Vitality in the architectonic environment

More and more research studies are being conducted on the quality of architectural space as a factor affecting our mental, social and physical health. In the past, this topic was often overlooked in its complexity. The establishment of the specialized fields of building biology, biophilic architecture and the works of Christopher Alexander and Nikos Salingaros now allow us to consider various facets of this subject. This dissertation aims to analyse the topic of vitality in architectural space by studying the abilities and perceptions of its users. Therefore its aim is not to develop a new theory of “vitality-enhancing architectural space”, but to describe how this phenomenon is perceived by the users themselves. Users can provide feedback on which features of architectural space they see as the most important for the feeling of vitality. These features have been hierarchically organized based on their perceived importance, analysed and described in detail to allow their relatively easy application in practice. This dissertation will also allow a comparison with other existing works on the studied topic.

It is as a matter of fact renascence and update of practice, that was used in the past either consciously or unconsciously, but during modern age forgotten or inhibited. Dissertation also brings in resume of practical instrumental means of vital architectural creation.
Investigation of yeast properties during the beer fermentation

In this diploma thesis, the properities of brewery’s yeasts during the operation beer fermentation in the selected brewery and dependences of diferent agents and their influence on viability and vitality of the yeasts were studied. Other observed properities were physiological state, pH, temperature, a value of soft and coarse sludges, a value of dissolved oxygen in wort, the level of wort’s fermentation. It were collected the samples of fermenting wort and washing yeasts for determination. The used yeasts were observed from the first time until thein last used (generally 4 times). pH, temperature, apparent fermenting and total numer of yeast cells were measured in the collected samples. The used yeasts were washed before every other application and vitality was determined. The number of death cells were observed by this method. The number of death cells never fell bellow 5 %. The value of dissolved oxygen in wort is also the important agent for the growth of yeasts. The touchs with low number of oxygen showed that the time for fermentation had to be longer (by about 3 or 4 days more). The touchs with higher number of sludges showed that the sludges haven’t any influence on the yeasts and the procces of fermentation. Confirm the accuracy of the repeated used of the yeasts was the object of this thesis.

Non-structural carbohydrates (NSC) are one of the most important organic compounds in the plant growth. Concentration of NSC was studied in the mature Scots pine trees (Pinus sylvestris L.) with artificially disrupted phloem connection between the crown and the roots by stem girdling and in the controls within the vegetation seasons 2013 -- 2014. The highest concentrations of NSC in the needles were at the beginning of the vegetation season, before the needle flush and beginning of radial growth of the stem. While the concentration of the soluble saccharides was not different between the girdled and control trees, the concentration of starch was significantly higher in girdled trees at the beginning of a season. In contrast to the needles, the highest concentrations of NSC in the wood were found in the summer and lowest in the June. The starch content in the wood o girdled trees was significantly higher at the end of the season. The concentration of soluble saccharides was similar in all variants of experiment for the entire season. The NSC may provide and information about the physiological vitality of the tree, however their use as a simple indicator is complicated by the normal seasonal dynamics. The results provide an important contribution to understanding of underlying physiological processes necessary for the correct prediction of the tree survival time.

The goal of the work was to find out, what influence different ways of regeneration of silver fir have on its growth speed and overall vitality seven years after plantin. Study was carried out on research area ÚZPL -- LS LČR Svitavy, district of Radiměř. The fir was planted on area partially covered by forest wall, in v gab, with simultaneous planting of fir and larch. There was total of 7 research plots located on acidic stands between 4th and 5th LVS (forest vegetation grade) and variables measured were: losses, height, terminal growth of particular years, width of root collar, total number, length and width of branches, length of needles, colour of needles, shape of crown and trunk, frost and other damage. Results show, that different manners of planting (cover) do have an measurable influence on growth rate and vitality of silver fir. The best results were reached on area partially covered from multiple sides by standing forest and in gab, where frost damage was minimal.

Seed vigor was evaluated as germination percentage at low temperature (10 °C) and drought (-0,2 MPa), and also as germination parameters (energy, rate and index). 133 double-haploid lines (DHL) and their parents were evaluated in six environments. The total variation consisted of 51% by environments, 8% by lines and 37% by their interaction. The better parents showed higher vigor when harvested in worse location. The other parameters were controlled similarly (34,7 -- 41,0% location, 24,9 -- 32,9% year and 8 -- 37% by the lines. The results can be used as support for breeding for higher grain vigor not only for better agronomic but also for higher malting value.
