Content of succrose in authentic honey from Czech Republic

The work is focused on the analysis of data on sugar composition of honey coming from beekeepers in the Czech Republic. The database includes a number of years and several hundred locations. The aim is to test the hypothesis that sucrose content in honeys vary in individual years in connection with the course of the weather, especially the sufficiency or deficiency of rainfall, so that in rare cases it can even unadulterated honeys sucrose content exceed the limit specified standard. It will formulate recommendations for action at the findings of the high sucrose content of genuine honeys to prevent unauthorized punishment.
Množení česneku kuchyňského - jarního a ozimého nepaličáku - v podmínkách in vitro

In this Bachelor's thesis we studied influencing of bulbs multiplications growing in vitro by adding grow-regulators PP 333, CEPA, ABA to the medium or increase of sucrose concentration in the medium. The culture from segments of inner clove part was cultivation on primary MS medium and 0.5mg/l GA3 was added and 2iP was replaced by the similar concentration of BAP for the cultures from apical meristems. The experiment was made with 2 unregistered garlic genotype with working names Emilia and 'Japo'. We also observed CO2 and ethylene production by the growing plants and influencing bulbs multiplications by these gases. Explantats made from segments of inner clove part after ribavirin application were tested if they contains OYDV virus and DNA analysis was made in order that we could verify genetic stability of in vitro growth plants. In this work were found statistically significant differences in the production of onions in the individual genotypes of garlic on tested media.
Study of thermal properties of sucrose

This work is focused on the study of thermal properties of materials that are used for accumulating heat on the basis of phase transition. These are mainly sugar alcohols (mannitol, erythritol) or carbohydrates (glucose) respectively. Thermal properties, thermal in particular, and thermal conductivity and specific heat capacity were measured in a wide range of temperatures comprising of the transformation from the solid to the liquid phase in the entire volume of substance. To study the temperature dependence of thermal properties, transient methods were used, that allow to determine the diffusion and relaxation time, thermal absorption coefficient and a parameter characterizing the properties of resources based on the fractal model of heat transfer (shape and the heating of the sample). Thermal parameters were determined from those values for defined ideal heat source (planar) and defined ideal heating (leap). There are results of three sucrose samples produced in the Opava refinery (unrefined, refined, clean). Differences in their thermal properties were studied. In addition, thermal properties of a pure solution (commercial) of sugar in distilled water at different concentrations were studied (1:1, 3:1, 1:3). The results presented in this thesis may be used to more efficient use of thermal energy in heating of dwellings, both with the use of solutions, which have a higher heat capacity than water and also with the use of phase transformation.
Proposal of systems for targeted transoprt of compounds of sweet taste

This bachelor thesis is focused on possibility of immobilization sweeteners on polymeric system. In the theoretical part information about sweeteners, carrier systems and the possibility of immobilization technique were reviewed. In the experimental part preparation of nanoparticles of polyhydroxybutyrate (PHB) was optimized. The PHB particles were characterized by dynamic light scattering (DLS). PHB particles were activated by plasma treatment. The surface morphology was studied using infrared and Raman spectroscopy. Immobilization of following substances with a sweet taste was tested: glucose, fructose, sucrose, mannitol, sorbitol and erythritol. From these compounds only sucrose was immobilized on PHB particles successfully. Prepared immobilized particles were exposed to the artificial stomach juice, intestinal juices and bile acids and amount of released sucrose was monitored. Finally, long-term stability prepared particles was measured and also the amount of sucrose released was determined.
Exogenní a endogenní faktory tvorby cibule u česneku kuchyňského v podmínkách in vitro

In this thesis the study of effects exogenous and engoenous factors on garlic (Allium sativum L.) plats in vitro was done. Species 'Lukan', 'Lan', 'Bjetin', 'Slavin' a 'Havran' were observed with their reactions to growth regulators added to MS medium with 0,5 mg.l-1 2iP, 0,2 mg.l-1 NAA - variants with PP 333, ABA, CEPA and increased sucrose content. During the growth of the plant, ethylene, ethane and CO2 concentrations in culturing bulbs were determined and plant growth was monitored, namely bulb creation and mass increase. At the end of the experiment, the stability of donor plant and explant was verified using DNA markers in 'Lan' and 'Lukan' species.
Vliv osmotického stresu na růst rostlin bramboru v in vitro podmínkách

Osmotic stress influences productivity and crop yield of plants significantly. This bachelor thesis is a literature review summarizing the acclimation of plants under osmotic stress and tolerance of plants to osmotic stress, observes issues of abscisic acid in the relation to osmotic stress and other markers as osmoprotectants and dehydrins. Screening experiment in vitro culture of potato plants (Solanum tuberosum L.) in agar stiffened media containing stressors as sodium chloride, sucrose and polyethylene glycol was carried. Plant growth parameters such as solids content and length of roots was observed. RIA method was used to determine the content of abscisic acid in plant leaves influenced by stressors.

This thesis deals with the determination of undeclared or illicit added sugar and is evaluated by HPLC (High-Performance Liquid Chromatography) with refractive index detection. The approximate determination was carried out by Fieheh reactions II. Labeling of samples was also evaluated in accordance with legislation. Samples of honey were collected from the trade network, beekeeping markets and stores and directly from beekeepers and were purchased in the fall of 2013. Samples directly from the beekeeper came from the Christmas Markets in Brno 2013. Totally 21 samples out of which 13 samples were purchased in the market network, 6 came directly from the beekeeper and 2 were from beekeeping markets and stores. It was observed if the samples meet the legislative requirements for the kontent of fructose, glucose and sucrose. It was also assessed whether the samples meet the legislative requirements for mandatory labeling of honey data. Meet the legislative requirements of 4 samples out of 21 samples.
The influence of competition and soil fertility on dynamics of storage in species \kur{Plantago lanceolata}

This study investigates influence of strong competitor and soil fertility on growth and carbohydrate storage production of Plantago lanceolata, the usually subordinate species in plant communities, using experimental approach. As a dominant competitor Poa pratensis was used.
