The Economics of Happiness: Are the busier employees the happier ones?

The thesis analyzes the relationship of the perceived happiness level and the level of work busyness of full time employees. For this purpose, a questionnaire was built which mapped happiness and busyness with considering both subjective and objective variables. 205 employees participated in the questionnaire survey. The sample showed above-average education, a high level of busyness and above-average salary (or wages). It has been shown that the work happiness level depends on the level of workload: the unbusy employees are the least happy ones. Influence of ambitions or influence of the importance of work area to the relationship of happiness level and workload level has not been confirmed.
Expression of osmotin, an antifungal protein from Nicotiana tabacum in Escherichia coli

Plants have evolved a huge variety of proteins involved in the defense against pathogens and adaptation to stressful environments. Plant proteins whose expression is strongly induced in response to infection by pathogens belong to the group of pathogenesis-related (PR) proteins. The family of PR-5 proteins constitutes a group of cysteine-rich proteins including thaumatin, zeamatin and also osmotin. Osmotin is a cationic protein of 205 residues and molecular weight of 24 kDa. It was discovered and characterized in cells of Nicotiana tabacum var. Wisconsin 38. The plasmid harbouring cDNA of osmotin from Nicotiana tabacum was constructed for transformation of Escherichia coli. The osmotin gene was prepared in fusion with histidine tail to facilitate the isolation and purification from bacterial cells. Selection of transgenic colonies was based on antibiotic resistance. The hexahistidine-tagged osmotin was overexpressed in heterologous system by using pET expression vector and purified using immobilized metal affinity chromatography. The expression of osmotin was detected and antifungal activity was tested.

The book "Artificial Intelligence Methods in geoinformatics", (editors: Voženílek, V. - Dvorsky, J. - Husek, D) issued by the University of Palacky in Olomouc (in 2011. 184 pp. ISBN 978-80-244-2945-8) is the final outcome of the project GACR 205/09/1079 - "Artificial intelligence methods in GIS." The aim of the book was to summarize the results of research on the application of artificial intelligence methods for processing, visualization and interpretation of spatial information in the area of geographic information systems. The project was a joint research activity of experts from various research fields of Palacky University in Olomouc, VSB-Technical University of Ostrava, and Institute of Computer Science, Academy of Sciences Czech Republic targeted to the integration of methods used in solving problems of artificial intelligence and methods of processing spatial data, whose integration is essential for the development of new modern research methods and technologies.
Igor Stravinskij: Story of the Soldier

In m y thesis I deal with life and work of Igor Stravinskij, concentrating on one of his masterpieces, Story of the Soldier. In the first part I write about life and work of this important russian composer,then I continue with the description of my concept and direction of the work which had its first night in November 2005 in theatre Minor. This thesis contains detailed description of the scene and costumes for Story of the Soldier, together with certain problems connected with its presentation. In my diploma thesis I mention personality of Charles Ferdinand Ramuz and Sergej Pavlovič Ďagilev too , both tightly connected with Igor Stravinskij.
