first, theoretical section is focused on the definition and development of logistics, as a tool for increasing effectiveness and competitiveness of the company. Part of this section is also a description of the concept of minimizing total logistic costs. Second, practical section describes logistic processes and tasks as they are performed in selected company. In this section, author focused mainly on purchasing, warehousing, and material flow through out the company during the whole process of production of final product. As a part of this block is also a identification of main deficiencies and proposed improvements. Čínský realitní trh - současný stav, trendy a vyhlídky

The aim of this work is to describe the phenomena of bubble in the financial, but especially in the real estate market; based on gained information, the current situation of Chinese real estate market is described. This work is divided into theoretical and more practical part. The theoretical part is devoted to the definition of bubble, bubbles in asset market, real estate market bubbles specifics and its causes analysis, identification and implications for the economy. Also, the Japanese real estate bubble is described. The second part describes the evolution of Chinese real estate market and provides overview of the milestones in its evolution. Moreover, it deals with specifics of the Chinese real estate market and explains the factors behind the rise in prices. Further in second part, situation on Chinese housing market is compared with the one in Japan during real estate bubble. Furthermore, this thesis explains how state government can coordinate, regulate and intervene on the market and comes up with short-term outlook.
