Metabolic Effects of L-Carnitine Supplementation in Hemodialyzed Patients
Cibulka, Roman ; Racek, Jaroslav (advisor) ; Zima, Tomáš (referee) ; Eiselt, Jaromír (referee)
Chronic kidney failure is associated with many kinds of metabolic disorders caused by the kidney disease and also attributable to dialysis treatment. Phenomena such as accumulation or deficit of various substances and dysregulation of metabolic pathways participate in the pathogenesis of these changes. One of these disorders, which we studied in more detail, is a deficit of carnitine. Carnitine is a substance which plays an essential role in beta-oxidation of fatty acids by catalyzing their transport into the mitochondrial matrix. It enables obtaining of energy, namely in muscle cells including myocardium. Patients with chronic kidney failure treated by hemodialysis are known to have decreased carnitine concentration in plasma and tissues due to its impaired synthesis in kidneys and the great loss across the hemodialysis membrane during dialysis sessions. A single hemodialysis session reduces plasma free-carnitine concentrations to about one-third of their predialysis values because of small molecule of free-carnitine. On the other hand, renal elimination of acylcarnitine, which is physiologically ineffective, may be impaired in chronic kidney failure, leading to increased blood concentrations of acylcarnitine. Considerable evidence suggests that carnitine deficiency and abnormalities of carnitine...

Mechanisms of activation and modulation of the TRPV1 receptor: relationship between structure and function
Touška, Filip ; Vlachová, Viktorie (advisor) ; Teisinger, Jan (referee)
Vanilloid Receptor Subtype 1 - TRPV1 - is an important member of the TRP ion channel family. The physiological role of this ion channel is to integrate the nociceptive signals from the peripheral nervous system. TRPV1 participates in the detection of noxious stimuli on the primary afferent sensory neurons: of the chemical stimuli, such as capsaicin, resiniferatoxin, inflammatory mediators, endogenous vanilloids, and of the low pH (<6.5), and of the noxious heat stimuli (>43řC). Apart from the high expression on the dorsal root ganglion neurons and trigeminal ganglions, the functional expression of this receptor has been also found in the central nervous system, where its role remains elusive, however. Structural-functional studies which have been carried out since the receptor was first cloned, that is 13 years ago, have revealed several specific characteristics. Nonetheless, a number of questions remain to be answered, particularly those concerning cellular mechanisms and regulation of TRPV1's activation, modulation, and its complex function in the nervous system. The aim of this diploma thesis was to characterize, using electrophysiological and fluorescence techniques, the activation and desensitization kinetics of recombinant TRPV1 receptor in relation to the dynamic changes of intracellular calcium....

Ullrichová, Jana ; Šatínský, Dalibor (advisor) ; Matysová, Ludmila (referee)
Development and validation of HPLC method for separation and determination of active substances in pharmaceutical preparation Valetol Jana Ullrichová Abstract A method for the determination of paracetamol, propyphenazone and caffeine by the high- performance liquid chromatographic method with ultraviolet detection has been developed and validated. The analysis was performed at the room temperature in an isocratic mode on the reversed phase ODS Hypersil 5 µm C-18 column (250x4,6mm). A mobile phase (water : 2-propanol : diethylamine : methanol (50 + 15 + 3 + 32) adjusted to pH 7.5 by means of phosphoric acid) was suitable for the separation and the determination of paracetamol, caffeine and propyphenazone. UV detection was applied at 273 nm. Injected volume was 5 µl, flow rate 0,5 ml.min-1 . The developed method is sensitive and selective and can be applied for the routine studies of pharmaceuticals in the tablet form.

Analysis of the approved territorial plan of the small community and the proposal for its complement and change
MÜLLEROVÁ, Veronika
This thesis concerns an analysis of the territorial plan of human settlement Svatý Jan nad Malší. Surrounding villages, such as Sedlce and Chlum nad Malší, were described in terms of the greatest changes, closer look can be provided on the cadastral plan of the town Svatý Jan nad Malší. Based on the study of territorial planning documentation and field exploration, individual parts of the territorial plan were evaluated, inconsistencies with actual condition were pointed out and necessary changes were proposed. To achieve completeness, questionnaire survey was conducted, from which the most problematic sections emerged, mainly work opportunities in village, condition and maintenance of roads and possible options of new construction. At the end, general maps  of territorial plan were designed in ArcGIS software according to particular cadastral areas with advised changes.

Main results and benefits of work Benefits of this work can be seen both in theoretical and in ernoirical level. With respect to theoretical part the analysis of the newest trends in development of institutional theories is very important benefit. The need of proactif engagment of public sector into increasing of competitivness of region by form of strenghtening of inovation potencial and ability to learn is part ofthese trends. In this connection three questions are discussed, which are in centre of interest of institutional theories in the last time: a) necessity of territorial proximity of firms as a condition for successfull process of learning b) the character ofrelations among sectors c) other sectors than high-tech sectors can be prosperous on the basis of practical experiencies of author can be possíble to suppose, the 3 factor impede to wider application of institutional theories in Czech Republic: a) complicated concept of institutional theories b) underestimating of concept of of institutional theories, misunderstanding of function of public sector c) insufficient data base Typologies of actors and instrurnents of regional policy are important benefits of work as well. First typology ranges of actors of regional development in compliance with scale activities. Types of instruments disponible to...

Germination and early growth of buckwheat (Fagopyrum aesculentum) after low-temperature plasma discharge treatment in air gas generated in various apparatus were tested. Three pretreatment times were used: 180 s, 300 s and 600 s. Number of germinating achene, length and weight of sprouts were measured. The data was analysed with two-way ANOVA. A significant influence of the type of plasma apparatus, time of exposure and combination of both factors were found. The positive effect on germination and early growth was observed after application of GlidArc plasma. The opposite effect of PDBV apparatus, which is characterized by plasma with a high power density, was observed. Shorter pre-treatment times for this apparatus must be therefore used.

Talová, Dagmar ; Palaščáková Špringrová, Ingrid (advisor) ; Pánek, David (referee)
Title ofwork: Inteďerence ofan overactive bladder by n. tibialis stimulation Aim: The aim of this work to was authenticate whether a 12-week-long Stoller's afferent nerve stimulation can influence the activity of the detrusor of the urine bladder through its inhibition and in this way improve some objective and subjective parametres of urgent incontinence (uroflowmetry, filling cystometry, OAB V8 questionnaire). Method: The group consisted of seven female patients with overactive bladder. The therapy of afferent nerve stimulation lasted for 12 weeks. Before the start of the therapy, and a:fter its completion all the patients were examined for urodynamic functions (uroflowmetry, cystometry). All the patiens also filled in the OAB V8 questionnaire- before and after the therapy- in which only subjective feelings were assessed. Results: The hypothesis in which we presupposed the descensus of the hyperactivity of the urinary bladder was validated with 75% of the patients which was demonstrated by urodynamic examination. Further hypothesis in which we presupposed the increase of the capacity ofthe urinary bladder was not possible to validate or refutate as the average increase of the capacity of the urinary bladder was statistically insignificant. Stoller's afferent nerve stimulation has a long-term...
