Synthesis and evaluation of novel disinfectant agents

This diploma thesis deals with the synthesis and use of new quaternary ammonium salts (QACs). These substances have been commonly used for decades as disinfectants, antiseptics, antimicrobial additives, preservatives, and currently their decontamination potential using micellar catalysis is being investigated. The theoretical part contains information about the mechanism of action on individual types of microbes, an overview of physicochemical properties, especially solubility and determination of logP, micelle formation. Furthermore, microbial resistance and the mechanism of cross-resistance between QACs and antibiotics (ATB) are described here. The experimental part mainly deals with the synthesis of new QACs, which we expect to have an antimicrobial effect and the potential of creating a micellar microenvironment for micellar catalysis. The first series of reactions is focused on the preparation of three molecules of monoquaternary ammonium salts from the starting material benzylamine, to which two homologous aliphatic alkyl chains C8, C10 and C12 and a hydroxyethyl group are attached. Design of a hydraulic system for long-distance water transport

The Ph.D. thesis aims to design a hydraulic system for long-distance water transport by the serial connection of axial pumps. Specifically, there is presented an analysis of the axial centrifugal pump prototype, which was tested. The prototype has a specific design. An asynchronous electric motor with a hollow-shaft rotor is used. The pump's impeller is pressed into the hollow shaft. Some advantages follow from the design compared to common pumps and pumping stations. For example, it is possible to designate a compact design of a pump and lower pressure load of pipe, so the elastic material for the pipe's wall can be used. However, the main disadvantage of this design is related to the large rotating diameter, where the pump's seals are located. It leads to quite large mechanical losses on seals and in the context of relatively small hydraulic power also to small efficiency. Therefore one part of the Ph.D. thesis is focused on the pump's seal. The result of the work is a new design of the mechanical face seal with a spiral grooves. The seal was manufactured and subjected to the test. During the testing of the prototype axial pump, it was also revealed inappropriate hydraulic design. Therefore it was necessary to redesign the impeller and guide vanes of the pump.
Hydrokinetic swirl turbine

There has been a notable increase in interest in renewable energy harvesting over the past two decades. This has been accompanied by the emergence of new technologies and research, which are driving developments in this area forward. One of the sources is also kinetic energy of river streams and tidal currents. The aim of this thesis is to formulate fundamental equations for the utilisation of kinetic energy of water and to design a hydraulic model of a swirl turbine runner. A further aspect of this thesis is to verify the previously formulated equations in the computational modelling software Ansys CFX and to analyse the effect of the draft tube on the power output of the turbine.
Hydraulic solution of the swirl turbine for locality of Rosenauer mill

This thesis focuses on the design of a swirl turbine for the site of Rosenauer Mill, where the gross hydraulic head HB is 1 m and the flow rate is Q is 1.6 m3/s. The beginning of the work is dedicated to microturbines and explains why a swirl turbine is suitable for this location. The procedure applied for developing the hydraulically active surfaces of the turbine is then presented. Firstly the design of the blades is based on analytical formulas, which are further refined using the CFX Ansys program. The turbine is closely affected by the inlet and draft tube, therefore their design is also included in this thesis. The hydraulic calculations yield the resulting parameters and characteristics of the designed turbine, calculated from the entire assembly using CFD computations. The final section of the work is devoted to the design of main structural components such as the gear, shaft, and bearings. The hydraulic design developed in this thesis serves as the basis for implementing the swirl turbine and constructing a small hydraulic power plant at the Rosenauer Mill.
Performance Assessment of the Selected Company according to Model Start Plus and Proposals for the Improvement

The aim of this thesis is to analyse the use of multi-criteria models to evaluate managerial approaches and corporate performance with an emphasis on non-financial aspects. In this thesis, the evaluation of the company is processed using the Start Plus model. Based on the identified opportunities for improvement of the company, a proposal for improvement is developed. The results of this thesis showed that the company is rated well according to the model. The total score is 410 points. The weaknesses are mainly in the areas of employees, customers, social performance and not defining the vision and mission. Recommendations were made at the end of the thesis to address these weaknesses.
Application of amines for the preparation of chiral cyclic compounds

