Flodrová, Dana ; Toporová, L. ; Macejová, D. ; Laštovičková, Markéta ; Brtko, J. ; Bobálová, Janette
Bottom-up proteomic approach was used for detailed characterization of proteins from\ntwo human tumour cell lines representing major clinically different types of breast\ncancer. The aim was to show the differences between them on proteomic level. Here\nwe present almost 100 unequivocally identified proteins out of which 60 were mutually\ndifferently expressed for MDA-MB-231 and MCF-7. Some well-known breast cancer\nmarkers like annexins A1, A2 and vimentin were found in the MDA-MB-231 cell line.\nOn the other hand, MCF-7 cells were found to be positive for cytokeratins and keratins\nand thus we were able to distinguish both cell lines sufficiently.

Comfort of mothers during childbrith
HANZALOVÁ, Martina
This thesis for a bachelor's degree is dealing with the comforts of expectant mother in the process of delivery. A theoretical part is divided into three main chapters. The first chapter covers the delivery description and its classification. The second chapter deals with an inclusive nursing care for a mother-to-be in the process of delivery and a midwife's care during every particular labour phase. The last chapter describes all aspects which affect the delivery process. The exploratory part of this thesis is focused on the contentment of women in labour with the offer of comforts during their delivery. I have set two aims. The first aim was focused on the women awareness of pharmacological and non-pharmacological techniques which cope with the pain in the process of delivery. The second aim was focused on the women contentment with comfort offer in the labour ward and the treatment of nursing staff. I have chosen three research questions: Which possibilities of pharmacological and non-pharmacological techniques do the women know and use to ease the pain in the process of delivery? Which relaxation aids have been offered in the process of delivery? To what extent the woman has been satisfied with the ambience and the nursing staff treatment? I chose the method of qualitative research. I carried on a survey to get the detailed and comprehensive information. The whole research was transferred to two main categories and its subcategories. During investigations it emerged that most respondents found the information on the Internet, in professional literature, antenatal courses etc. Hydroanalgesia appeared to be the most known alternative to ease the pain in the form of shower or bath. As far as pharmacological possibility is concerned the respondents mentioned only epidural analgesia. The comforts category describes general comforts which the midwife offers to the expectant mothers in the process of delivery. Antenatal preparation was given to most respondents with the previous consent and they were offered drinks in the labour ward. Most respondents were also offered non-pharmacological techniques to ease the pain such as relief positions, gymnastics ball, massage, hydroanalgesia etc. No respondent was offered epidural analgesia, however, one respondent was relieved with the help of pharmacotherapy. The thesis also deals with the people present in childbirth and the influence of ambience. It was shown that most respondents had no reservations. The category which assesses the women contentment with the midwives attitude towards expectant mothers shows that most women were satisfied. The midwives were not only mental support but also they took care about their comfort and informed about the process of delivery. There was only one case when the woman was not satisfied and she would appreciate to get more information from the midwives. Unprofessional attitude of the midwives has an influence on the respondent assessment. The research shows that public health needs not only routine work but mainly human attitude and communication. Midwives should behave sympathetically, be assertive towards the women in labour and relieve the pain with the help of all available techniques in such a demanding situation as delivery is.

