Role of Psb28 proteins in the biogenesis of the Photosystem II complex in the cyanobacterium Synechocystis sp. PCC 6803

The thesis focuses on the role of Psb28 proteins, namely the Psb28-1 and its homolog Psb28-2, in the biogenesis of the Photosystem II complex (PSII) in the cyanobacterium Synechocystis PCC 6803. The aims of this work were to localize the proteins within the cells, and to determine their function. A fraction of both Psb28 proteins was identified in the monomeric PSII core complexes but most proteins were found in the unassembled protein fraction associated with thylakoid membranes. Psb28-1 was mostly detected as a dimer while Psb28-2 as a monomer. Psb28-1 also differed from Psb28-2 by its higher affinity to the PSII core complex lacking CP43 antenna. Characterization of Psb28-less mutants suggested regulatory function of the proteins in PSII biogenesis in connection with chlorophyll biosynthetic pathway. Analysis of preparations isolated using FLAG-tagged versions of Psb28 proteins showed their association with Photosystem II - Photosystem I supercomplexes, especially under increased irradiance, and supported a role of Photosystem I in the PSII biogenesis.

Development of electronisation of local administration of Zábřežsko micro region in relation with e-government

Thesis "Development of an electronisation of local administration of Zábřežsko micro region in relation with eGovernment"examines the issues of the development of electronisation of autonomy in relation to legislation and eGovernment. The topic of the public administration has been increasingly discussed in recent years not only in the context of increasing demands for processing speed and information, but also to facilitate transparency and sub-processes in public administration. Theoretical part describes the role of eGovernment in the public administration, the process of electronisation of public administration and individual services of eGovernment (Czech POINT, data boxes, basic registers, communication infrastructure of public administration), which are being in place with municipalities. The practical part evaluates the situation of software and hardware equipment, legislatively bound on a contract software and service of eGovernment on a sample of villages Zábřežsko. On the basis of the results of the questionnaire researches are determinated recommendations for sample of 20 micro region Zábřežsko.

Evaluation of the European Cohesion Policy in Selected Region

The diploma's thesis is aiming to evaluate European policy of social cohesion and its growth options in the selected region. The evaluation is connected to the Elbe cycle route and it is based on analyses. The theoretical part of the thesis introduces the subject of European policy of social cohesion while defining its legislation and framework of the legislation. It also defines the embodiment of regional politics within legislation of the local government i.e. the Czech Republic. Analyses of the development of the selected project i.e. the Elbe cycle route and its impact on Hradec Kralove district will be dealt with in the practical part of the thesis. Business results of entities participating on the Elbe cycle route project will be gathered and used in order to evaluate selected indicators. Final assessment will be based on benchmarking. Method of comparison, analyses and syntheses will be used to evaluate gathered data. Applicable data will be gathered by research of related literature. Information will be also provided by the executive entities connected to the project, regional websites focussed on cycle tourism and by the town of Smiřice. Data gathered by the entities that cooperate on supervision of the Elbe cycle route project will be used for the statistic assessment.

Vote and Voting Behaviour of Citizen of the Town Lanškroun

This graduation thesis Vote and Voting Behaviour of Citizen of the Town Lanškroun deals with vote and voting behaviour of inhabitants, who have permanent residence in the town Lanškroun. In thesis the returns of particular elections to municipal government of the town are characterized from years 2002, 2006, 2010 and 2014. Subsequently the thesis is focused on the voting behaviour of the population. Informations about voting behaviour of citizen were obtained from performed questionnaire research. The thesis is divided into two parts. The first part of graduation thesis is theoretical, which defines the basic concepts related to the topic of elections. It focuses mainly on concepts such as local democracy, local politics, vote and voting behaviour. The practical part focuses on the characteristics of the town Lanškroun, analyzes individual municipal vote and on the basis of results of the questionnaire results explains the voting behaviour of the population of Lanškroun.

Influence of meadow management on the migration behavior of selected species of butterflies

Migrations are the key process and one of the basic biological features, which allow survival of endangered species in fragmented landscape. In the last decades, agriculture intensification, industry and urbanization caused that number of natural butterfly habitats has significantly decreased. The localities originally suitable for butterflies are currnetly converted to farmland or building plots. This PhD thesis aimed to find a compromise solution of meadow management, which should allow a commercial use and concurrently do not jeopardize the continued existence of endangered or threatened species. The main idea of this study was in the intentional emigration causing in the middle of flight period, which should lead into resettlement of the population to another (potentially more suitable) locality. Possibility of aimed manipulation with butterfly emigrations would be considered as a major breakthrough in their protection. The investigated species Phengaris nausithous (Bergsträsser, 1779) and Phengaris teleius (Bergsträsser, 1779) are both considered to be flagship species for protection of European butterflies at open meadows. This study is based on research, which was carried out in 2013-2015 at 17 patch in localities Dolní Labe, Czech Republic. In above mentioned years, one selected meadow was mowed in the middle of flight season and the emigrations were then monitored using mark-release-recapture method. Results of population and migration characteristics were calculated in MARK 8.1 software. Subsequently, the effect of mowing on emigration (comparison of emigrations from the meadow before and after mowing) was evaluated using Statistica 13. The hypotheses of this study were based on the assumption, that the loss of resources after well-timed mowing should increase the activity of adult butterflies. During the monitoring, number of emigrations was increased only in flight season 2015. Unfortunately, the statistical evaluation did not show significant effect of mowing on migrations. On the other hand, the hypothesis dealing with colonization of nearest meadows was confirmed. Effect of intentional emigration caused by management interventions was unfortunately not proved. However, we found out, that even though the population at one mowed meadow decreased clearly, the metapopulation size at the locality remained more or less stable. At investigated (mowed) meadow, there was observed a sharp decline of P. nausithous abundance. On the other hand, P. teleius population was only slightly reduced. It is therefore conceivable, that P. teleius are not as susceptible to innapropriate term of mowing. The results obtained in our research conclusively displayed no possible positive effect of mowing in the middle of flight season and a necessity of suitable management maintenance at localities inhabited by Phengaris butterflies.

