A Comparison of the Determination 25(OH)D in the Human Serum by Methods UHPLC-MS/MS and CMIA Abbott Architect

In this rigorous work UHPLC-MS/MS and Abbott Architect CMIA methods for determination of 25(OH)D (calcidiol) in human serum were compared. Human serum samples were firstly analysed by the principle of CMIA and subsequently identical samples were processed by UHPLC-MS/MS method that was developed and validated in Research Laboratory of III. Internal Gerontometabolic Clinic of University Hospital in Hradec Králové for 25(OH)D3 and 25(OH)D2.clinical monitoring. Comparison of these methods was performed using the statistical software NCSS 2007 (Number Cruncher Statitistical Software, Kaysville, Utah, USA). Descriptive statistical methods described basic properties of both result sets (mean, median, standard deviation, etc.). The evaluation was tested using pair T-tests to assess the diversity or consensus between the two datasets. Correlation of the two sets of values was investigated by regression analysis. Based on statistical analysis of the results we can conclude that the values obtained by UHPLC-MS/MS and CMIA Abbott Architect methods are statistically significantly different (p = 0.0000), but the correlation is very good (r = 0,8832). Therefore the both methods are well comparable and the results can be converted to each other.

RNDr. Dagmar Solichová, Ph.D. Diploma Thesis Title: Comparison of extraction procedures for HPLC determination of liposolubile vitamines in serum In this thesis were compared extraction methods, solid phase extraction (SPE) and liquid liquid extraction (LLE) used for separation of human serum fat soluble vitamins: A (retinol), E (α-tocopherol), D2 (ergocalcipherol), D3 (cholecalcipherol), 25- (OH)D3 (calcidiol). Vitamins were then determinated by high performace liquid chromatography (HPLC). As a baseline method used the procedure that was developed and partially validated the thesis: Application of the SPE technology and monolithic columns in HPLC analysis of biologically active substances (Horčičková 2009). This procedure was further optimized in order to increase the extraction of target analytes 25(OH)D3, D2 and D3 and shortening the preanalytical phase. ACCULTURATION OF INDIVIDUAL WITH BEHAVIOUR DISORDER - PROGRAMME "BOROUK"

Socialization of an individual with behavioural disorder {--} programme ,,Borouk University Decree (Diploma) Work Abstract The aim of the thesis is to monitor situations of clients with problems of behavioural disorder, who was allowed from a hospital and protective training in Educational Institution and Canteen {\clqq}Husův dům`` in Dvůr Králové na Labem, and an evaluation of the socializational programme {\clqq}Borouk``. In a practical part of the thesis the author chose a quatitative research, in which she analysed data of clients allowed from 2001 to 2005. The programme {\clqq}Borouk`` is evaluated on the basis of data about resocialization of those clients. There is a case study enclosed in the text. In the closing part of the thesis, the programme {\clqq}Borouk`` is evaluated as a suitable programme, which helps clients with the behavioural disorder to socialize. The results show that a wide range of aspects of lifes of a group of examined clients were improved. In spite of their suitable integration and related elimination of negative social phenomenons, it is necessary to provide the clients with behavioural disorder with lifelong assistance and support.
