Methodology for experimental study on the release of humic acids from hydrogel application forms.

Hlavnou náplňou bakalárskej práce bolo navrhnúť a optimalizovať metódu experimentálneho štúdia uvoľňovania huminovej látky z gelových matríc, ktoré boli vytvorené pomocou bežných kozmetických a farmaceutických zahusťujúcich látok. Pre štúdium uvoľňovania tejto aktívnej látky bola zvolená difúzna aparatúra vo vertikálnom usporiadaní, tzv. Francova cela. Výluhy gelových vzoriek boli podrobené spektrofotometrickým meraniam. Vzniklé interakcie medzi roztokmi lignohumátu a rôznych typov zahusťujúcich látok boli experimentálne stanovené. Celý proces optimalizácie metódy pre uvoľňovanie huminovej látky z gelových foriem bol diskutovaný.
Preparation of humic hydrogels for appliaction in cosmetics

This bachelor´s thesis is focused on the preparation and characterization of hydrogels containing humic substances and assessment of their potential usefulness in cosmetic practice. Main objective of the work was to develop a literature review focusing on the potential use of humic substances in the cosmetics and pharmaceutical industries. Furthermore, the aim was to prepare model hydrogels addition of soluble humic substances in traditional cosmetic compositions and their characterization by basic methods of analysis parameters. The main investigated parameters were consistency resulting gels (assessed subjectively and subsequently by rheology), their internal pH and dry matter. The experimental results showed that incorporation of humic substances into hydrogels resulting gels acceptable characteristics for further use in cosmetic applications.
The Study of Aggregation of Humic Acids in Different Xenobiotic Environments

Theme of the presented bachelor thesis is the study of the aggregation behavior of the humic acids (HA) in the environment of extraneous substances. As extraneous substances, three cationic surfactants were selected (CTAB, TTAB and Septonex). In the experimental part of the thesis, the humic acids were characterized in detail using different methods (elemental analysis, ash content determination, titration acidity determination). The experimental part of the thesis is dealing with the structure of created HA-surfactant systems. These systems were characterized using the results of the elemental analysis and FT-IR spectroscopy. The key part of the experimental part of the study is represented by the selection of proper methods and by determination of the critical aggregation concentration of the humic acids in the surfactant environment. For these purposes, two key methods were selected in the frame of the presented bachelor thesis: isothermal titration calorimetry and dynamic light scattering. The first method is sensitive owing to the energetic fluctuations, in the case of the dynamic light scattering we are able to detect well the changes in the particle size caused by mutual interactions of particular substances in the system.
Study on structural and thermodynamic aspects of the interactions of humic substances with organic ions.

This bachelor thesis deals with the study of thermodynamic and structural aspects of the interactions of humic acids with organic ions. Methylene blue (dye) was used as a model organic ion. The interactions were studied by isothermal titration calorimetry and dialysis in diffusion cells. The calorimetric experiments showed exothermic nature of the interactions. Furthermore it was proved that increasing concentration of humic acids causes increasing of reaction enthalpy. The dialysis experiments found that increasing concentration of humic acids causes increasing of apparent equilibrium constant. The value of this constant was lower for modified humic acid due to the reduced content of the binding sites.
Application of trihalomethane formation potential to evaluation of drinking water treatment efficiency

The aim of bachelor thesis was to develop a literature search for the potential creation of oftrihalomethanes (THMs FP) as indicators of water quality in surface water treatment. The THM FP test was monitorated to evaluate the effectiveness of current technological methods of treatment of natural water with special consideration of water with increased content of humic substances. The attention was focused on comparison of the THM FP values with the normally applicable water quality parameters; it means the concentration of dissoluble organic carbon (DOC), UV254, CODMn, CODCr. The obtained evidence suggests that THM FP test allows sensitively assess water quality in term of content of chemically and biologically easily biodegradable organic substances. The potential of THM formation has been proved as a valuable tool for quantify non-specific organic contaminants in treated water, which can usefully complete previously used criteria for assessing quality of treated water.
Bond strength of metal ions in humic gels

This thesis deals with the study of bond strength of Cu2+ ions in humic gels. Two methods of extraction were used during the study, sequential and fractional extractions. During the fractional extraction leaching of copper proceeds in single agents, whereas during the sequential extraction leaching proceeds gradually in all extraction agents. Destilled water, 1M MgCl2, 1M HCl and 0,025M NH4EDTA were applied like extraction reagents. Amounts of Cu2+ ions in leaches after extraction were measured by UV-VIS spectrometry. By using extractions was obtained the fractionation of Cu2+ ions according to bond strength in humic gel.
Study on phase transitions in humic substances

The work is aimed to examine the potential applications of temperature modulated differential scanning calorimetry in study on phase transitions in humic acids. Experiments will deal with both humic standards and the solid rests after sequential extraction of specific molecular fractions to reveal their role in secondary structure of humic acids.
Influence of humic substance HS 1500 on tolerance of Sterlet to nitrite

The aim of the thesis was to assess the effect of humic substance HS 1500 on the tolerance of Sterlet (Acipenser ruthenus) to nitrite. Preparation Huminfeed was used as a source of substance HS 1500. Tolerance of Sterlet to nitrite was assessed on the basis of the results of acute toxicity tests and the results of haematological and biochemical blood examination of fish that were exposed to increased concentrations of nitrite in the presence and absence of preparation Huminfeed.
Influence of selected factors on reproduction of fish \kur{Betta splendens}

The Siamese fighting fish (Betta splendens; order Perciformes, family Osphronemidae) is a fish occurring in the area of south east Asia. Above branchial organ (so-called labyrinth) allowes it to breath an air oxygen. It is a territorial sort of fish, males are brawling to each other. In a breeding fish take alive or frozen food and also artificial feed. The fish is bubblenest, spawns are put into a foam nest, built by a male, that takes care about the posterity for some time. The task of the work was to verify the influence of chosen feeds sorts and concentration of humic substances in water for a success in reproduction of Betta splendens fish. The experiment was performed in an aquarium room at institute of biological disciplines AF USB. The experimental work started on 12. 12. 2011 and was finished on 25. 2. 2013. Tested fish were by particular couples put into tanks, at which there were breeding afterwards. It was evaluated the frequency of spawns and the number of embryos. From the result of the work there is obvious that the feeding by a beef heart was preferable for fish´s fertility in a monitored time area about approximatelly 75% in comparison to feeding by a chicken breasts muscles. The presence, or more precisely the concentration of humic substances obviously did not affected the fertility. All fish´s groups embodied higher grade of fertility at the beginning of the monitored time area (probably higher fertility at younger - more vital fish). Siamese fighting fish is very demanded fish at aquarists, this is why is very important to evaluate certain parameters of breeding and rearing of these fish.
