Extracts from vermicomposts produced under different conditions

The main purpose of this study was to determine if aeration and extraction time influence on the properties of extracts produced under different conditions. The leachates for our experiment were obtained from five different kinds of vermicompost (from horse manure, digestate, marc, apple pomace and from kitchen waste). Has always been prepared 1kg of vermikompost and using special extraction equipment for leaching in 9 liters of water. Attempt always proceeded 48 hours and at predetermined intervals, were taken control samples. Every 1h, 6h, 12h, 24h and 48h. The leachates were prepared in two variants. The first variant was without aeration at a constant temperature 30 °C and stirring 750 rpm. The second variant was with aeration at a constant temperature 30 °C and stirring 750 rpm. In this work we are evaluated in terms of leachate contents of ammonium nitrogen(N-NH4+), nitrate nitrogen (NO3--N), total nitrogen, dissolved organic carbon (DOC). Analytical equipment placed on the apparatus measured the remaining variables evaluated in this work - pH, conductivity and dissolved oxygen. The leachates produced with aeration generally reached higher values than the leachates produced without aeration. This difference was only 6%. The highest increase during exgraction was the ammonium nitrogen in a variant with aeration. Other endpoints were higher by only a few percent . The extraction time effect on the properties of extracts was clear. The highest rates of increase was in the first 24 hours, when values were rising most. The remaining 24 hours have had such importance. Time certainly had a great influence on the properties of the extracts, but the last 24 hours of leaching characteristics has not changed significantly, so it is not necessary to infuse for 48 hours.
Influence of selected parameters on biotechnological production of polyhydroxyalkanoates

The aim of this work is to study microbial production of polyhydroxyalkanoates (PHA). Theoretical part is focused on production of PHA using microoganisms and transgenic plants. Bacterial strain Cupriavidus necator H16 was used for laboratory production of PHA. Various waste oils were used as sole carbon and energy source. Salt of propionic adic and 1-propanol were used as intermediate for 3-hydroxyvalerate monomer (3HV) unit. Incorporation of 3HV to polymer can improve material features of PHA. The major part of experimental work was focused to study influence of aeration (concentration of dissolved oxygen) to bacterial growth, selected metabolic pathways and formation of PHA. Furthermore, influence of aeration to monomer composition of polymer was evaluated. According to experimental conclusion of this work it was approved dependance between aeration and monomer composition of PHA. Moreover, it was approved that higher concentration of oxygen supports bacterial growth and influences PHA content in cells. In addition, NADPH is one of the substrates influencing flux of acetyl-CoA throughout the metabolism; higher intracellular concentration of NADPH inhibits TCA cycle and enhances accumulation of PHA in cells. For this reason, specific enzymatic aktivity of several selected intracelular enzymes were measured, including those enzymes which can generate NADPH.
Process of destratification at Brno dam reservoir

This thesis deals with the process of destratification at the Brno dam reservoir within the activities of a larger project "Clean Svratka", aimed to improvement of water quality in the catchment area upstream the Brno dam. At the Brno dam the project "Implementation of measures at the Brno Reservoir" has been in progress for the third year. Its aim is the proposal and implementation of measures to suppress the cyanobacteria mass development in the dam lake. One of the key measures is aeration and mixing of dam water through a 20 aeration tower. The theoretical part of this thesis focuses on seasonal conditions in the dam lake, the technology suitable for destratification and aeration and possibilities of their use. In the practical part of the thesis the seasonal destratification and the impact of meteorological conditions are monitored.
Comparison of aeration elements

The content of the thesis is to compare the aeration elements used in the aeration tank for biological treatment of wastewater. In order to compare, the work contains description and explaination of: ? basic division of functions and some aeration systems, ? transfer of oxygen, ? the use of pure oxygen instead of air, ? placement of aeration elements into the activation tank. Since the aeration elements are plentiful, work is focused on the most widely used pneumatic and mechanical aeration systems.
Aeration at WWTP´s.

Problems of aeration dimensioning at WWTP´s.

The objective of this work is to compare aeration systems of primarily mechanical and pneumatic aeration, as well as to compare various manufacturers of aeration components in terms of their advantages, positioning in aeration tanks, operation, use, efficiency and energy requirements. The second part is a description of the waste water treatment plant in the town of Česká Třebová and the development of an alternative calculation for the biological part with circulation activation, a circular sedimentation tank and a design for an aeration system in the aeration tank.
Eutrophication of water reservoirs

The first part of this thesis is focused on general explanation of eutrophication, its causes and consequences. The possible solutions of eutrophication are discussed briefly and a few examples of eutrophic reservoirs in the Czech Republic are added. The second part of this thesis is focused on ongoing modifications of water reservoir Brno and finally the effectiveness of measures are being compared.
Container sewage treatment plants up to 500 PE

The aim of this bachelor´s thesis was making a survey of companies involved in producing container type waste water treatment plants in the domestic market. In the first part of the bachelor´s thesis there is an overview of types of plants, including the method of implementation, use and placement and subsequent operation. In the practical part there is a list of companies active in this field in the Czech Republic that were compared to each other in terms of technological, machine-technological and financial aspects depending on the population for which they are intended for. Data provided by these companies and their processing can be used in selection of container of waste water for expert and unprofessional public, which is considering the acquisition of container plant.