Eating habits of patients with metabolic syndrome
NOVÁČKOVÁ, Anna
Metabolic syndrome (syndrome X, Reaven's syndrome) is frequently occurring disease in Czech Republic. The diagnosis often detected only separated parts of metabolic syndrome which are usually not connected with complex of MS. Medical conditions co-occurred within this syndrome are following: elevated fasting plasma glucose (insulinoresistance), abdominal (central) obesity (women waistline > 88 cm, men waistline > 102 cm), elevated blood pressure, high serum triglycerides (> 1,7 mmol/l), and low high-density cholesterol (HDL) levels (women 1,25 mmol/l, men 1,0 mmol/l). The incidence of MS can be divided into the prevalence of MS itself (this correspond with 30%) and the prevalence of each individual part of MS which correspond with 80% population. The most frequently reasons of MS are so called environmental impacts as e.g. central obesity, insulinoresustance syndrome, fatty liver, systemic inflammation, intestinal alterations, hypertension, hypertriglyceridemia, foetal malnutrition, low basal energetic outcome, sleep apnoea syndrome, changes of gut flora, hyperglycaemia and cumulation of visceral fat. This bachelor´s work is concerned with eating habits, life style and other factors which could be reason of described metabolic syndrome. The aim of this study is evaluation of eating habits and life style of patients suffered from MS before syndrome outbreak, as well as participation of obesity in MS outbreak assessment. Two explorative questions were formulated on the basis of described goals: First research question: Did the MS patients follow principles of healthy eating before syndrome outbreak? Second research question: Does obesity have a significant effect on MS outbreak? Definition and characterization of MS in relation to diet is described in theoretical part of my work as well as explanation for reasons of MS outbreak, descriptions of each separated diseases of MS complex and their treatment. Metabolic syndrome in connection with obesity and its classification into each level is defined inthis part. Obesity treatment together with prevention and treatment of MS are suggested in terminal part of this section. In practical part of work, the results of own research are presented. This section is focused on: 5. evaluation of eating habits before MS outbreak; 6. obesity incidence in MS patients; 7. patient´s life style; 8. the presence of individual diseases participate in MS. Research part of my bachelor´s work is prepared with using of quantitative assessment, which was done by questionnaire investigation in southbohemian and westbohemian region. Research population consists of thirty gender-nonspecific and age various patients diagnosed as having MS. The research questions were focused on mapping of respondent´s eating habits, life style (smoking, movement activity, daily eating portions and breakfast regularity), previous and current illnesses as well as diseases of respondent´s relations. The aim of described investigation was evaluation of obesity, life style and diet participations in MS outbreak. The results did not prove observance of healthy eating principles in MS patients before and after syndrome outbreak, while most of respondents were overweight or obesity. It can be assumed that genetic predispositions play role in MS outbreak. Additionally, very interesting is detected occurrence of MS in many individuals younger then forty years old. The present work extended the knowledge about risk factors of MS outbreak together with eating habits, obesity and MS outbreak connections. When reasons of MS seem to be more clear, it is easier to establish new more effective prevention. On the basis of the results, it can be arranged diet suggestions useable as effective prevention of MS outbreak.

The Tax Assignation and Theater
Gerneš, David ; POKORNÝ, Jiří (advisor) ; VÁVROVÁ, Hana (referee)
Bachelor thesis aims based on an analysis support system theaters in the Czech Republic, the analysis of the tax system in the Czech Republic and analysis of tax assignations in the Slovak Republic, to investigate whether for the Czech theater network, this measure was beneficial and whether it would be worth it to deal with its introduction into practice. The thesis also examines the impact of the introduction of the assignation of income tax on Czech public budgets and polls. Within the survey sent to 120 respondents who were asked about their relationship to culture and whether they would be introducing a possibility for assignations and what area non-profit sector would contribute. On the basis of these steps is then designed a system of assignations suitable for Czech Republic.

Paramedic knowledge of first aid overdose legal recreational drugs
VAŇATA, David
In this bachelors thesis topic: Rescue worker knowledge of first aid for overdosed people on legal recreational drugs . I decided to define the theoretical part of a group of substances that come under legal recreational drugs . I also characterised the history and composition of those drugs , their effects on the human body , signs of intoxication and procedure of a rescure worker and application of first aid. The plot of this work, is to point out the knowledge of a rescue worker in providing first aid to patients who used and are suffering from poisoning their body from any legal drug. I am also interested in whether this procedure theory on it, is consistent to the latest knowledge and practices of a rescue workers in their field. In the practical part of my bachelor thesis, I decided to focus on a group of rescue workers of South Bohemia, with whom I have met during my practical training . As my thesis sample , I used a quantitative method that is based on 50 anonymously answered question forms . I have not specified my hypotheses. I expect this work to be help for current and future rescue workers in their studies , and also, I hope in deeper acknowledgement, improvement of working procedure, and especially, bigger attention to the substances that are tolerated by our society, although very dangerous.

Natural houses in gardens
Formánková, Kristýna ; Kunt, Miroslav (advisor)
Thesis discusses natural houses which are supposed to be part of our lives in the future. These houses may be for someone and they might prefer smarter building with non-recyclable materials when choosing a new housing. But it is necessary to think about our future and sustainable development. We also need to tell ourselves what is really a priority for our planet. There comes a time when they will prefer building materials which can be used somehow after some time or be recycled. This is related to the design of the garden. It is designed to fulfil an ornamental, economic and social function. The garden is an integral part of each house, therefore it is important for each element used to be creating the garden as a linked harmonious whole. The study deals with the concept of a natural houses home in accordance with the maximum use of the garden.

