The effect of IDGF2 protein on expression of selected genes in cell line Cl.8+

The imaginal discs growth factors (IDGFs) are proteins able to induce proliferation of Cl.8+ cells proliferation in collaboration with insulin. They are also connected with response to starvation stress and innate immunity. A genome-wide analysis of Cl.8+ cells treated with IDGF2, which was performed in our lab, detected tens of genes transcriptionally induced by this protein. The aim of this work was to confirm this effect with some of these genes using quantitative PCR.
Methodology for the phenotyping resistance to Plum pox virus in peach

Sharka disease, which is caused by the Plum pox virus (PPV), is the most economically important virus disease in fruit trees worldwide. Since it is not possible to completely eradicate PPV in the Czech Republic, it is necessary to find another way to control this pathogen. The most promising seems to be the breeding of new cultivars for resistance. This methodology provides a comprehensive protocol for testing PPV resistance in peach. It is based on visual assessment of disease symptoms on leaves and fruits, and determining the presence of the virus in plants using ELISA and RT-PCR techniques.

Methodology of hop latent viroid (HLVd) diagnostics

Viroids belong to the pathogens, which cause important economic losses in quality and quantity of hop cones. Hop (Humulus lupulus L.) as a perennial crop generally grown for nearly twenty years at the same site is seriously endangered by viroid infection. A foolproof diagnostics of viroid infection is necessary for the assessment of healthy state in hop plants.Molecular hybridization (dot-blot) and RT-PCR methods enable a reliable determination of latent hop viroid (HLVd). The methodology consists of the diagnostics as well as its utilization within the evaluation of hop healthy state.
Methodical procedure to utilize Real Time RT-PCR (QRT-PCR) and RT-PCR for potato spindle tuber viroid (PSTVd) detection

The main result is the establishment of molecular detection methods for potato spindle-tuber viroid (PSTVd) in breeding materials from potato Solanum tuberosum. These techniques are able to sensitively and reliably detect this quarantine disease during any developmental phase of potato.
Methodics of molecular detection of Barley yellow dwarf virus in vectors by RT-PCR

Barley yellow dwarf is one of the most important viral diseases of cereals in the world. It can cause significant yield losses in most cereal species, including wheat, barley, rice, corn, oat and rye. In this methodology we provide assays for reverse transcription - polymerase chain reaction (RT-PCR) for sensitive detection of barley yellow dwarf virus directly in the vectors, in aphids. This way it is possible to evaluate infectivity of vectors and from that conclude subsequent measures. The methodology is meant to serve the employees of State Phytosanitary Administration.

Methodology of resistance evaluation of plum, Prunus domesica L., clone C5 to the Plum pox virus and mixed infections with another two viruses

The methodology encloses the procedure of preparation and ínoculation of transgenic plants of plum with the individual viruses, the maintenance of field orchard, the evaluation of virus symptoms in leaves, serological and electronmicroscopical detection of viruses in leaves of plum trees, determination of relative concentration of PPV by DAS-ELISA, detection of PPV-Rec, ACLSV and PDV by RT-PCR, and determination of risk of recombinations between PPV-Rec and the homologous transgenic transcript in P. domestica, clone C5.

A metodical approach to detect viruses belonging to a genus Vitivirus infecting grapevines in the Czech Republic

A metodical approach describes detailed procedure to detect two important grapevine viruses belonging to a genus Vitivirus. Two independent methods are recommended for the detection: RT-PCR and DAS-ELISA.
