Analysis of active phenolics in some cereal products

Presented diploma thesis was focused on study of phenolic substances in various cereal products. Flours, brans and germs from various cereals, flakes and extruded cereals were chosen as samples. Phenolic substances were analyzed by spectrophotometry, RP-HPLC/UV-VIS and RP-HPLC/PDA/ESI-MS. Studies of saccharide content which were analyzed by spectrophotometry and HPLC/RI was incorporated to this thesis. Differences in the content of phenolic substances released from glycosides after acid hydrolysis of samples and differences in saccharide content determined after acid and enzymatic hydrolysis of samples were investigated. Simple sensory analysis of extruded cereal products of different flavour (cocoa, honey, chocolate) was carried out too. In analyzed samples the highest content of total phenolics and total flavonoids was measured in wheat germs. Especially in wheat germs and also in buckwheat and oat flakes, maize brans and cereals products Teddy (cereal balls with cocoa) and Ferda (cereal grains with honey) high levels of individual catechins, phenolic acids and flavonoids were determined too. The quantity of phenolic substances increased for several times in samples processed by acid hydrolysis in comparison to non-hydrolyzed samples. The high content of total saccharides, reduced saccharides and individual mono- and disaccharides was found mainly in extruded products. Concentration of monosaccharides increased in acid hydrolyzed samples in spite of disaccharides concentration that was higher only in some of hydrolyzed samples. In several cases final disaccharide concentrations were lower in hydrolyzed samples than in non-hydrolyzed ones.
Evaluation of selected dietary supplements - climacterium

A dietary supplement is a product that contains nutrients (e.g. vitamins, minerals, herbs, amino acids or fatty acids) intended to supplement the diet. The aim of the diploma thesis was to suggest a prescription for preparation of three types of dietary supplements which are used to reduce of climacteric symptoms, their production and determination of energetic (nutrition) value on account of detected content of proteins, fats and saccharides in dietary supplements. A part of the diploma thesis was to suggest suitable wrapping and price of prepared dietary supplements. The three dietary supplements were used for analyses, namely capsules, herb syrup and herb drops. The content of proteins was determined by Kjeldahl´s method, total fats were determined by Soxhlet extraction and saccharides were determined by titration with potassium permanganate (Bertrand´s method). Dietary supplements in form of capsules were particular analysed on content of heavy metals (lead, cadmium and arsenal). In herbal drops was in addition determined alcohol content (ethanol). There was from the measured data found out, dietary supplement in form of capsules was chosen as the product with the lowest energetic (nutrition) value.
Analysis of main wine saccharides

This diploma thesis deals with determination of carbohydrates in wine. Theoretical part attends to production of wine from grape to treatment and training of wine. It refers saccharides in wine, especially glucose and fructose. The study provides an overview of the available sources concerning the possibilities of determination of carbohydrates in wine by chromatographic methods and Skalar automated chemistry analyser. The study shortly refers simple analytical methods for determination of wine saccharides. The experimental part based on this search deals with analysis of saccharides by high performance liquid chromatography with refractive index detector and UV detector on aminoalkyl column and determination of reducting sugars by Skalar automated chemistry analyser. Finally, the results of these methods were compared.
Dietary supplements in sport nutrition and possibilities of monitoring their effect on efficiency of sportsman.

This work deals with dietary supplements intake and its impact on the performance rise and muscle mass growth of bodybuilders. Twenty-five students (bodybuilders) were chosen for the purpose of this survey; they were divided into four groups according to the dosage of creatine and other dietary supplements (proteins and maltodextrine). Changes of the measured parameters (weight; bustline; weistline; limb girth; body lipid content; creatine and urea content in the urine) were compared during the survey. The tested dietary supplements impact on the muscle mass growth was evaluated on the basis of the acquired results.
Characterization of cereal products with fruit component for baby nutrition

Present bachelor thesis deals with the characterization of milk cereal products intended for baby food. The theoretical part focuses on the morphology of cereal grains, fortification, an overwiev of supplementary components used to fortification and their properties. For the practical part eight kinds of cereal dairy products of three different producers were chosen. In these products total polyphenols, total flavonoids, reducing and total saccharides and proteins were analyzed. The highest level of polyphenols was measured in product Sunar multicereal. The highest amount of flavonoids (more than 50 % of total phenolics) was found in Nestle fruit product. The highest level of total and reducing saccharides contained Nestlé honey flavour. High amount of proteins was found in products Sunar with multicereal flavour and pears with biscuits. Using HPLC/UV-VIS analysis vitamins C and E were determined. The highest content of vitamin C contained Nestlé with cookie lavor, while high vitamin E level was found in Sunar pear flavor and biscuits. Using HPLC/RI individual saccharides were examined. In all products high amount of lactose was found, mainly in the products of Sunar. Nestlé products contained mostly glukose, while in products Hami sucrose was detected as the most abundant saccharide.
Characterization of waste rice substrates and their potential utilization by microorganisms

The aim of this bachelor thesis was to characterize some cereal substrates. Based on the results of group parameters the most suitable substrate for cultivation of model industrial microorganisms was proposed. Analysis of total phenolics, reducing sugars and total sugars was performed spectrophotometrically. Using high-performance liquid chromatography with refractometry index detection content of individual mono-and disaccharides was measured. Saccharides were extracted from cereal substrates by water and then acid hydrolysis was applied. Two rice substrates were then used as nutrition sources for cultivation of two model industrial microorganisms: yeast Rhodotorula glutinis and bacterium Bacillus subtilis. Before use to production media substrates were processed by chemical and enzyme hydrolysis and analyzed. Production of microbial biomass in media with modified waste substrates was evaluated.
Active substances in cereal products for baby food

