This diploma thesis is about soybean as functional food. Functional food contain components with positive effect on human health. In soybean there are lot of these components and very valued are phytoestrogens. Phytoestrogens has estrogenic activity with positive effect in prevention of osteoporosis, breast and prostate cancer, cardiovascular disease and menopausal problems. In experimental part was measured level of isoflavones in different kinds of soy food. The method for separation was high performance liquid chromatography with mass spectrometry.

Diploma thesis on the theme Concentration of selected phytoestrogens in the diet of dairy cattle and their distribution in to the blood and milk deals about transmittance of phytoestrogens from real fed TMR in the South Moravian conventional breedings of dairy cows into the blood plasma of animals and to milk as a product intended for hu-man consumption. A review of literature is devoted to the occurrence of phytoestrogens in forage and feed, their metabolism and effects on animals and humans. The experimental part of the thesis is devoted to the identification and quantification of selected phytoestrogens in feed and their penetration into blood plasma and milk. Determination of phytoestrogens was performed by HPLC - MS analysis.

Phytoestroegens are biologically active compounds naturally occuring in plant materials. This group consists mainly of polyphenols characterized by structural similarities with female hormone 17-beta-estradiol. Plants produce phytoestrogens as their secundary metabolities, primarily due to the reaction as a response to various stress. Agents with estrogen effect contained in foodstuffs bring very positive impact on our health. On the other hand, some of the estrogen compounds may be harmful to the health depending on the concentration and the complex state of the consumer, as well as mycoestrogens, xenoestrogens and antiestrogens. Also for this reason is necessary to detect and quantify them in the sample of food. The major problem encountered in the studies of their effect is large amount of a variety of other substances in biological material, and according to their complicated structure was important to establish a reliable method, that was used also in the practical part of my experiment. It is an UPLC-MS/MS -- combination of Ultra High Performance Liquid Chromatography and Tandem Mass Spectrometry preceded by complex sample preparation which involves also SPE purification ("solid phase extraction").
Monitoring of Phytoestrogens Expression to the Milk and Milk Products

Phytoestrogens are very intensively studied in recent years because they show different pharmacological effects on human health. For example, it was demonstrated that phytoestrogens reduce the risk of certain cancers (breast, prostate or colon), act as antioxidants in the body and affect the activity of certain enzymes. In addition, also it was demonstrated number of positive effects of soybean phytoestrogens on the health status of women in the menopause, such as reducing the risk of atherosclerotic cardiovascular disease or osteoporosis. Various technologically processed soy beans are important part of the feed rations mainly for high-lactating dairy cows, because they are a valuable source of the energy and high-quality protein, despite their negative effect on reproductive performance. Phytoestrogens consumed from the feed rations are hydrolyzed in the rumen of dairy cow and together with their metabolites are absorbed in the small intestine into the blood. They can be excreted from the body in urine or milk. From this perspective, cow's milk may become a source of phytoestrogens substances, especially equol in the human nutrition. On the other hand, some research results suggest possible potential risks associated with phytoestrogens intake in the diet on human health. At the some age (infants and young children) and group (vegetarians) categories may be dietary intake of phytoestrogens undiserable. According to most experts, a beneficial effect on human health prevail the potential risks. However, the risks associated with phytoestrogens intake in food can not be ignored and therefore phytoestrogens are still actual problem in the field of scientific research.
Evaluation of selected dietary supplements - climacterium

A dietary supplement is a product that contains nutrients (e.g. vitamins, minerals, herbs, amino acids or fatty acids) intended to supplement the diet. The aim of the diploma thesis was to suggest a prescription for preparation of three types of dietary supplements which are used to reduce of climacteric symptoms, their production and determination of energetic (nutrition) value on account of detected content of proteins, fats and saccharides in dietary supplements. A part of the diploma thesis was to suggest suitable wrapping and price of prepared dietary supplements. The three dietary supplements were used for analyses, namely capsules, herb syrup and herb drops. The content of proteins was determined by Kjeldahl´s method, total fats were determined by Soxhlet extraction and saccharides were determined by titration with potassium permanganate (Bertrand´s method). Dietary supplements in form of capsules were particular analysed on content of heavy metals (lead, cadmium and arsenal). In herbal drops was in addition determined alcohol content (ethanol). There was from the measured data found out, dietary supplement in form of capsules was chosen as the product with the lowest energetic (nutrition) value.

We live in tough times. We experiencing high fyzical and psychical pressure whitch corrupting our healthy livestyle. This restriction takes its toll in the form of lifestyle diseases such as diabetes mellitus or obesity. That's why I decided to concentrate my bachelor thesis to healthy nutrition in the form of functional foods. I belive that conclusions of this thesis can show the way to people whos looking for healthy life. Literary part of this work is focused to functional foods, their general characteristics and the main representatives. Furthermore, the importance of biologically active substances contained in them. I pay attention only few of them because huge range of this topic. From the group of antioxidants for example vitamin E and vitamin C and from the micronutrients selenium and carotenoids. I also examine fiber becouse of high incidence of colorectal cancer disease in the Czech Republic. The experimental part of my work is focused on phytoestrogens, their structure, content in foods and effects on human health. I'm extracting phytoestrogens using the high-performance liquid chromatography in the scope of the experiments of various plant materials.

Bachelor thesis entitled The influence of dietary habits on the incidence of cancer in the literature review discusses the importance of nutrition in disease prevention. Describes some negative factors on human health. It also deals with the issue of phytoestrogens, their occurrence in foods and their effect on human health. The last section focuses on recommendations of daily routine and diet for people with a genetic predisposition to cancer. The practical part deals with the evaluation questionnaires from respondents with cured cancer.
Natural sources of lignans in our diet

This bachelor thesis deals with lignans (phytoestrogens) in our diet. Preliminary of the work is focused on the characteristic of lignans, which belongs to the not entirely explored group of substances contained in various parts of the plant. The work reviews the most utilized crops (soy, flax, and cereals), from which was isolated a large amount of lignans and other compounds with estrogenic activity. Furthermore, there is introduced the list of substances contained in crops and mechanisms of their action in the human body. Their use is very beneficial in the prevention of serious diseases, so-called civilization. At the end of the work, there are highlighted various methods of separation of lignans from plants.