Effect of snowpack on runoff generation during rain on snow event.
Juras, Roman ; Máca, Petr (advisor) ; Ladislav , Ladislav (referee)
During a winter season, when snow covers the watershed, the frequency of rain-on-snow (ROS) events is still raising. ROS can cause severe natural hazards like floods or wet avalanches. Prediction of ROS effects is linked to better understanding of snowpack runoff dynamics and its composition. Deploying rainfall simulation together with hydrological tracers was tested as a convenient tool for this purpose. Overall 18 sprinkling experiments were conducted on snow featuring different initial conditions in mountainous regions over middle and western Europe. Dye tracer brilliant blue (FCF) was used for flow regime determination, because it enables to visualise preferential paths and layers interface. Snowpack runoff composition was assessed by hydrograph separation method, which provided appropriate results with acceptable uncertainty. It was not possible to use concurrently these two techniques because of technical reasons, however it would extend our gained knowledge. Snowmelt water amount in the snowpack runoff was estimated by energy balance (EB) equation, which is very efficient but quality inputs demanding. This was also the reason, why EB was deployed within only single experiment. Timing of snowpack runoff onset decrease mainly with the rain intensity. Initial snowpack properties like bulk density or wetness are less important for time of runoff generation compared to the rain intensity. On the other het when same rain intensity was applied, non-ripe snowpack featuring less bulk density created runoff faster than the ripe snowpack featuring higher bulk density. Snowpack runoff magnitude mainly depends on the snowpack initial saturation. Ripe snowpack with higher saturation enabled to generate higher cumulative runoff where contributed by max 50 %. In contrary, rainwater travelled through the non-ripe snowpack relatively fast and contributed runoff by approx. 80 %. Runoff prediction was tested by deploying Richards equation included in SNOWPACK model. The model was modified using a dual-domain approach to better simulate snowpack runoff under preferential flow conditions. Presented approach demonstrated an improvement in all simulated aspects compared to the more traditional method when only matrix flow is considered.

Design of Experiment for Non-Stationary Processes of Production
Jadrná, Monika ; Macák, Tomáš (advisor)
The doctoral thesis is concerned with the services sector and the area of mass production. Particularly, the optimization of the product portfolio of the travel agency and the optimization of production rounds of ammunition. The theoretical part deals with the current overview of discussed topic. Further, the terminology and methods of the decision-making process are defined to support decision making. The theoretical basis of research focused on the choice of appropriate input variables in the area of services, and on the choice of a particular material option in the production area and appropriate equipment for the production. The theoretical part forms the basis for the practical part of the thesis. For the doctoral thesis was chosen an enterprise operating in the defined sector. Product portfolio for the services sector is optimised using Fuzzy logic and Fuzzy sets so that the enterprise can maintain its competitiveness in todays highly ambitious market. Product portfolio for manufacture is optimised for achieving desired properties of the product. The main aim of the thesis is to propose a new methodological approach for the management of selected business processes in their nonstationary time course. The aim of the practical implementation is to verify the functionality of the proposed methodological approach, both in the area of services and in the field of mass production.

Non-invasive methods for sex and steroid homones determination in gibbons of the genus Nomascus
Bolechová, Petra ; Bartoš, Luděk (advisor) ; Pavel, Pavel (referee)
The gibbon primates of the family Nomascus are classified as critically endangered species, and, to date, basic understanding and information about their biology is missing. With regard to the status of these animals in the wild and captive populations in zoos, being familiar with their reproduction, may improve captive breeding programs. Data collection in this study was to be carried out by the practical use of non-invasive methods (polymerase chain reaction for DNA extraction and enzyme immunoassays), using faecal samples for analysis. The first method, polymerase chain reaction, was used for sex determination in juveniles, because of their coat colour and visual similarity of secondary sex characteristic in both sexes; it is not possible to determinate sex without handling the animal. Another main purpose of this study is to try and answer the hypothesis regarding the ovarian cycle of females, factors influencing their hormone concentration and also the onset of sexual maturity in females and the timing of their fur colour change. Hypotheses were checked by monitoring the concentration of progesterone and oestrogen faecal metabolites and by evaluation of the composition of breeding groups of gibbons with the ZOO influence. During a four year period (from 2010 till 2014), there were a total of 51 animals analysed from 16 different zoos with a faecal sample count totalling 1618 samples. The results confirmed the use of noninvasive methods for sex determination, and thus ensuring the maximum welfare standards. Endocrinological analysis confirmed the hypothesis of the influence of the environment (ZOO) to the hormone concentrations and female´s pregnancy effect in conjunction with her age on the final results of the male - father hormone concentrations. The initial information in this study is the confirmation of the onset of ovarian cycle in young females without connection to their fur colour change and a significant factor of a mother´s impact and her territoriality. This study is the first to present a long term monitoring of ovarian cycles in females and hormone concentrations of other individuals, both male and female, and from various age groups. The results allow us to understand the possible impact of zoo environments on the reproductive status of these gibbons and contribute to the general improvement of breeding management.