Demography and Dispersal Ability of a Threatened Saproxylic Beetle: A Mark- Recapture Study of the Rosalia Longicorn (Rosalia alpina)

This thesis focuses on demography and mobility of an endangered and strictly protected icon of European saproxylic biodiversity, the Rosalia longicorn beetle (Rosalia alpina). Using the mark-recapture method we estimated the size of the population, adult longevity, and dispersal ability of the Rosalia longicorn on three hills in the Ralska Upland, Czech Republic. Furthermore, we assessed the distribution pattern of another 15 populations inhabiting small patches of old beech forest in this area. Factors affecting local survival and the species conservation are discussed.

Monitoring of surface changes caused by underground mining in the Karviná region

Geodetic monitoring of the Louky locality revealed irregularities in the development of the subsidence depression and it also helped to document the changing terrain due to ongoing reclamation activities. Repeated GNSS measurements recorded not only the size of subsidence of individual points, but also the size and direction of horizontal displacement of these points, which revealed the influence of undermining in the area of interest, and in its surroundings. It turned out that particularly the significant tectonic fault A forms a barrier of the influence of underground mining. The subsidence depression develops differently than the models of the predicted subsidence expected, especially in the area between the tectonic faults X and A. The method of aerial photogrammetry recorded not only the subsidence caused by underground mining, but also the extent and size of the changes caused by the ongoing reclamation of the surface.

Provision of alternative refuse collection for a real estate surrounded by foreign property

The thesis deals with possible ways of drainage and wastewater disposal. The issue of wastewater discharge to the environment has been closely monitored in recent years, and there is an apparent effort to improve the current situation.The dissertation discusses and compares three selected options – access to improved sanitation, building reservoirs and domestic wastewater treatment plant.The final section is dedicated to a comparison of acquisition and operating costs over time and to subsequent evaluation of the most suitable option. For the sake of clarity and assessment of potential solutions, the locality in the administrative area Vysoké Mýto - Domoradice has been selected as a model. The local authorities have been designing a public storm sewer, and given that it is surrounded by land, this issue will need to be addressed.

Sharing Local Information for Faster Scanning-Window Object Detection

Cílem této dizertační prace je vylepšit existující detektory objektů pomocí sdílení informace a výpočtů mezi blízkými pozicemi v obraze. Navrhuje dvě metody, které jsou založené na Waldově sekvenčním testu poměrem pravděpodobností a algoritmu WaldBoost. První z nich, Early non-Maxima Suppression , přesunuje rozhodování o potlačení nemaximálních pozic ze závěrečné fáze do fáze vyhodnocování detektoru, čímž zamezuje zbytečným výpočtům detektoru v nemaximálních pozicích. Metoda neighborhood suppression doplňuje existující detektory o schopnost zavrhnout okolní pozice v obraze. Navržené metody je možné aplikovat na širokou škálu detektorů. Vyhodnocení obou metod dokazují jejich výrazně vyšší efektivitu v porovnání s detektory, které vyhodnocují jednotlivé pozice obrazu zvlášť. Dizertace navíc prezentuje výsledky rozsáhlých experimentů, jejichž cílem bylo vyhodnotit vlastnosti běžných obrazových příznaků v několika detekčních úlohách a situacích.

HUMAN ACTION RECOGNITION IN VIDEO

Tato disertační práce se zabývá vylepšením systémů pro rozpoznávání činností člověka. Současný stav vědění v této oblasti jest prezentován. Toto zahrnuje způsoby získávání digitálních obrazů a videí společně se způsoby reprezentace těchto entit za použití počítače. Dále jest prezentováno jak jsou použity extraktory příznakových vektorů a extraktory pros- torově-časových příznakových vektorů a způsoby přípravy těchto dat pro další zpracování. Příkladem následného zpracování jsou klasifikační metody. Pro zpracování se obecně obvykle používají části videa s proměnlivou délkou. Hlavní přínos této práce je vyřčená hypotéza o optimální délce analýzy video sekvence, kdy kvalita řešení je porovnatelná s řešením bez restrikce délky videosekvence. Algoritmus pro ověření této hypotézy jest navržen, implementován a otestován. Hypotéza byla experimentálně ověřena za použití tohoto algoritmu. Při hledání optimální délky bylo též dosaženo jistého zlepšení kvality klasifikace. Experimenty, výsledky a budoucí využití této práce jsou taktéž prezentovány.
