Stress Factors of Volunteers Working With Older People in Residential Facilities in České Budějovice

The bachelor's thesis considers stress factors of volunteering in long-term residential care. The theoretical part of the thesis focuses on the themes of volunteering, ageing and old age and identifies several selected stress factors. The practical part of the thesis represents the data interpretation from in-depth semi-structured interviews with volunteers who are managed by volunteer centres in České Budějovice and who also visit older people in cooperating long-term residential care.
Stress management in a selected company

The thesis deals with the issue of stress and stress management in a selected organization. The aim of the thesis is to examine the stress factors and the impact of stress in the selected organization, including suggestions for changes in the current stress management in the organization. The sub-objectives of this thesis were to find out how the employees handle stress, how they approach stress management and also how they prevent stress and to recommend solutions if deficiencies are found. Based on this information, recommendations and suggestions were made to improve the current issues in the selected organization. For all the suggestions and recommendations, financial costing of the cases were graphically made. From the results, it is evident that the respondents are aware of the dangers of stress and burnout, irregular diet or lack of relaxation techniques, but do not practice any of these and do not actively address them. Therefore, I recommend assistance in implementing changes from the company and management, whether it be more emphasis on creating separate and enclosed spaces in the offices or other methods aimed at creating places of calm. The stated aim of the thesis has been fulfilled and its contribution to practice can be considered significant if the promising chance of using some of the proposals in the offices of OVB Allfinanz a. s. in České Budějovice turns into reality.
Stress management and professional focus of workers

The thesis deals with stress management and the problem of stress in the workplace. The methodological approach is based on a questionnaire survey of three selected companies - Madeta a. s. (Madeta), ESW Bohemia k. s. (ESW) and the Central Institute for Inspection and Testing of Agriculture (ÚKZÚZ). The questionnaire was created on the office.com web-site and it contains 23 questions. The data are processed using appropriate graphs and tables through Microsoft office Word and Microsoft office Excel. The results show that in Madeta, most of the respondents are under stress (labourers) - they work in three shifts, in a fast pace most often individually and they are unable to resist the stress. In ÚKZÚZ (administrative workers and non-production type technicians), they work the morning shift in a calm pace and show little stress. Employees are aware of the consequences of long-term stress and therefore take care of themselves through appropriate activities. In ESW (administrative workers and production-type technicians), they work mainly in the morning shift and often in teams. Working in a team helps to reduce stress. Employees take care of themselves in appropriate ways to reduce stress.
The extent of stress factors in the work of social worker in social services for seniors

The bachelor thesis is focusing on the description of the most important stress factors for social workers in social services with the elderly. The theoretical part presents thepractice in field of home personal assistance and residential home care and summarizes possible stress factors in both areas. The practical part identifies the most challenging moments and situations for the social workers in both areas. The foundation of this thesis is therefore a senior, a social worker and stress factors arising from his work with the elderly.

The main aim of this Bachelor thesis is to understand better how to deal with stress, which types of stress factors do we differentiate and how to predict them. The research is based on cooperation with company, which produces a machinery for automotive and medical industry. Accumulation of phenolic compounds in Spirogyra in stress conditions

Phenolic substances represent one of the major groups of secondary metabolites of higher plants that play a role in a number of biotic and abiotic interactions. Among algal groups, these substances are produced, for example, by conjugating algae (Zygnematophyceae), which are currently considered to be the closest living relatives of higher plants. The common function of phenolic substances is protection against the harmful effects of ultraviolet radiation, or even an excessive amount of photosynthetically active radiation, which was supported by the induction of their synthesis after exposure to UV radiation in several ecophysiological studies. However, the range of applied stress factors and their effect on changes in phenol levels is limited. In this diploma thesis, the genus Spirogyra, for which the chemical structure of a number of specific phenolic substances is known, was exposed to selected stress factors and their influence on cell morphology, maximum quantum yield Fv/Fm and quantity of phenolic substances was monitored. A significant increase was observed after application of UV-B radiation, higher level of PAR, low temperature and osmotic stress, while at high temperature this effect was not evident. Changes in the contents of anthocyanins and photosynthetic pigments in barley leaves under the effect od climate change factors

