Fatigue Properties of Ultra-fine Grain Copper Produced by ECAP Method

This diploma thesis describes properties of ultra-fine grain Cu prepared via ECAP procedure. The influence of fatigue loading with positive mean stress on S-N curve (i.e. fatigue life), cyclic plastic behaviour and grain size was investigated. It was found that tensile mean stress leads to shorter lifetime in comparison with fatigue loading with zero mean stress. During main part of the lifetime, significant hardening of UFG Cu was observed. There is no distinct effect on microstructural orientation and stability.

This work deals with fatigue properties and stability of microstructure of ultrafine-grained (UFG) copper prepared by severe plastic deformation by means of equal channel angular pressing (ECAP) method. The effect of different fatigue loading regimes and thermal exposition on microstructural changes was investigated and the fatigue lifetime curves were experimentally determined. The research attention was focussed on localization of cyclic plastic deformation and fatigue crack initiation in UFG structure. Experimental results indicate that after stress-controlled fatigue loading (both symmetrical and asymmetrical) the microstructure remains ultrafine; no grain coarsening was observed. Contrary to this, strain-controlled fatigue loading results in formation of bimodal structure. Grain coarsening was observed also after thermal exposition at 250 °C for 30 minutes. Annealing at lower temperatures does not result in grain coarsening or development of bimodal structure. Fatigue loading results in development of surface relief in form of cyclic slip markings. Their density, distribution and shape differ for particular fatigue loading regimes. Differences in crack initiation mechanism in low- and high-cycle fatigue region were found. Nevertheless, the characteristic feature for all loading regimes was stability of UFG microstructure in the region of cyclic slip bands and fatigue cracks.

REPRESENTATION This theses focused on representation in term sof civil law, family law and business law. This issues is processed komplex and contains all the basic attributes of representation. Theses is based on current treatment of private law and it highlights the changes in new civil Code effective since january 2012. It evaluates legislation in comparison with future changes. It refers to mootness, uncertainties and inaccuracies contained in the current Civil Code. The work focuses on the representation in general and its different types based on the Civil Code. It also contains a modification in the representation of the Commercial Code, which focuses on representing corporate and procuration. The presentation draws on family law representation and guardianship of minors, guardianship.

The diploma thesis is focused on evaluate the efficiency of wastewater treatment plants in the village Veverská Bítýška. Part of the thesis is a literary research, which defines: the basic information about water, wastewater, wastewater treatment and related legislation. Other items of work are characteristic of interest territory and characteristic of selected wastewater treatment plant. In the context of the methodology of the diploma thesis are described selected indicators of water quality and individual determinations in the field and laboratory. Results for the set of indicators of water quality are processed in tables and the graphs, which are accompanied by brief descriptions. Efficiency of wastewater treatment plants is calculated from the acquired measurement results. At the end of the diploma thesis its the overall assessment. Efficiency of wastewater treatment plants always meet the set legislative limits. Values were almost always over 90%.
Deforestrace a její ekologické souvislosti

This bachelor thesis is focused on deforestation and its ecological connections. The main aim is giving an idea of whole issue of deforestation, causes and consequences to local ecosystems especially animals. In the next step it delimits causes and consequences of climate change caused deforestation. By the survey method was evaluated current situation of rainforest felling on the islands Borneo and Sumatra, setting up palm plantations, threatening the populations of orangutans, proboscis monkeys and also impacts to inhabitants living in this area. The results showed that deforestation has a demonstrably negative impact to animal and plant species and community of people living in affected area.

The thesis deals with the issue of dispersed greenery, its division and function in the landscape. By the research method has been characterized 6 selected localities. The coefficient of ecological stability and its changes since 1990 has been calculated for selected localities and then compared by the comparative method. The main aim was to map the state and changes of dispersed greenery (mainly woody plants) along the railway tracks. The condition of greenery along railway tracks and its unkeep was evaluated and compared, which is primarily aimed at keeping the greenery in a condition ensuring safety and continuity of the rail traffic. An integral part of the thesis is a proposal of measures for greenery along the tracks.
ANALYSIS OF SEQUENCE SPECIFIC INTERACTIONS BETWEEN DNA AND P53 FAMILY PROTEINS BY ELISA, SLOT-BLOT AND EMSA

DNA-protein interactions of core domains of p53, p63 and p73, members of tumor suppressor p53 family, were investigated by multiple methods with regard to verifying the sequence specificity with which short target oligonucleotides/long DNA fragments can be recognized. The sequence specificity of core domains and wtp53 full protein binding to specific sequence in both types of DNA substrates was confirmed in solution, on surface and in gels by ELISA, slot-blot and EMSA.

It explores his transformation from a film enthusiast to a renowned creator with many million dollar budgets. By analyzing his early films and comparing with contemporary films, I am discovering how his formal and narrative practices have changed in connection with his entry into mainstream production.

Title of diploma thesis: Study of renal accumulation of antivirals using cellular model Administration of antiviral drugs may be in some case associated with severe nephrotoxicity. This adverse effect can be based on active renal transport and accumulation of the drugs in the renal cells. Several experimental in vitro methods are available to study the mechanisms responsible for drug uptake in the kidney including standard renal cell lines or renal slices. The aim of this study was to evaluate usefulness of isolated rat renal cells to determine which renal transporters may be involved in transport of selected antivirals into the renal cells. The freshly isolated rat renal cells were prepared by two-phase collagenase perfusion method. To evaluate contribution of active and passive transport mechanisms to the renal uptake, accumulation at normal and low incubation temperature was evaluated. To confirm previous information on mechanisms of the renal transport, we also determined which groups of transporters contributed to the renal accumulation of the studied antivirals.
