Development of model system for study of bacterial adhesion on lung epithelium of CF patients

Cystic fibrosis is an inherited disease bearing a number of health difficulties. The main complication is a chronic colonization and infection of respiratory tract with specific microorganisms - especially Pseudomonas aeruginosa. The lung infection with this microorganism is the most common cause of all of death in these patients. The colonization of respiratory tract is mediated by the series of adhesive structures such as lectin PA-IIL. Currently, the most widely used therapy is an antibiotic treatment. Due to the increasing resistance to antibiotics another methods for treatment are being searched. One possibility is a passive immunization of patients with chicken antibodies. For this purpose, we prepared antibodies against one of the adhesive structures of P. aeruginosa - recombinant lectin PA-IIL. These antibodies be able to recognize a native lectin PA-IIL, expressed by P. aeruginosa. To test the ability of antibodies to prevent adhesion of bacteria on the lung epithelial cells a suitable model system was necessary to develop. Chicken antibodies as a tool of passive immunization against microbial diseases of respiratory tract

Pseudomonas aeruginosa and Burkholderia cepacia are opportunistic human pathogens. Hazards pose to immunocompromited patient groups, most of whom are patients suffering from cystic fibrosis, for which is the infection caused by Pseudomonas aeruginosa or Burkholderia cepacia, often lethal. Bacteria exhibit resistance to most antibiotics and the immunization of patients, paradoxically, worsens the patients' condition. Hen antibodies, unlike mammalian do not activate the complement cascade and do not cause the inflammatory response, therefore, seem to be a suitable tool to protect high risk populations as a means of passive immunization. In this work were prepared specific chicken antibodies against significant virulent factor - bacterial lectins PAIIL and BClA responsible for adhesion. Antibody specificity was demonstrated by ELISA and Western blot. Antibodies were affinity purified and cleaved to fragments. Pneumocytes type II from the lungs of patients with Cystic Fibrosis were isolated and cultured. Tests of the adhesion of bacteria cells were performed on them using ELISA. Comparison of the biocidal activity of disinfectants against biofilm

A common vector of nosocomial infections is Pseudomonas aeruginosa. This opportunistic pathogen is resistant against different classes of antibiotics as well as against biocides. Some P. aeruginosa strains are able to form biofilm. The biofilm formation on medical devices represent a common occurrence that can lead to serious illness and death. Bacteria in biofilms are more resistant than their planktonic counterparts. Therefore it is important to find disinfectants that will be effective also against biofilm. Bactericidal efficacy of four disinfectants has been evaluated on biofilm and on planktonic forms of P. aeruginosa.
Production of mcl-PHA by selected bacteria

The aim of this work is to study the possibility of microbial production of polyhydroxyalkanoates with medium chain length (mcl-PHA). For production two bacterial strains were used: Pseudomonas putida and Pseudomonas aeruginosa. Theoretical part of bachelor thesis deals with the biosynthesis of polyhydroxyalkanoates, their material properties and microbial producers. In experimental part selected bacteria were cultivated on different substrates to evaluate their ability to utilize various carbon sources and accumulate mcl-PHA. Content and composition of mcl-PHA accumulated in biomass were analyzed by gas chromatography with FID. Pseudomonas putida produced more mcl-PHA in medium containing canola oil as sole carbon source when compared with Pseudomonas aeruginosa. P. putida produced 2,11% mcl-PHA in medium containing glycerol as carbon source.

Development of antibiotic resistance in the region Písek in 2000-2010

This bachleor´ s dissertation is aimed to the developement of antibiotical resistance by choosed kinds of bacteria, particulary Pseudomonas aeruginosa, Escherichia coli and Klebsiella pneumoniae in the region of Písek and also in the whole Czech Republic during the period 2000-2010. There is described matter of the antibiotical resistance, antimicrobial agents and also information about the EARS-Net system in the beginning of this work. The practical part contains the evaluation of resistance spread in the region of Pisek according to the data of clinical microbiology of Nemocnice Pisek, the spread of resistance in the Czech Republic according to the EARS-Net and also the consumption of individual groups of antibiotics according to the State Institute for Drug Control. Antibiotical resistance is spreading constantly, for example because of inappropriate usage of the antibiotics or insufficient information about pathogens that cause infectious diseases the most frequently.
