Determination of selected components in human urine with electrophoresis in short capillary.

Capillary zone electrophoresis is frequently used in various analyses. In this diploma thesis a hydrodynamic sample introduction method controlled by pressure pulse has been proposed for short-capillary electrophoresis. The base electrolyte flushes sample from the loop of a six-way sampling valve and is carried to the injection end of the capillary. At the time when the sample zone reached the capillary, a short pressure impulse is generated in the electrolyte stream, which provides injection of the sample into the capillary. Then the electrolyte flow is stopped and the separation voltage is turned on. The amount of sample introduced to the capillary is controlled by the duration of the pressure pulse. This new sample introduction method was tested in the determination of ammonia, histidine, creatinine, uric acid and hippuric acid in human urine and for rapid screening of the contents of the inorganic ions in cerebrospinal fluid and blood plasma. The determination was performed in a capillary with an overall length of 10,5 cm and two base electrolytes was tested - 50 mM MES + 5 mM NaOH (pH 5,10) and 1 M acetic acid + 1,5 mM crown ether 18-crown-6 (pH 2,40). Involvement of donors of blood plasma to the donor's registry bone marrow

The Bachelor thesis is focusing on the possibility to exploit the potential of regular blood plasma donors for inclusion into the register of bone marrow donors. Based on the assumption, that this is the almost identical population and that the criteria for blood plasma donating are similar to the criteria for bone marrow donation, plasma donors are tested periodically on the infectious diseases transmitted through blood, they have experience with the intravenous collection and plasma donors are largely complying with the age limit needed for bone marrow register. The theoretical part describes anatomy and physiology of blood plasma and bone marrow and current knowledge on the issue for donating of blood plasma and bone marrow. Describes types and indications of bone marrow transplantation, registers of blood plasma and bone marrow donors, the motivation for donating, reliefs and remunerations for donors. (thesis): Basis: This diploma thesis follows the bachelor thesis that concentrates on the eating habits and water intake of blood plasma donors before plasmapheresis. Goals: The research had four goals: 1) explore to what extent does providing of the educational material influence the awareness of blood plasma donors. 2) To map subjective state of health and lifestyle of plasma donors regarding their physical activity, healthy, regular, varied diet, smoking and sleep schedule. 3) To explore the knowledge of regime measures and risk behaviour before donating blood plasma. To evaluate donors' eating habits and water intake before plasmapheresis and reveal potential deficiencies. 4) Evaluate the knowledge of a given problem with those respondents who were provided with educational material in comparison with those respondents who got no educational material. Methods: An educational material was created regarding regime measures of blood plasma donors. This educational material served as the basis for standardized anonymous questionnaire. Methods: The serological typing of HLA antigens using the lymphocytotoxic test was performed among plasma donors of the Transfusion Department of the University Hospital Hradec Kralove. On a sample group of 200 plasma donors was determined the frequency of HLA antigens by blood group and gender on the basis of a serological examination. The collected data of the observed sample of aforementioned group were processed using Microsoft Excel in relation to the gender and blood group of the donor. Results: It was discovered that within the observed sample there is the highest frequency of HLA-A antigens 2, 1 and 3; HLA-B antigens 7, 44, 35 and HLA-C antigens 7, 6, 4. Objective of this diploma thesis was to evaluate internal environment of racehorses according to physical load. Experiment was done on ten english thoroughbred horses. These horses were prepared in training for the plain racing. Blood collections for analysis were done at rest, 5 and 10 minutes after physical load. Analysis were done by metabolic profile test. Biochemical and hematologic measurements were studied on horses. All horses showed high values of selen, from 278,62 to 337,48 umol/l. Thanks to biochemical measurements, at creatine kinases, the biggest value that was find out was 8,13 ukat/l at rest, after 5 minutes of rest, it was in range of 3,72 - 8,23 ukat/l and after 30 minutes, in range between 3,01 - 12,88 ukat/l. At rest the lactate was established at biggest value 6,58 mmol/l, after 5 minutes of rest values on all tested horses increased to biggest measured value 8,98 mmol/l. Factor LDH has increased too, after 5 minutes of rest it has increased from the standing point to the biggest measured value at 9,97 ukat/l. Intensive training of race horses causes that the tracked measurements arent going back to the optimal values after the 30 minute rest. Majority of tracked measurements, especially biochemical, exceeds upper minimum of reference values. We conclude that it would be appropriate adjusting the reference values and standards for english thoroughbred in physical load.

(thesis): Background: This study explores the issue of eating habits and water intake of blood plasma donors. Objectives: The aim of this research was to map the blood plasma donors' awareness of well-balanced diet, evaluate their eating habits and water intake. Inquire donors' knowledge of plasmapheresis, their awareness of food suitable for replenishing proteins after the collection of blood plasma and iron in low levels of haemoglobin. Compare the differences in eating habits between men and women. Map the frequency of rejected plasma donors due to low haemoglobin, high level of leucocytes or chylous plasma. In the conclusion the respondents evaluate their own knowledge of the given problems. Methods: The research was conducted in a method of a standardised anonymous questionnaire. Overall data of 300 respondents attending a private plasma centre were processed while maintaining ethical principles. The research took place in 14 days, from 18th November 2015 until 2nd December 2015, so as to avoid a situation in which one client fills more than one questionnaire. The questionnaires were filled anonymously by clients waiting for their plasma to be taken or during the process in a donation chair. Plasma protein shave many important functions in the organism. Gamma globulins are carriers of immunoglobulins which play animportant role in the immune response. Their contentis primarily given by the health burden of the organism. The aimofthe study was to determine the individual protein fractions in the blood plasma of ewes and lambs, comparemutual relations between total plasma proteins and thein variol factions and assess the concentrations of individual plasma proteins, mainly globulin, in connection with the aktivity of the thyroidgland, physiological state, and increments. The sampling was carried out in the spring (25.3.) and the autumn (14.10) 2013. The individual protein fractions were determined from the serum by the means of electrophoresis. The results show that the concentration of proteins in the blood plasma of bothewes and ewe lambs and ram lambs was high due to haemoconcentration, heat stress during sampling, grazing young green forage, comprising a large number of protein aceous substances, and increasing demands on energy for milk production, especially in the early stagesoflactation. Theconcentrationofproteins in theblood plasma oflambspertains to thegrowthproduction. It establishes a correlation between the concentration of plasma proteins and the aktivity of the thyroidgland. It also confirmes higher concentrations of plasma proteins of lambs correlating to thein higher daily gain. Due to the high concentration of total protein in the plasma, the level of its fractions was high as well. After the conversion to a percentage, the level of - globulins in blood plasma in all categories was within normal limits in the range of 14-27%. The concentration of - globulins in blood plasma increased in relation to the parasitological findings coccidia of the genus Eimeria and gastrointestinal nematodes. Relations between the kontent of plasma proteins in the blood of the lambs and ewe sobserved were in most casespositive. There was a strong dependence between the total protein and globulins. Even among other fractions of plasma proteins and globulins correlation coefficient was almou always positive.
