Magnetic resonance imaging (MRI) provides a useful noninvasive method to study the long-term migration and fate of transplanted stem cells in the central nervous system in vivo. Grafted adult as well as embryonic stem cells (ESCs) labeled with superparamagnetic nanoparticles survive in the host organism and migrate preferentially into a lesion site, where they populate the damaged nervous tissue. The migration is not affected by the route of administration; the lesion is populated with the same number of cells after intracerebral grafting as after intravenous injection. Less than 3 % of transplanted mesenchymal stem cells (MSCs) in a cortical photochemical lesion differentiated into neurons and none into astrocytes, while most ESCs (70 %) differentiated into astrocytes and only 5 % into neurons. The intravenous injection of MSCs or of the mononuclear fraction of the bone marrow, which includes hematopoietic and nonhematopoietic stem cells, progenitors and lymphocytes (BMCs), as well as the mobilization of endogenous BMCs with G-CSF (granulocyte colony stimulating factor) significantly improved the recovery of hind limb motor function and sensitivity in rats with a spinal cord compression lesion and significantly increased the spared white matter volume in the center of the lesion. Attention training using EEG biofeedback in patients with brain injury

Celosvětové statistiky jsou pravidelně plněny varovnými čísly, která oznamují, kolik lidí každý rok podlehne následkům traumatu mozku. Díky zlepšující se lékařské péči se zvyšuje počet lidí, kteří přežívají i závažná poranění mozku. Tato poranění však sebou nesou řadu komplikací a to raných i pozdějších. Finanční prostředky vynakládané na léčbu pacientů po poranění mozku jsou značné. Přesná čísla nejsou známa, ale podle Smrčky (2001) v roce 1990 např. v USA dosáhly roční náklady na mozková traumata 25 miliard US dolarů. Tato částka zahrnuje náklady spojené jednak s přímou léčbou pacientů, ale i s následnou rehabilitací, rekvalifikačními kurzy atd. Řada pacientů je díky následkům úrazu v dlouhodobé pracovní neschopnosti nebo trvalé invaliditě. Častými následky po poranění mozku jsou mimo Jlne deficity kognitivních funkcí, zvláště pozornosti. Tato disertační práce se zaměřuje na možnost využití metody EEG biofeedback při tréninku pozornosti u pacientů po poranění mozku, Jednou z terapeutických aplikací metody EEG biofeedback jsou právě poruchy pozornosti. Photodynamic therapy (PDT) is currently a well-established method in this area, but targeting its effect on tumor vasculature has not been considered until recently. In comparison with PDT, vascular-targeted photodynamic therapy (VTP) takes the advantage of a much shorter interval between administration of the photosensitizer (PS) and irradiation (drug-light interval) and thus turning the effect of cytotoxic agents to tumor vasculature. It is intended to disrupt the oxygen and nutrients supply to malignant cells and thus cause their damage and death. It is a modern trend to buy more and more often a variety of herbal food supplements in order to strengthen or restore human health. Many scientists have been researching Myrica rubra plant for its potential therapeutic effects. In our research we used sesquiterpenes isolated from this plant, in order to determine their effects on the activity levels of antioxidant enzymes in a differentiated Caco-2 cell line derived from intestinal epithelium tumor that have maintained characteristics of healthy cells. Caco-2 cells are being used as a basis for testing of substances and their effects on non-cancerous cells. The cells were incubated for 24 and 48 hours with a concentration 25 µg/ml of tested substances. Postprandial thermophily in reptiles

Many ectothermic vertebrates, including some species of reptiles, raise their preferred body temperature after feeding, termed postprandial thermophily. Postprandial thermophily enable the animal to digest its meal at higher rate. However, most studies documenting this phenomenon have relied upon laboratory thermal gradients, with grossly oversimplify an animal's environment and the thermal gradient data may sometimes be misleading. On the other hand, in field or field enclosure, thermoregulation is just one concern among many (such as reproduction, predator avoidance, foraging) and the current biotic and abiotic factors may influence actual thermoregulation behaviour of the specimen. So there is the need for caution in interpreting results. Future research into the thermal ecology of ectotherms should take special care in the design of experiments and methodology. Study animals should be given microhabitats that approximate those they prefer in the wild.

