Rada, Vojtěch ; Junasová, Veronika (referee) ; Šteffan, Pavel (advisor)
This diploma thesis is focused on wireless charging technology according to the Wireless Charing 2.0 standard by the NFC Forum and the design of an ASIC circuit of a device functioning as a listener, where the primary emphasis is placed on the digital design of this circuit. In the beginning the work deals with a general theory, gradually discussing all the standards from the NFC Forum that are necessary for the control protocol design according to the Wireless Charging 2.0. Subsequently, the analysis of a real communication in the development kit for this standard is conducted, based on which the system design of the digital circuit is made. The designed circuit is described by VHDL language and verified by running the basic simulations to check the functionality of the designed parts of the circuit. In the final part of the thesis, a prototype module with the proposed system is fabricated, implemented in an FPGA with a connected battery charger circuit and its functionality is tested in practice.
Test benches for microcontroller kits
Žaludek, Ivo ; Burian, František (referee) ; Bradáč, Zdeněk (advisor)
This thesis deals with the design of a test bench for functional testing of educational kits. These kits are in the form of standalone printed circuit boards, for instance, a module with a microcontroller and its peripherals, such as a display, keyboard, or external memory. A comparison of commercially available testers is conducted, and the requirements for testing of each printed circuit board are identified. Based on these requirements, the design of both the hardware and the software of the test bench was developed. The final stage of the project is the evaluation of the functionality of the test bench.
Influence of production environment on the occurrence of contaminating Penicillium in smear ripened cheese
Veselá, Vendula ; Brázda, Václav (referee) ; Vítová, Eva (advisor)
The topic of this thesis is the monitoring of the environment in a cheese production plant specializing in cheese ripened under a smear rind. The aim is to evaluate the occurrence of Penicillium mold as the main contaminant microorganism, depending on the sampling location and production time. Air samples were collected throughout the production process using the sedimentation method at 21 designated locations. The total number of microorganisms, selected groups of microorganisms, and the target microorganism (Penicillium mold) were monitored in the collected samples using cultivation (according to ČSN standards), microscopic, and molecular diagnostic techniques. The aim was to evaluate the occurrence of Penicillium citrinum, chrysogenum and commune species in dependence on the sampling location and production time. As part of the production screening, the broader microflora was first evaluated, followed by monitoring for contaminating molds. Using macroscopic and microscopic methods, 36 morphologically distinct colonies were selected. Microscopic analysis confirmed the presence of bacteria, yeasts and molds of the genera Penicillium, Cladosporium and Aspergillus. In order to identify the microorganisms present, an analysis using the polymerase chain reaction (PCR) method was performed, preceded by DNA isolation using the NucleoSpin Microbial DNA kit and phenol extraction. Based on real-time PCR results, the occurrence of contaminating Penicillium and Cladosporium molds was detected even in the early stages of production. Molds from these genera are found throughout the entire facility except for the single room that serves as a curd cooler before forming. Aspergillus mold, in the form of yellow mold with white mycelium on the edge of the colony, was detected only once in the drying room where cheese ripening occurs, and confirmed by microscopic observation. The genus Cladosporium was found in five morphologically distinct forms in the production environment. In addition, DNA was isolated from 11 morphologically distinct bacteria. Real-time PCR identified this DNA as belonging to coryneforms, with four of them belonging to the genus Brevibacterium. The genus Penicillium was represented by 13 morphologically distinct colonies, real-time PCR confirmed the presence of Penicillium commune and Penicillium chrysogenum. In addition to the mentioned molds, 11 morphologically distinct bacteria were detected, from which DNA was isolated. Real-time PCR identified this DNA as belonging to coryneforms, with 4 of them belonging to the genus Brevibacterium. The results of this work completely map the contamination of the production environment of the plant, and based on these results, methods for environmental sanitation were proposed to ensure the decontamination of the production process.
Optimization of mycovirus detection methods obtained from NGS data analysis
PÍCHALOVÁ, Barbora
One well-known pathogenic fungus Armillaria causing Armillaria root disease which is characterised by infecting the root system of woody plants, resulting in basal decay and the development of a white syrrocium under the bark of trees. Several genome sequences of mycoviruses have already been published in research on this disease, including viruses from the Mymonoviridae, Botourmiaviridae, Partitiviridae, Virgaviridae as well as a newly described group of viruses known as ambi-like viruses. In this study, alongside ambi-like viruses and partitiviruses, by in silico analysis of virus sequences obtained via NGS methods unveiles the identification of previously unknown beny-like viruses within Armillaria spp. from the Czech Republic. To confirm the presence of beny-like viruses in vitro, reverse transcription with PCR was used. Total RNA was isolated using a commercial kit and its presence in the samples was confirmed electrophoretically. The sequence obtained using the Illumina system's NovaSeq 6000 sequencing platform was used to design the triple pairs of primers Beny F1 and Beny R1, Beny F2 and Beny R2, and BV1 F and BV1 R, in which potential ORFs were designed. The primers were designed to target the longest ORF encoding RdRp, the most conserved gene in RNA viruses. The DNA fragment obtained by reverse transcription was amplified by PCR followed by reamplification. Finally the samples were evaluated electrophoretically for the presence of viral single-stranded RNA.
Thrombin generation in pathophysiological conditions
Malíková, Ivana ; Kvasnička, Tomáš (advisor) ; Hirmerová, Jana (referee) ; Úlehlová, Jana (referee)
Introduction: Thrombophilic conditions may be accompanied by increased thrombin generation (TG), especially during pregnancy when the increase in thrombin generation results from a series of changes in the haemostatic system. During administration of anticoagulant treatment, it is desirable to reduce the increase in thrombin generation. Aims of the study: The first aim of our study was to compare TG in individuals with a proven factor V Leiden mutation and individuals with elevated levels of factor VIII. Another aim was to compare patients with VTE and also with or without proven FV Leiden mutation that are taking different anticoagulation treatments. The final aim was to assess thrombin generation in pregnant women and their response to LMWH administration during severe thrombophilic conditions. Group of patients and methods: There were included 170 healthy blood donors in the control group, the congenital and acquired thrombophilia group included 44 individuals with FV Leiden mutation and 38 individuals with factor VIII level >150 % without other proven thrombophilia. 347 subjects with VTE taking direct coagulation inhibitors were included in the anticoagulation group. Sixty women with a prophylactic dose of LMWH were included in the group of pregnant women. Kit Technothrombin® TGA RC Low and RC...
