Thermophilic microorganisms application to biodegradation
Varmužová, Tamara ; Márová, Ivana (referee) ; Babák, Libor (advisor)
This thesis is focused on study of biodegradability of modified polyurethane elastomeric films in synthetic medium with minerals and vitamins on tempered shaker by mixed thermophilic aerobic bacterial culture Bacillus and Thermus genera. In most cases addition of all used fillers (carboxymethyl cellulose, hydroxyethyl cellulose, acetylated cellulose, acetylated starch and glutein) led to increased biodegradability of elastomeric films with modifying agent in comparison with elastomeric films without modifying agent (referential). The growth of cultures was strongly increased in presence of elastomeric films modified by 10 % acetylated cellulose and 10 % carboxymethyl cellulose. Elastomeric film biodegradation mechanism included probably two stages: abiotic destruction of elastomeric films and consequent utilization of degradation products by bacterial culture.
Application of thermophilic Culture for Organic Compounds Biodegradation
Piechová, Jana ; Burdychová, Radka (referee) ; Babák, Libor (advisor)
The aim of this work is focused on biodegradability study of modified polyurethane materials (foams and foils) in glukose medium and biodegradation of whey. The biodegradability was tested with using mixed thermophilic aerobic bacterial culture of the Bacillus and Thermus genera. The cultivations were practised in the fermentor and in Erlenmayer flasks in a heated shaker. Maximal reduction of COD and culture growth has been achieved by biodegradation of polyuretane in the fermentor. Main effect of degradation was abiotic destruction of PUR materials and consequent utilization of degradation products by bacterial culture. These tests indicated that the best modification agent for biodegradation was 10% carboxymethyl cellulose. The polyurethane foam was more decomposed then the polyurethane foil. Biodegradation of whey without proteins in the fermentor was detected decrease of CHSK and lactose concentration about 50 % in 20th hour. Optimal conditions for whey biodegradation with the thermophilic bacterial culture were pH 6,5 and temperature 60 °C.
Biotransformation of nitro aromates
Tomeček, Jaromír ; Bednařík, Karel (referee) ; Friedl, Zdeněk (advisor)
Contamination by nitroaromatic compounds, such as explosives, pesticides or other nitroaromatic compounds, is worldwide environmental problem. This text focuses on contamination by compounds from military and ammunition industry, most especially 2,4,6-trinitrotoluene (TNT), 1,3,5-trinitro-1,3,5-triazinane (RDX) and 1,3,5,7-tetranitro-1,3,5,7-tetrazocane (HMX). First goal of this work is to provide the reader with a aerobic biodegradation and transformation processes of nitroaromatic compounds named above. The second goal is to compare the toxicity of initial nitroaromates and their transformation intermediates.
Modification of polyurethane by biodegradable poly(hydroxybutyrate)

Degradation possibilities of synthetic macromolecular materials centred on biodegradations
Zichová, Miroslava ; Ondruška, Vladimír (referee) ; Babák, Libor (advisor)
This work deals with the degradation possibilities of synthetic macromolecular materials. Mechanical, chemical and chemo-physical degradation is described, but the work is focused on biodegradation. Polymer biodegradation is its decomposition by microorganisms. The aim of the work is to show the advantages of biodegradation and to arrange the knowledge acquired about the biodegradation of particular macromolecular materials. There is resumed the information about the degradation of polyurethanes as an exception among synthetic polymers, which are mostly resistant to biodegradation. Attention is also paid to biodegradable polymers.
Composting of urban green areas in urban compost facility in Březnice
BARTUNĚK, David
This thesis deals with the use of municipal biowaste, which processes compost facility in the city Březnice. In this work are dealt optimal conditions for composting, composting grass biomass then composting technology and at the end concept of composting in the city Březnice with a total processing technology and equipment. The aim of my thesis was to outline the composting and propose optimal solutions for the construction of a composting facility. Part of this work is also an experiment with ion exchange capacity, which proves the importance of following the correct composting process for the production of quality compost. The contribution of this work rests primarily as such its integrity and organization. It is possible to find there the right parameters for creating the right conditions of the composting process, including the elements necessary for actual registration of compost.
ENDOCRINE DISUPTIVE CHEMICALS: MECHANISMS OF WHITE ROT FUNGI BIODEGRADATION
Cajthaml, Tomáš ; Křesinová, Zdena ; Čvančarová, Monika ; Maternová, H. ; Filipová, A.
The main aim of this study was to elucidate mechanisms involved in EE2 and other EDs compounds degradation by white rot fungi. Results showed simultaneous involvement of ligninolytic enzymes and cytochrome P-450 in biodegradation. The present results indicate the advantage of utilization of WRF cultures for organopollutants degradation.
Functional thin films for applications using advanced oxidation processes
ŠRAM, Vlastimil
This diploma thesis aims to optimalization the process of magnetron sputtering and creating of thin layers for use in advanced oxidation processes. During the work was created range of TiOx layers. For this process was used physical method of sputtering called PVD. The photocatalytic activity of the deposited films was tested by degradation of organic dyes Acid Orange 7. Furthermore, the layer was analyzed on surface morphology (SEM) and the layer thickness (profilometry). Study of created layers was focused on the link between the characteristics of each layer, deposition parameters and photocatalysis properties. Based on these results, the layers were applied in a system using AOP for the decomposition of organic substances. The first chapter is devoted to a summary of existing knowledge of photocatalysis and its principles. Another chapter is devoted to the theory and methods of applying thin layers and summary of knowledge of the low-pressure discharges. In the exprimental section there are described various components of the apparatus. Furthermore, the experimental part of the work focuses on the analysis of the optimization process of applying thin layers on titanium oxide. The last chapter of the thesis contains the results of the experiments on the basis of is designed another research progress of this issue.