This approach has many advantages compared to traditional ways of drug administration, hence we observe intense research in this area in last decades. The mechanism of action of enhancers is not exactly elucidated yet. They reduce reversibly barrier properties of skin by nonspecific interactions with components of the stratum corneum (SC), i.e. proteins and lipids, or change the partitioning between the SC and the drug. The aim of our work was to determine the cytotoxic effects of selected transdermal permeation enhancers comparing IC50 values of new substances synthesized in our department with clinically used agents. This thesis deals with the business process based transformation of two businesses - Lázně Aurora s.r.o. and Bertiny lázně s.r.o. The outline of a system development analysis is based on a theoretical framework of the business process reengineering and the production management in a contemporary dynamic and turbulent corporate environment. A conversion into a holding arrangment with parent corporation Slatinné lázně Třeboň a.s. and two subsidiaries Lázně Aurora s.r.o. and Bertiny lázně s.r.o. is proposed in relation with restructuring and redesigning all business processes. The outline of new Slatinné lázně Třeboň group based on business process management precedes a detailed analysis of current state and future lookout of spa industry from the point of view of the new business entity. This analysis implies that current management techniques facing the challenging environment cannot guarantee a substantial economic growth of the businesses. Within a whole host of named reasons there are changes related to the transformation of the national health sector, strengthened by demographic changes. Current management practices and enterprise enablers of process management are evaluated through Process and Enterprise Maturity Model (PEMM). Last part of this thesis presents the outline of the new business system development analysis. The goal is to overcome the dynamic environment issues by engagement of business process management and to prepare a production system for serving different segments of clients that are characterized by considerably different needs and requirements. New model of core business processes is proposed in order to meet this goal. It comprises a description of process relations and positioning within the new enterprise structure, including the main reasons of such an alignment.

Mesenchymal stem cells and the possibility of their transdifferentiation into insulin producing cells.

Mesenchymal stem cells (MSCs) have been demonstrated in almost all tissues of the body. Their main source is bone marrow and adipose tissue. These cells are multipotent, e.g. they are capable of differentiating into a variety of cell types. They are able to migrate into damaged tissues. Their other relevant property is a specific suppression of imunity. In the body they serve as precursors for specialized cell types and they also participate in formation of specific tissue microenvironment. Their properties represent a great potential in a wide range of clinical therapies. Besides other possible applications they could be used in the therapy of diabetes mellitus type one. During this disease insulin producing -cells are destroyed. MSCs have been used in experimental in vitro and in vivo studies to differentiate into insulin producing cells. However these cells are not able to produce sufficient amounts of insulin to exclude the supportive administration of exogenous insulin. Therefore there is a need for further research in this field of possible therapy.

Comprehensive characterization of cellular suspensions by means of elastic light scattering

Studies of yeast Saccharomyces cerevisiae - a model eukaryot organism - often require determining the number of cells in a suspension. One of the methods often used for studying cell suspensions is the measuring of elastic scattering of light passing through the suspensions. Usual output of those measurements is the angular dependency of the intensity of scattered light, which bears information about the size of scatterers. This thesis studies time-dependencies of the intensity of scattered light and relationship of their statistical properties to the number of scatterers in a unit volume. For this purpose a simple model was proposed, which describes the connection between this quantity for a monodisperse suspension and the dependency of standard deviation of time-behaviour of the scattered intensity on the mean value of the same time-behaviour. An apparatus for computer controlled measurements of angular and timedependencies of scattered light intensity was completed for carrying out the experiments. Elastic scattering was measured on suspensions of yeast Saccharomyces cerevisiae and a model system (suspensions of 8m polymer particles). DIPLOMA THESIS IS DEVOTED TO THE STUDY OF PREPARATION OF PURE BELITE CLINKER FOR THE POTENTIAL INCREASE OF KINETICS OF THE HYDRATION PROCESS BY CHEMICAL ACTIVATION. THE THESIS OF THIS WORK IS BASED ON RESEARCH FINDINGS CONCERNING BELITE CLINKER AND RESEARCH AT THE INSTITUTE OF THD. THE THESIS AIMS TO DESIGN COMPOSITION OF THE RAW MEAL BURNING BELITE, FOLLOWING MODIFICATION BY SULPHATE AND POTASSIUM CARBONATE IN ORDER TO POTENTIALLY INCREASE THE REACTIVITY OF THE BURNT BELITE CLINKER AND THE LABORATORY FIRING OF PREPARED SAMPLES.
