Evaluation of SDS-PAGE tuber protein profiles for their potential use in potato cultivar characterization

The use of various types of gel electrophoresis for characterization and identifying plant varieties is well estabilished. This diploma work deals with the possibility characterize potato cultivar (Solanum tuberosum L.) by the help of electrophoretic technique SDS {--} PAGE tuber protein profiles. Soluble potato tuber proteins can be classified according to their molecular weight (kDa) into three groups: First, the major tuber protein patatin (37 {--} 44 kDa); second, a group constituted of protease inhibitors (6 {--} 24 kDa); and third group containing all other proteins. First two group manifest genetic variability, which is necessary evalute for its potential using of potato cultivar characterization.
Evaluation of tuber cooking quality in selected table potato cultivars cultivated under organic and conventional crop management

The aim of this diploma thesis was to evaluate the effects of conventional and organic crop management on cooking quality of potatoes. In one-year potato field trials (year 2010) with different crop management (conventional versus organic), 13 table potato genotypes (Adéla, Bionta, Karin, KE 100/10, Laura, Madona, Magda, Princess, Red Anna, Rosara, Solara, Satina,Terka) were used. The cooking quality (i.e. colour of the flesh, consistency, mealiness, moistness, flavor and darkening) was subjectively assessed using sensory analysis according to VORAL (1996) method. Only structure of boiled potato tubers was significantly affected by different crop management (4.8 % from total variability). Genotypically declared cooking types in used potato genotypes were confirmed in both production systems.
Reaction of certified seed and farming seed in selected variety potato

The main aim of this bachelor work was influence of certified seed and farmer seed in selected potato varieties. In 2008 attempts were established with certified seed and farmer seed. The selected varieties were evaluated by the number released plants, production of potatoes, proportion of market tubers and starch kontent in tubers. Comparisons were 4 varieties with different lenit of growing season {--} Magda, Adéla, Ditta, Bionta.
Monitoring of fulfillment of consumption basket in the selected kindergartens

The main aim of the Bachelor thesis in the theoretical part was to outline the issue of boarding in kindergartens, to explain the term ,,consumption basket`` and subsequently to provide analysis of single parts of the consumption basket in regards to its benefits for the organism of a child. In the practical part, the explanation on the principle of calculation of consumption basket is given in the first place. The main aim of the practical part is then to analyse consumption basket in selected kindergartens and carry out, based on the selected criteria of classification, evaluation of fulfillment of consumption basket in the respective kindergartens.
The influence of density growth potatoes on decree tuber

In the potato production type, 380 {--} 400 m above sea level, was studied density 40 000, 45 000, 50 000, 55 000 plants per 1 ha on the yield potato, average number of tubers on the plant, proportion and average weight of tubers. The maximum yield of tubers were recorded at density 45 000 plants per 1 ha (varieties Impala, Adéla, Futura). Density grow it was not influence at proportion market tubers. Highest density growth the meaning reduction average weight of tubers.
Hydrolysis of potatoes

This work compares acid, alkaline and enzymatic hydrolysis of potato starch with simultaneous application of various pressures (0MPa, 0,3MPa and 1MPa). Acid hydrolysis was made by sulphuric acid of intensity 3, 6, 9 and 12 %, alkaline hydrolysis was made by sodium hydroxide of intensity 3, 6, 9 and 12 % and four enzymatic preparations for starch hydrolysis were used for enzymatic hydrolysis. Hydrolysis effect was measured via GE and added by alcohol yield. There was positive influence of increased pressure 0,3MPa and 1Mpa on hydrolysis process visible in this experiment.
