Testing the effects of a mixture of beta-glucans and antimicrobial extracts

The bachelor thesis aims to describe the effects of a mixture of selected natural substances in conjunction with -glucan obtained from the cell wall of the studied yeast species. The theoretical part describes bioactive substances and compounds with antioxidant activity. Subsequently, selected species of plants and herbs are characterized as direct sources of bioactive substances. In parallel, the following chapters of the theoretical part present as sources of -glucan representatives of yeast species with their morphological characterization. - glucan, its structure, biological effects, as well as the principles of methods used in the determination and preparation of bioactive mixtures of plant extracts and -glucan are described. In the experimental part of the work, attention is paid to the preparation, cultivation of yeast, isolation, and determination of -glucan content using the enzymatic method. The amount of polyphenols, flavonoids, and antioxidant activity in aqueous plant extracts is determined. The content of fatty acids, carotenoids, and coenzyme Q is determined in yeast. Subsequently, the antimicrobial and antioxidant effects of mixtures of plant extracts and -glucan are tested.
Optimization models in population protection

This master thesis utilizes operational research principles to model and address emergency events, with a focus on rapidly and effectively making decisions related to population sheltering and the allocation of security forces during such crises. The theoretical section provides fundamental information about the current state and application of risk management in civil security while defining terms directly associated with population protection and crisis management. Moreover, it explores the historical application of mathematical programming and optimization in crisis management processes. In the practical segment of the thesis, the fundamental traffic problem is adapted to address the issue of establishing permanent civil protection shelters in the Brno-Židenice district. The models are solved using the GAMS program, building upon a comprehensive risk analysis that meticulously identifies key components of the population protection system. Furthermore, potential risk scenarios for the system are formulated using the FMEA method. The results obtained, which outline optimal allocation strategies for residents and security forces, are visually presented on maps. Recommendations are provided for future planning and crisis management in emergency events.

Studium kožních onemocnění pomocí monovrstevných lipidových modelů

Ochrannou funkci kůže zajišťuje nejvrchnější vrstva epidermis, stratum corneum (SC). Funkce této bariéry je závislá na složení a struktuře extracelulární hmoty ve SC. Hlavní složky této hmoty představují ceramidy (Cer), vyšší mastné kyseliny (VMK) a cholesterol zastoupené v téměř ekvimolárním množství. Změny ve složení mezibuněčné matrix jsou spojovány s rozvojem různých kožních onemocnění (např. atopická dermatitida). Ceramidáza je enzym štěpící Cer a glukosylované ceramidy (prekurzory Cer) za vzniku lysosfingolipidu (LysoSph) a mastné kyseliny. Patologická aktivita tohoto enzymu vede k dysfunkci kožní bariéry a rozvoji kožních onemocnění, např. Farberovy choroby. Cílem této práce bylo studium monovrstevných lipidových modelů, jejichž složení simuluje vysokou aktivitu ceramidázy v kůži. Lipidové modely byly studovány pomocí Langmuirových izoterm a Brewsterovy úhlové mikroskopie na rozhraní vzduch-kapalina a na pevném podkladu mikroskopií atomárních sil. Z hodnocení monovrstev...
