Pathobiochemistry of lysosomal storage disorders: Study of Fabry disease and generation of cellular models of X-linked disorders.

Human autopsy or biopsy tissue samples, mouse models and cell cultures of various types represent the most common materials in the investigation of cell pathogenesis of inherited diseases. This dissertation is devoted to all these approaches in the study of two X-linked lysosomal storage diseases, Fabry disease (FD,α-galactosidase A (AGAL) deficiency) and mucopolysaccharidosis type II (MPSII, idunorate-2- sulfatase (IDS) deficiency). The primary goal of the work was analysis of lipid blood group B antigens with terminal α-galactose (B-GSL) in the pancreas of FD patients with blood group B (FD-B).,In addition to the main glycosphingolipid (GSL) substrate, globotriaosylceramide (Gb3Cer), B-GSLs represent another minor substrate of AGAL. The deposition of undegraded B-GSL has been demonstrated in FD-B pancreas where it was significantly higher than in other organs such as the kidneys and lungs which accumulate mainly Gb3Cer. High concentration of lipid and non-lipid B-antigens was primarily confirmed in exocrine acinar epithelial cells of FD-B, accompanied by massive accumulation of ceroid (secondary sign of lysosomal storage). Crowdfunding for theatre

This work is focused on the issue of crowdfunding for theater or non-profit sector as a tool. It covers all phases of a campaign; setting campaign goals, identifying and quantifying all costs associated with it. I am aiming at using the full potential of the tool and the volume of work involved. I also briefly deal with the motivation of the donor. The last chapter describes three case studies of successful campaigns. The practical output of the work is a campaign budgeting tool, an instrument that enables authors to identify and quantify all costs associated with the campaign and determine more precisely what the target amount should be. The tool proved that if you collect CZK 50.000, the cost of campaigning is about 30% of the amount; the higher amount you collect, the lower campaign costs you reach.
Tandemová hmotnostní spektrometrie sfingolipidů s aplikací pro metabolické studie a diagnostiku sfingolipidos

In recent years, mass spectrometry (MS) become the dominant technology in lipidomic analysis and widely influenced research and diagnosis of diseases of lipid metabolism, e.g. lysosomal storage disorders (LSD) characterized by impairment of the lysosomal functions. Defects in lysosomal processing of sphingolipids SFL belong to the category of sphingolipidoses. This condition has severe and even fatal clinical outcome. The primary aim of this work was to establish quantitative and qualitative methods of SFL analysis useful for research and diagnosis of LSD. At first, semisynthesis of mass labeled lipid standards utilizing immobilized sphingolipid ceramide N-deacylase was performed. Established methods of quantitative analysis were then used to prove the increased excretion of urinary SFL in LSD with characteristic storage in the kidney. Determination of excreted urinary SFL was found useful for differential diagnosis of prosaposin and saposin B deficiences for which routine enzymology is failing. MS also enabled monitoring of individual molecular species (isoforms) of SFL, which led to the finding that their urinary pattern is changing in some LSD. Liver gangliosides in cholestasis induced by bile duct ligation.

Gangliosides are sialic acid-containing glycosphingolipids located on the cell surface of all animal cell types. They play a role as receptor molecules, share in cell-to-cell interaction and protect the cell against harmful environmental factors by increasing of rigidity of cell surface. This diploma thesis studies an influence of experimental cholestasis on hepatic ganglioside composition. Cholestasis was induced by bile duct ligation in Wistar rats. A significant increase of total lipid bound sialic acid and b-series gangliosides (GD1b, GT1b, event. GD3) was found in cholestatic liver when compared with controls. These results found in obstructive cholestasis correspond with the results Majer et al. Biomed. Chromatogr., 21, 446-450 (2007), described in 17α− ethinylestradiol induced cholestasis, but the increase of b-series gangliosides was milder in our study. As a second point, an effect of modulated heme-oxygenase 1 (HO-1) activity was investigated in cholestatis induced bile duct ligation (HO-1 activator- hemine, HO-1 inhibitor- Sn-mesoporphyrin). An increase of a total lipid sialic acid was found in Sn-mesoporphyrin treated animals, but a decrease of some a- and b- series gangliosides was observed. Only a few cultural organizations in the country are devoted to development and continual work with individual fundraising. Many other cultural NGOs stands at the beginning of creating a strategy and planning individual fundraising in general. For this reason, this diploma thesis deals with potential and usable strategies in individual fundraising, donors motivation based on Czech surveys, case studies of cultural organizations with the potential in individual giving and other aspects. The summary should provide basic knowledge framework for the creation of strategies for cultural organizations that may find out desire and motivation to pursue potential of individual fundraising in the future. This thesis seeks to change stereotypes in thinking about individual fundraising as a way of raising funds without the potential and historical context.
The communicational specifics of the role of a production manager in theatre

The aim of this thesis is to map professional communication of production manager in theatre, from the point of view of specifics in theatre environment. It also expresses and compares professional manager communication in a medium-sized company with professional communication of production manager in repertory theatre. And it also determines optimal communication, which is necessary for this profession, and also the most frequent deficiencies, which also appears in this professional communication. Theoretical part of this thesis is focuses on terminology of communication and personal management. Practical part of this thesis is to focus on evaluation a questionnaire forms, which was realized on sample of theatrical productions and focused on communication skills.
