Determination of lipids and fatty acid profile in a barley caryopsis

The diploma thesis deals with the determination of lipids and fatty acid profile in a barley caryopsis.(Hordeum vulgare). The theoretical part describes the synthesis of fatty acids and their degradation in plant material, secondly, it described the possibilities of lipid extraction and their determination and the possibilities of determination of fatty acids. In the experimental part method of lipid extraction on automated extractor Fex ®IKA and determination of fatty acids by GC-FID were optimized. For analysis of fatty acids two capillary columns SLB-IL 100 and Supelcowax were compared. Twenty varieties of barley from the year 2008 and twenty varieties from the year 2009 were compared based on the content of lipids and the representation of fatty acids in a barley caryopsis. The diploma thesis was realized in the Research Institute of Brewing and Malting, Plc. in Brno.
The assessment of chosen characteristics of cheese fats

This bachelor’s thesis deals with assessment of selected fat characteristics of natural edam cheese. The theoretical part deals with general characteristics of natural cheese and structure of individual components. The main objective was to introduce and optimize methods in a laboratory to simple analysis of fat in cheese. Fat numbers were determined. To optimize were used samples, which were purchased at regular food store. With optimized methods were analyzed samples of cheese produced in cooperation with the Institute of Food Technology of Tomas Bata University in Zlin.
The assessment of fatty acids in cheeses

This bachelor thesis deals with the determination of fatty acids in natural Edam cheese. The theoretical part briefly characterize the natural Edam cheese, main attention is paid to lipids, fatty acids and the possibility of their determination by gas chromatography. In the experimental part the content of fatty acids in the model samples of natural Edam cheese produced at Tomas Bata University in Zlín was determined. The method according to ISO 1735 was chosen for extraction of lipids from the samples , the method according to ISO 5509 using methanol solution of potassium hydroxide was chosen for the esterification of fatty acids ,. The resulting methyl esters were determined by gas chromatography with FID detection.
The application of microorganisms to the degradation of lipids

This bachelor thesis is focused on the study of commercial products for degradation of lipids in waste water. This commercial products contain microorganisms with lipolytic activity. Theoretical part of this thesis describes lipids, lipases, mikroorganisms which produce lipolytic enzymes and basical principles of cleaning of waste water. In this part analytical methods of lipids isolation are presented too. The practical part of this thesis is concerned with the study of three commercial products. These products were tested for the content of microflora. Subsequently the restriction analysis was performed. Using this method the microorganisms were identified as Bacillus sp. Furthermore the growth curves were determined. This curves show growth of biomass. Then the pH during cultivation and lipolytic activity of microorganisms were determined. Lipolytic activity was determined by spectrofotometric method with using of p-nitrophenyllaurate.
Utilization of biomass for the energy purposes

The theoretical part of work focuses on the issue of biomass used for energy purposes, in particular agricultural waste, which can be used as a substrate for biogas station. Furthermore, examines the composition of polymer substrates, which affects most biogas process. The aim of this work was to measure each sample of enzyme activity of biomass, which is used as a raw material for biogas plants.
Role adipokinetic hormone in fruit fly \kur{Drosophila melanogaster}

The aim of the thesis was evaluation of the adipokinetic hormone (AKH) effect on the fruit flies Drosophila melanogaster with AKH production deficiency, and adenosine receptor dysfunction. A nutrition level was measured in those flies to evaluate involvement of AKH and adenosine into the metabolic pathways. Results revealed that application of external Drome-AKH significantly increased the free glycide level in the fly bodies. On the other hand only limited effect of hormonal treatment on other nutrients (lipids, proteins) was recorded. The role of adenosine was unclear, but its involvement cannot be excluded.
Analysis of low abundant lipids in vernix caseosa using chromatographic methods and mass spectrometry

The aim of this work is to develop a separation method for characterization of unknown lipids in vernix caseosa. We focused on very low abundant lipids which have not been previously described in vernix caseosa. The total lipid extract was fractionated using low pressure normal phase liquid chromatography to obtain sub-fractions. The sub-fractions were further separated in high performance liquid chromatography mode to isolate classes of lipids. This procedure enabled us to characterise lipid classes by mass spectrometry and identify one of the class as acyl-ceramides with non-hydroxylated fatty acids.
Determination of optimal conditions for cell growth and accumulation of storage lipids of the alga Trachydiscus minutus

Algae are becoming a strategic source of fuels, food, feedstocks, and biologically active compounds. Algal lipids are among the most promising potential products. The yellow-green alga Trachydiscus minutus (class Eustigmatophyceae) is unicellular freshwater alga accumulating lipids as its storage reserves. The influence of various growth média, temperature and light intensity on the growth and accumulation of storage lipids of alga Trachydiscus minutus was studied. The optimal growth conditions were determined. Storage lipids were quantitatively determined fluorometrically. Accumulation of lipids was monitored by fluorescence microscopy.

Main thought of this work is research of digestive apparatus of senior people in relation to (healthy) lifestyle. In introduction I explain basic terms connected with nutrition, health, illness and prevention. In the next chapter I put attention to identify health condition and signals that human body gives when it not getting all that it needs. The content part of this work is focused on determinants that influence our health. I put attention to complex point of view. Reader will get information what is the optimal drinking regime, what macronutrition and micronutrition body needs, what it the categorization of those and what food we can get those from. I go to detail in needs of movement, rest, sleep and mental hygiene. This material generalizes actual needs of healthy lifestyle that can be used as help, to build and to strengthen in long term right habits for health and vitality.