Ongoing climate change and its manifestations have a significant impact on agricultural crops, which are exposed to a wide range of abiotic (drought, high or low temperature, elevated UV radiation) and biotic (pathogens, pests) stress factors that can dangerously affect their growth and yield. One of the very important phenomena of climate change is the increasing concentration of CO2 and other greenhouse gases in the atmosphere. Manifestations of climate change, such as high temperature and drought, interact together with increased CO2 concentration, and these interactions can have serious effects on vegetation. This diploma thesis examines the effect of climate change factors and stress factors on the content of pigments with the importance of plant protection against stress, namely photosynthetic pigments and anthocyanins in barley leaves. The aim of the thesis is to find out whether and what are the differences in the response to these factors in two varieties of barley, differ in their resistance to photooxidative stress. Characterization of biological effects of extracts obtained from yeasts and autotrophic microorganisms

Carotenogenic yeasts and microscopic autotrophic microorganisms are increasingly being studied in the biotechnology industry because of their ability to produce a variety of substances with a wide range of utilization. These substances mainly include lipids and carotenoids, which are used in the food and pharmaceutical industries. The aim of this bachelor thesis is to determine the way these microorganisms behave in the presence of stress factors, and which of them would have a positive effect on the production of metabolites that can be further used in various industries. The theoretical part is dedicated to a characterization of substances produced by carotenogenic yeasts, cyanobacteria and microalgae, which will be further analyzed in the experimental part. The metabolites mainly include carotenoids, which are a wide group of naturally occurring pigments that possess, among other things, antioxidant and provitamin activity. No less important substances produced by these organisms are lipids, which are part of cell membranes, they also serve as a reservoir of energy, and the substances contained in lipids are part of the precursors of many hormones. Yeast cell walls also contain -glucans, which have many beneficial effects on immunity and human health. Due to this fact, interest in their research have been rising recently. The analytical methods used to determine these substances are also mentioned, as well as stress factors and their effects on the cells of the mentioned organisms. The experimental part deals with the exposure of these microorganisms to oxidative stress and stress caused by heavy metals. The aim was to analyze which concentrations of these stress factors are lethal to microorganisms, and which of them are optimal for increase of the production of the above-mentioned substances. Of the yeast strains, the strains Sporidiobolus pararoseus and Rhodotorula mucilaginosa achieved increased production of metabolites due to selenate ions. In contrast, cobalt had a beneficial effect only on the accumulation of lipids and -glucans in cells. The positive effect of selenium was also observed in the strain Chlorella minutissima and Arthrospira maxima, however, the most remarkable changes in the production of metabolites were achieved by the addition of the optimal concentration of the solution inducing oxidative stress.
Monitoring the influence of stress factors on yeasts of the genus Metschnikowia using molecular techniques

Molecular techniques are used widely nowadays, especially in the identification of various species of microorganisms, and provide reliable and accurate results in a relatively short period of time. This work deals with the characterization of Metschnikowia yeasts exposed to various stress factors and the monitoring of their genomic DNA by PCR-DGGE and PFGE methods. The work was focused on the analysis of ribosomal rDNA, specifically the ITS1 and 5,8-ITS2 regions and genes encoding the domains D1/D2, that are part of the large 26S rDNA ribosomal subunit, which are commonly used in the characterization of fungal eukaryotic communities. Three types of stress factors were selected for the experiments: osmotic stress (NaCl environment), oxidative stress (addition of H2O2 to the medium) and nutritional stress (addition of hemp flour / leaves and flowers as a carbon source). The analysis was performed for particular strains Metschnikowia andauensis, Metschnikowia pulcherrima, M. chrysoperlae, M. shanxiensis, M. sinensis and M. zizyphicola. The results showed that addition of the NaCl, H2O2 and hemp components at higher concentrations to the production media does not disrupt the ribosomal DNA when detected by PCR-DGGE. Mutations have not been observed by comparing these strains with yeast that was cultivated under optimal conditions. Despite stress factors, PFGE analysis of karyotypes showed that DNA of some yeast species does not prove any damage and remain intact. Remaining strains proved certain degree of damage, and bands were not detected on the gel for these strains. In the given circumstances, it can be stated that the high adaptability of these species to a stress environment makes them promising biotechnology producers. These yeasts have great potential for usage in agriculture as a tool for biocontrolling of fruit or vines.