Organocatalysis, along with catalysis by metal complexes or enzymes, has become one of the very useful tools of asymmetric synthesis. Different activation modes or their combinations into sequences (domino reactions) enable effective access to enantiomerically enriched complex substances. The objective of this work was the preparation of chiral molecules from simple precursors activated by amine-based organocatalysts. In particular, we were focused on a study of asymmetric allyl substitution of Morita-Baylis-Hillman carbonates and two-component domino reactions on sulfur-containing heterocycles. The thesis deals with the organocatalytic allyl amination of Morita-Baylis-Hillman carbonates with aromatic amines catalyzed by β-isocupreidine. The corresponding allylic amines were obtained in high yields (90-96%) with moderate enantiomeric excess. It was possible to get substances of high optical purity (ee 82-99%) by recrystallization of selected products. Furthermore, the developed method was applied in the preparation of enantiomerically enriched β-lactams, which represent valuable precursors, for example, in the synthesis of the drug Ezetimibe. Possibilities of increasing electric supply from Nuclear power plant Dukovany

Subject of the thesis is to describe the ongoing possibilities to increase the electric supply from the Dukovany Nuclear Power Plant and monitoring the selection of the new ultimate heat sink of important technical water system in Dukovany Power Plant. In the first part of the thesis there is a short overview of nuclear power plants in the Czech Republic and around the world. In the following chapters there is a description of a project dealing with increasing the electric supply from the Dukovany Nuclear Power Plant – project Safety 16 Terra EDU [16] and its parts – subproject Utilization of project reserves [19] and subproject with the goal to reduce the shutdowns. In the last part there is a monitoring the selection of a new ultimate heat sink – tower fan – in case of loss current cooling towers due to an extreme wind.

Master´s thesis is the proposal of real business plan for the re-opening of a music club Beseda in Hodonín city. Business plan was set in as reactions for proposal of city Hodonín, which is offering zero rental of area associated with operation of a club to potencional client. As a main information source for procesing of this work i used output of consultation with previous user of this club with assumption, that progress of situation of this club is perfectly reflecting current situation on this market.
Study of sediment transport, flood protection and outline of flood plan in chosen part of the river Thaya

Bachalor's thesis „Study of Sediment Transport, Flood Protection and Outline of Flood Plan in Chosen Part of the River Thaya“ is divided into four parts. In the first one, I describe chosen part of the river Thaya from the Nové Mlýny to the Břeclav weir with focus on geographic, geological and hydrogeological conditions, hydrotechnical structures and river adjustments, biota and protected areas, demography and Lednice-Valtice Cultural Landscape. In the second part, I determine, by bed load samples granulometric analysis, the source of sediments, which appeard under the Břeclav weir. In the third part, I describe chosen area from the view of flood protection, I describe historical floods and I assess capacity of chosen problematic profiles using HEC-RAS program. In the last part, I focus on flood protection of the village Lednice and of the part of Lednice-Valtice Cultural Landscape, I describe problems of this area in context of flood protection and I suggest outline of the Flood Plan of the Village Lednice.
TRANSFORMATION OF THE FORMER BARRACS IN HODONÍN

The object of this thesis is an urbanistic study called Transformation of former barracks in Hodonin. Urbanistic study involves not only urbanistic solution of the barracks area, but also examines the potencial of the city expansion in this locality. Study is focused on realistic model of finishing the city structure and proposal of public spaces in surroundings of historical barracks buildings. Although the city tried to connect this territory back to the city structure and not to limit it’s natural development - the area turned into chaos by selling out lands around the barracks. Not only urbanism is the problem here. Buildings, that would deserve the attention, suffer. The historic barracks are the next object in this thesis. In detailed architectural study is their transformation, which considers historical, constructial, economical and social potencial of these buildings. Reconstruction respects mass and historical value of buildings.