Present bachelor thesis is focused on the study of antioxidant substances in cereal products for baby food. The theoretical part is aimed at describing the anatomical structure and chemical composition of cereal grain, characterization of antioxidants with the focus on natural antioxidants, the technology and nutritional parameters of cereal products. The experimental part deals with the content of phenolic compounds, flavonoids, and the total and reducing sugars in the total of 12 kinds of cereal products with different flavour – 5 types of oak pap, 3 types of fiber pap and 4 corn paps. The work includes the analysis of ascorbic acid by titration method and the analysis of tocopherol, carotenoids and catechins by high performance liquid chromatography as well. The highest content of total phenolics and flavonoids was found in corn pap with raspberries and also in cereal pap with chocolate. High levels of vitamins E and C were found predominantly in flavoured fiber pap.
The dietary accessories to support of overweight reduction

This bachelor´s thesis deals with the possibility of using alternative sweeteners to reduce overweight and treat type 2 diabetes. In the experimental part of this thesis was verified the minimal effect of reduced calorie sweetener Extra-Linie from Favea Ltd. including erythritol on blood glucose concentration.
The lipid quality of our economically significant fish species

The level of fish consumption in the Czech Republic is very low. One of the possible factors influencing this situation is low awareness of the customer or consumer. The thesis "The Lipid Quality of our economically significant fish species" works with the list of nineteen economically most significant fish species not only including fish from the semi-intensive and intensive breeding, but also such species that are interesting for the consumption from the sport fishing point of view. The samples taken from particular fish were primarily used for the analysis of the fat content and fatty acid composition. The analysis established very interesting results in this field and created basis for further analysis for establishing atherogenic and thrombogenic index or for formation of nutritive values mentioned as "Big 7" (fat content and the ratio of fatty acid out of it, sacharide content and the sugar ratio in it, protein content, ash content and energy value) by particular fish species according to the regulation ES 1169/2011. The results of this work confirmed that the fat content and its composition are highly influenced by the type of ingested nourishment. The fish bred in semi-intensive way with the highest fat content are Common carp (7,62 ? 2,9 %) and Silver carp (6,87 ? 1,37 %). On the contrary the predatory fish evinced the fat content usually within 1%. The fish from extensive breeding or the types caught in sport fishing are in the fat con-tent more balanced. The fattest fish in this group is Silver Bream (5,52 ? 0,44 %) and the lowest content evinced Goldfish (1,14 ? 0,38 %). Also the species living in the aquaculture and living in the free waters as well were compared (Common carp, Rainbow trout, Peled, Tench, Wels catfish). The fatty acid composition significantly depends on the fat content. The predatory fish contain 195 - 219 mg×100 g-1, the most (p0,05) was found by the peled from the extensive breed (1048 mg×100 g-1 EPA + DHA). The participation of SFA by the most of the fish was under 30%, the content of MUFA was the highest by the Common carp (52,71 ? 7,75 %), the lowest by the European perch (18,83 ? 1,12 %). Pike, Zander and European perch contain relatively the most of PUFA, this is over 50%. All the fish species (Nile tilapia excluded) show in this study the values of atherogenic index below 0,5 and thrombogenic index below 0,4. The total energetic value was counted from the results of nutritional values presented in the "Big 7" list. The higher energetic value is directly proportional to the increasing fat content. The fish species included in this thesis show the total energetic value in the range of 500 - 900 kJ×100 g-1. The results of this study bring complete, laboratory proven information about the nutrition composition of our fish. This data can be appli-cable not only to increase the knowledge of the customers but mainly to be used by breeders and manufacturers of fish as an implement of fulfilling the duty of stating the nutrition facts on their products according to ES 1169/2011 on the providing infor-mation about the eatables to the consumers.
Vliv konzumace vybraných potravin sacharidové povahy na hladinu krevní glukózy

This thesis deals with the measurement of blood glucose levels after consumption of selected foods (glucose solution, milk chocolate, rice sandwiches with corn, fruit snacks with apricots and broken buckwheat multigrain bread) at regular intervals. The first part of this thesis is pursued to the literature review, which provides a comprehensive overview of the metabolism of carbohydrates, fats and proteins. It also discusses the differences in the carbohydrate metabolism of healthy people with diabetes type 1 and 2 It acquaints the reader with the concepts of blood glucose, glycemic index and glucose-tolerance test. The practical part includes the measurement of glucose in capillary blood and questionnaire survey concerning state of health, eating habits and the disease incidence in a selected group of volunteers. The experiment involved 12 students of Mendel University in Brno. Their blood glucose levels were measured at regular intervals (0, 30, 60, 90 and 120 minutes) after ingestion of the food. The glucose was chosen as a standard. From the measured values were calculated glycemic indices, which represented 100 for glucose, milk chocolate 40+-17,7, sandwiches, rice with corn 76+-25,8, fruit snacks with apricots 49+-17,9 and multigrain bread buckwheat 68+-21,3. The results were statistically processed and the effect of individual foods tested on blood glucose was evaluated.
